    A THERP/ATHEANA Analysis of the Latent Operator Error in Leaving EFW Valves Closed in the TMI-2 Accident

    This paper aims at performing a human reliability analysis using THERP (Technique for Human Error Prediction) and ATHEANA (Technique for Human Error Analysis) to develop a qualitative and quantitative analysis of the latent operator error in leaving EFW (emergency feed-water) valves closed in the TMI-2 accident. The accident analysis has revealed a series of unsafe actions that resulted in permanent loss of the unit. The integration between THERP and ATHEANA is developed in a way such as to allow a better understanding of the influence of operational context on human errors. This integration provides also, as a result, an intermediate method with the following features: (1) it allows the analysis of the action arising from the plant operational context upon the operator (as in ATHEANA), (2) it determines, as a consequence from the prior analysis, the aspects that most influence the context, and (3) it allows the change of these aspects into factors that adjust human error probabilities (as in THERP). This integration provides a more realistic and comprehensive modeling of accident sequences by considering preaccidental and postaccidental contexts, which, in turn, can contribute to more realistic PSA (Probabilistic Safety Assessment) evaluations and decision making

    ISOGO: Functional annotation of protein-coding splice variants

    The advent of RNA-seq technologies has switched the paradigm of genetic analysis from a genome to a transcriptome-based perspective. Alternative splicing generates functional diversity in genes, but the precise functions of many individual isoforms are yet to be elucidated. Gene Ontology was developed to annotate gene products according to their biological processes, molecular functions and cellular components. Despite a single gene may have several gene products, most annotations are not isoform-specifc and do not distinguish the functions of the diferent proteins originated from a single gene. Several approaches have tried to automatically annotate ontologies at the isoform level, but this has shown to be a daunting task. We have developed ISOGO (ISOform+GO function imputation), a novel algorithm to predict the function of coding isoforms based on their protein domains and their correlation of expression along 11,373 cancer patients. Combining these two sources of information outperforms previous approaches: it provides an area under precision-recall curve (AUPRC) fve times larger than previous attempts and the median AUROC of assigned functions to genes is 0.82. We tested ISOGO predictions on some genes with isoform-specifc functions (BRCA1, MADD,VAMP7 and ITSN1) and they were coherent with the literature. Besides, we examined whether the main isoform of each gene -as predicted by APPRIS- was the most likely to have the annotated gene functions and it occurs in 99.4% of the genes. We also evaluated the predictions for isoform-specifc functions provided by the CAFA3 challenge and results were also convincing. To make these results available to the scientifc community, we have deployed a web application to consult ISOGO predictions (https://biotecnun.unav. es/app/isogo).     Dynamic blockage of drippers as a function of the application of super simple phosphate and nítric acid in localized irrigation

    O superfosfato simples (SS) é uma alternativa aos adubos fosfatados que são utilizados via fertirrigação, como o ácido fosfórico, que provoca acidificação do bulbo úmido no solo, e o monofosfato de amônio purificado, que apresenta maior preço. No entanto, não se conhece o efeito de tal adubo no funcionamento de emissores, principalmente nos gotejadores, nem se é possível controlar o entupimento por SS por meio de aplicações de ácido. Assim, desenvolveu-se este trabalho com o objetivo de avaliar a possibilidade da utilização do superfosfato simples via gotejamento, como alternativa ao uso de outros adubos fosfatados, e a tentativa de controle do entupimento utilizando ácido nítrico. O trabalho foi desenvolvido em bancada de ensaios de gotejadores, localizada no Laboratório de Hidráulica da ESALQ/USP. Foram aplicadas doses de 50; 75 e 100% da solubilidade do superfosfato simples (SS), em 26 tubogotejadores de diferentes marcas comerciais, e ao fim de 360 horas de irrigação, foi aplicado ácido nítrico com pH 2, na tentativa de desentupir os gotejadores mais sensíveis. Diante dos resultados, foi possível inferir que o uso do superfosfato simples pode ser feito via fertirrigação, e o ácido nítrico, aplicado segundo esta metodologia, não proporcionou a desobstrução dos emissores.The super simple phosphate (SS) is an alternative to the phosphorous fertilizers which are used through fertigation, as the phosphoric acid, which triggers acidification of the wetted volume in the soil, and the monophosfato of purified ammonium, that has a higher price. However, the effect of such fertilizer in the emitters function, mainly in the drippers, is not known neither if it is possible to control the blockage for SS through acid applications. Thus, this study was developed in order to evaluate the possibility of the use of super simple phosphate trough drip irrigation, as an alternative to the use of other phosphorous fertilizers, and the attempt of controlling the blockage with nitric acid. The work was carried out in bench of drippers rehearsals, located at the Hydraulic laboratory in ESALQ-USP. Doses of 50; 75 and 100% of the super simple phosphate (SS) were applied in 26 emitter tubes with different commercial marks, and after 360 hours the nitric acid with a pH of 2 was applied, in the attempt of clearing the most sensitive drippers to the blockage. With the results it was possible to infer that: the use of super simple phosphate can be done by fertigation without causing damages to the system and the nitric acid, applied according to this methodology, did not provide the clearance of the emitters

    Carbon Monoxide Modulation of Microglia-Neuron Communication: Anti-Neuroinflammatory and Neurotrophic Role

Dysfunctional microglia contribute heavily to creating a toxic inflammatory milieu, a driving factor in the pathophysiology of several CNS disorders. Therefore, strategies to modulate the microglial function are required to tackle exacerbated tissue inflammation. Carbon monoxide (CO), an endogenous gaseous molecule produced by the degradation of haem, has anti-inflammatory, anti-apoptotic, and pro-homeostatic and cytoprotective roles, among others. ALF-826A, a novel molybdenum-based CO-releasing molecule, was used for the assessment of neuron-microglia remote communication. Primary cultures of rat microglia and neurons, or the BV-2 microglial and CAD neuronal murine cell lines, were used to study the microglia-neuron interaction. An approach based on microglial-derived conditioned media in neuronal culture was applied. Medium derived from CO-treated microglia provided indirect neuroprotection against inflammation by limiting the lipopolysaccharide (LPS)-induced expression of reactivity markers (CD11b), the production of reactive oxygen species (ROS) and the secretion of inflammatory factors (TNF-α, nitrites). This consequently prevented neuronal cell death and maintained neuronal morphology. In contrast, in the absence of inflammatory stimulus, conditioned media from CO-treated microglia improved neuronal morphological complexity, which is an indirect manner of assessing neuronal function. Likewise, the microglial medium also prevented neuronal cell death induced by pro-oxidant tert-Butyl hydroperoxide (t-BHP). ALF-826 treatment reinforced microglia secretion of Interleukin-10 (IL-10) and adenosine, mediators that may protect against t-BHP stress in this remote communication model. Chemical inhibition of the adenosine receptors A2A and A1 reverted the CO-derived neuroprotective effect, further highlighting a role for CO in regulating neuron-microglia communication via purinergic signalling. Our findings indicate that CO has a modulatory role on microglia-to-neuron communication, promoting neuroprotection in a non-cell autonomous manner. CO enhances the microglial release of neurotrophic factors and blocks exacerbated microglial inflammation.
