    The Hamburg Tornado (7 June 2016) from the perspective of low-cost high-resolution radar data and weather forecast model

    A tornado hit the northeastern suburbs of Hamburg, Germany, on 7 June 2016. It had an estimated strength of upper end F1 on the Fujita scale and was short-lived with an approximate duration of only 13 min and a path length of just about 1.3 km. We demonstrate that such a small-scale, extreme event can be observed and forecasted accurately by a low-cost radar and by an atmospheric model with low computational costs, respectively. Observations from a low-cost single polarized X-band radar covering the urban area of Hamburg with 60 m spatial and 30 s temporal resolution are analyzed with respect to their ability to capture the development as well as the track of the tornado. In contrast to the national C-band radar network, the X-band radar is capable of capturing the hook echo of the tornado as well as the circular pattern in rain rates, because of its higher resolution in space and time. High-resolution forecasts of the tornado event are conducted with the computational efficient Conformal Cubic Atmosphere Model (CCAM) in order to test the capability of predicting the tornado with a lead time of a few hours. A three step downscaling method is used to obtain a spatial resolution of 1 km with initial conditions taken from the NCEP analysis. Calculated severe weather indices clearly indicate a potential for a tornado. CCAM cannot explicitly resolve small scale tornadic features but the model simulates a strong convective cell only a few kilometers apart from the tornadic thunderstorm observed by the radar

    Tracing <i>Mycobacterium tuberculosi</i>s transmission by whole genome sequencing in a high incidence setting:A retrospective population-based study in East Greenland

    In East Greenland, a dramatic increase of tuberculosis (TB) incidence has been observed in recent years. Classical genotyping suggests a genetically similar Mycobacterium tuberculosis (Mtb) strain population as cause, however, precise transmission patterns are unclear. We performed whole genome sequencing (WGS) of Mtb isolates from 98% of culture-positive TB cases through 21 years (n = 182) which revealed four genomic clusters of the Euro-American lineage (mainly sub-lineage 4.8 (n = 134)). The time to the most recent common ancestor of lineage 4.8 strains was found to be 100 years. This sub-lineage further diversified in the 1970s, and massively expanded in the 1990s, a period of lowered TB awareness in Greenland. Despite the low genetic strain diversity, WGS data revealed several recent short-term transmission events in line with the increasing incidence in the region. Thus, the isolated setting and the uniformity of circulating Mtb strains indicated that the majority of East Greenlandic TB cases originated from one or few strains introduced within the last century. Thereby, the study shows the consequences of even short interruptions in TB control efforts in previously TB high incidence areas and demonstrates the potential role of WGS in detecting ongoing micro epidemics, thus guiding public health efforts in the future

    MicroRNA-24 regulates vascularity after myocardial infarction

    BACKGROUND: Myocardial infarction leads to cardiac remodeling and development of heart failure. Insufficient myocardial capillary density after myocardial infarction has been identified as a critical event in this process, although the underlying mechanisms of cardiac angiogenesis are mechanistically not well understood. METHODS AND RESULTS: Here, we show that the small noncoding RNA microRNA-24 (miR-24) is enriched in cardiac endothelial cells and considerably upregulated after cardiac ischemia. MiR-24 induces endothelial cell apoptosis, abolishes endothelial capillary network formation on Matrigel, and inhibits cell sprouting from endothelial spheroids. These effects are mediated through targeting of the endothelium-enriched transcription factor GATA2 and the p21-activated kinase PAK4, which were identified by bioinformatic predictions and validated by luciferase gene reporter assays. Respective downstream signaling cascades involving phosphorylated BAD (Bcl-XL/Bcl-2-associated death promoter) and Sirtuin1 were identified by transcriptome, protein arrays, and chromatin immunoprecipitation analyses. Overexpression of miR-24 or silencing of its targets significantly impaired angiogenesis in zebrafish embryos. Blocking of endothelial miR-24 limited myocardial infarct size of mice via prevention of endothelial apoptosis and enhancement of vascularity, which led to preserved cardiac function and survival. CONCLUSIONS: Our findings indicate that miR-24 acts as a critical regulator of endothelial cell apoptosis and angiogenesis and is suitable for therapeutic intervention in the setting of ischemic heart disease.     Eigenstate resolved infrared and millimeter-wave-infrared double resonance spectroscopy of methylamine in the N-H stretch Ðrst overtone region

    The eigenstate resolved, near-infrared spectrum of methylamine, has been measured at 5 MHz CH 3 NH 2 , resolution in the region near 6615 cm~1 using a molecular-beam laser spectrometer with optothermal detection. Part of the spectrum of the NH (symmetric stretch Ðrst overtone) has been assigned by applying 2l 1 a millimeter-waveÈinfrared double resonance technique. The spectra are very fractionated by intramolecular vibrational energy redistribution with an observed density of states between 20 and 30 states per cm~1 which is about four times the density of vibrational states calculated in the harmonic approximation. The coupling between the bright states and the bath is found to be rather strong (0.1 cm~1), and the lifetime for the redistribution is determined to be 3 ps for the vibration. 2l
