RAR/RXR and PPAR/RXR Signaling in Spinal Cord Injury

    Get PDF
The retinoid acid receptors (RAR) and peroxisome proliferator-activated receptors (PPAR) have been implicated in the regulation of inflammatory reactions. Both receptor families contain ligand-activated transcription factors which form heterodimers with retinoid X receptors (RXR). We review data that imply RAR/RXR and PPAR/RXR pathways in physiological reactions after spinal cord injury. Experiments show how RAR signaling may improve axonal regeneration and modulate reactions of glia cells. While anti-inflammatory properties of PPAR are well documented in the periphery, their possible roles in the central nervous system have only recently become evident. Due to its anti-inflammatory function this transcription factor family promises to be a useful target after spinal cord or brain lesions

(Autorenreferat)"This extended editorial introduces into the special issue on „The integration of qualitative and quantitative research (IQQR) in psychology” and also presents a genuine contribution to the debate. We begin with a brief description of five current methodological conceptions on IQQR provided in the literature and compare them concerning their specific perspectives on chances and limitations of mixed methods designs. Herein, we give our attention to the question of which of these designs may be easily adopted by psychological research and which, in contrast, may be particularly challenging. As a good example of the latter we identify designs in which qualitative and quantitative research findings are integrated in an approach that attaches equal value to both methodological strands. Specifically to this point, we then propose a Disposition-Meaning-Model which we use in order to discuss the chances of the future development of mixed methods in psychology. This editorial ends with a brief presentation of the papers contributing to this special issue and with the recommendation to make use of their thematic and methodological diversity for the further development of methods in psychological research." C5aR1 Activation Drives Early IFN-gamma Production to Control ExperimentalToxoplasma gondiiInfection

Toxoplasma gondii (T. gondii) is a parasite infecting animals and humans. In intermediate hosts, such as humans or rodents, rapidly replicating tachyzoites drive vigorous innate and adaptive immune responses resulting in bradyzoites that survive within tissue cysts. Activation of the innate immune system is critical during the early phase of infection to limit pathogen growth and to instruct parasite-specific adaptive immunity. In rodents, dendritic cells (DCs) senseT. gondiithrough TLR11/12, leading to IL-12 production, which activates NK cells to produce IFN-gamma as an essential mechanism for early parasite control. Further, C3 can bind toT. gondiiresulting in limited complement activation. Here, we determined the role of C5a/C5aR1 axis activation for the early innate immune response in a mouse model of peritonealT. gondiiinfection. We found thatC5ar1(-/-)animals suffered from significantly higher weight loss, disease severity, mortality, and parasite burden in the brain than wild type control animals. Severe infection inC5ar1(-/-)mice was associated with diminished serum concentrations of IL-12, IL-27, and IFN-gamma. Importantly, the serum levels of pro-inflammatory cytokines, including IL-1 alpha, IL-6, and TNF-alpha, as well as several CXC and CC chemokines, were decreased in comparison to wt animals, whereas anti-inflammatory IL-10 was elevated. The defect in IFN-gamma production was associated with diminishedIfngmRNA expression in the spleen and the brain, reduced frequency of IFN-gamma+NK cells in the spleen, and decreasedNos2expression in the brain ofC5ar1(-/-)mice. Mechanistically, DCs from the spleen ofC5ar1(-/-)mice produced significantly less IL-12 in response to soluble tachyzoite antigen (STAg) stimulationin vivoandin vitro. Our findings suggest a model in which the C5a/C5aR1 axis promotes IL-12 induction in splenic DCs that is critical for IFN-gamma production from NK cells and subsequent iNOS expression in the brain as a critical mechanism to control acuteT. gondiiinfection

[EN] The object of the DESEAS Project, funded by the EC, was to gather preliminary data on the abundance and maximum depth distribution of the rose shrimp Aristeusantennatus in the Mediterranean Sea. An exploratory survey was therefore designed with that goal in mind and conducted on the R/V García del Cid, sampling the maximum depths in three specific areas in the central and western Mediterranean, one off Ibiza (Balearic Islands), one off Calabria (western Ionian Sea), and one off the southern Peloponnesian Peninsula (Gulf of Kalamata, eastern Ionian Sea). The depths sampled ranged from 600 to 4000 m, with specimens of A. antennatus being collected down to 3300 m. There were three distinct boundaries marking the abundance of this species: 1500 m, relatively low abundance (<100 ind km-2). The known population structure of this shrimp species, with increasing proportions of males and juveniles with depth, was also recorded in the deep-sea regions in other areas of the Mediterranean. No evidence of any differences in gonad development or in the presence of spermatophores carried by females was found in any of the three sampling areas.
