    How to Evaluate Non-Growing Cells - Current Strategies for Determining Antimicrobial Resistance of VBNC Bacteria

    Thanks to the achievements in sanitation, hygiene practices, and antibiotics, we have considerably improved in our ongoing battle against pathogenic bacteria. However, with our increasing knowledge about the complex bacterial lifestyles and cycles and their plethora of defense mechanisms, it is clear that the fight is far from over. One of these resistance mechanisms that has received increasing attention is the ability to enter a dormancy state termed viable but non-culturable (VBNC). Bacteria that enter the VBNC state, either through unfavorable environmental conditions or through potentially lethal stress, lose their ability to grow on standard enrichment media, but show a drastically increased tolerance against antimicrobials including antibiotics. The inability to utilize traditional culture-based methods represents a considerable experimental hurdle to investigate their increased antimicrobial resistance and impedes the development and evaluation of effective treatments or interventions against bacteria in the VBNC state. Although experimental approaches were developed to detect and quantify VBNCs, only a few have been utilized for antimicrobial resistance screening and this review aims to provide an overview of possible methodological approaches

    Criterion-Validity of Commercially Available Physical Activity Tracker to Estimate Step Count, Covered Distance and Energy Expenditure during Sports Conditions

    Background: In the past years, there was an increasing development of physical activity tracker (Wearables). For recreational people, testing of these devices under walking or light jogging conditions might be sufficient. For (elite) athletes, however, scientific trustworthiness needs to be given for a broad spectrum of velocities or even fast changes in velocities reflecting the demands of the sport. Therefore, the aim was to evaluate the validity of eleven Wearables for monitoring step count, covered distance and energy expenditure (EE) under laboratory conditions with different constant and varying velocities.Methods: Twenty healthy sport students (10 men, 10 women) performed a running protocol consisting of four 5 min stages of different constant velocities (4.3; 7.2; 10.1; 13.0 km·h−1), a 5 min period of intermittent velocity, and a 2.4 km outdoor run (10.1 km·h−1) while wearing eleven different Wearables (Bodymedia Sensewear, Beurer AS 80, Polar Loop, Garmin Vivofit, Garmin Vivosmart, Garmin Vivoactive, Garmin Forerunner 920XT, Fitbit Charge, Fitbit Charge HR, Xaomi MiBand, Withings Pulse Ox). Step count, covered distance, and EE were evaluated by comparing each Wearable with a criterion method (Optogait system and manual counting for step count, treadmill for covered distance and indirect calorimetry for EE).Results: All Wearables, except Bodymedia Sensewear, Polar Loop, and Beurer AS80, revealed good validity (small MAPE, good ICC) for all constant and varying velocities for monitoring step count. For covered distance, all Wearables showed a very low ICC (<0.1) and high MAPE (up to 50%), revealing no good validity. The measurement of EE was acceptable for the Garmin, Fitbit and Withings Wearables (small to moderate MAPE), while Bodymedia Sensewear, Polar Loop, and Beurer AS80 showed a high MAPE up to 56% for all test conditions.Conclusion: In our study, most Wearables provide an acceptable level of validity for step counts at different constant and intermittent running velocities reflecting sports conditions. However, the covered distance, as well as the EE could not be assessed validly with the investigated Wearables. Consequently, covered distance and EE should not be monitored with the presented Wearables, in sport specific conditions

    Resonant nonlinear magneto-optical effects in atoms

    In this article, we review the history, current status, physical mechanisms, experimental methods, and applications of nonlinear magneto-optical effects in atomic vapors. We begin by describing the pioneering work of Macaluso and Corbino over a century ago on linear magneto-optical effects (in which the properties of the medium do not depend on the light power) in the vicinity of atomic resonances, and contrast these effects with various nonlinear magneto-optical phenomena that have been studied both theoretically and experimentally since the late 1960s. In recent years, the field of nonlinear magneto-optics has experienced a revival of interest that has led to a number of developments, including the observation of ultra-narrow (1-Hz) magneto-optical resonances, applications in sensitive magnetometry, nonlinear magneto-optical tomography, and the possibility of a search for parity- and time-reversal-invariance violation in atoms.Comment: 51 pages, 23 figures, to appear in Rev.     Mu Transposon Insertion Sites and Meiotic Recombination Events Co-Localize with Epigenetic Marks for Open Chromatin across the Maize Genome

    The Mu transposon system of maize is highly active, with each of the ∼50–100 copies transposing on average once each generation. The approximately one dozen distinct Mu transposons contain highly similar ∼215 bp terminal inverted repeats (TIRs) and generate 9-bp target site duplications (TSDs) upon insertion. Using a novel genome walking strategy that uses these conserved TIRs as primer binding sites, Mu insertion sites were amplified from Mu stocks and sequenced via 454 technology. 94% of ∼965,000 reads carried Mu TIRs, demonstrating the specificity of this strategy. Among these TIRs, 21 novel Mu TIRs were discovered, revealing additional complexity of the Mu transposon system. The distribution of >40,000 non-redundant Mu insertion sites was strikingly non-uniform, such that rates increased in proportion to distance from the centromere. An identified putative Mu transposase binding consensus site does not explain this non-uniformity. An integrated genetic map containing more than 10,000 genetic markers was constructed and aligned to the sequence of the maize reference genome. Recombination rates (cM/Mb) are also strikingly non-uniform, with rates increasing in proportion to distance from the centromere. Mu insertion site frequencies are strongly correlated with recombination rates. Gene density does not fully explain the chromosomal distribution of Mu insertion and recombination sites, because pronounced preferences for the distal portion of chromosome are still observed even after accounting for gene density. The similarity of the distributions of Mu insertions and meiotic recombination sites suggests that common features, such as chromatin structure, are involved in site selection for both Mu insertion and meiotic recombination. The finding that Mu insertions and meiotic recombination sites both concentrate in genomic regions marked with epigenetic marks of open chromatin provides support for the hypothesis that open chromatin enhances rates of both Mu insertion and meiotic recombination

    Classification of selected cardiopulmonary variables of elite athletes of different age, gender, and disciplines during incremental exercise testing

    Incremental exercise testing is frequently used as a tool for evaluating determinants of endurance performance. The available reference values for the peak oxygen uptake (VO2peak)(VO_{2peak}), % of VO2peakVO_{2peak} , running speed at the lactate threshold (vLT)(v_{LT}), running economy (RE), and maximal running speed (vpeak)(v_{peak}) for different age, gender, and disciplines are not sufficient for the elite athletic population. The key variables of 491 young athletes (age range 12–21 years; 250 males, 241 females) assessed during a running step test protocol (2.4ms−1;increase0.4ms−15min−1)(2.4 m s^{−1} ; increase 0.4 m s^{−1} 5 min^{−1}) were analysed in five subgroups, which were related to combat-, team-, endurance-, sprint- and power-, and racquet-related disciplines. Compared with female athletes, male athletes achieved a higher vpeakv_{peak} (P = 0.004). The body mass, lean body mass, height, abs. VO2peak(mlmin−1)VO_{2peak} (ml min^{−1}), rel. VO2peak(mlkg−1min−1)VO_{2peak} (ml kg^{−1} min^{−1}), rel. VO2peak(mlmin−1kg−0.75)VO_{2peak} (ml min^{−1} kg^{−0.75}), and RE were higher in the male participants compared with the females (P < 0.01). The % of VO2VO_2 at vLTv_{LT} was lower in the males compared with the females (P < 0.01). No differences between gender were detected for the vLTv_{LT} (P = 0.17) and % of VO2VO_2 at vLTv_{LT} (P = 0.42). This study is one of the first to provide a broad spectrum of data to classify nearly 500 elite athletes aged 12–21 years of both gender and different disciplines
