    The finite temperature QCD using 2+1 flavors of domain wall fermions at N_t = 8

    We study the region of the QCD phase transition using 2+1 flavors of domain wall fermions (DWF) and a 163×816^3 \times 8 lattice volume with a fifth dimension of Ls=32L_s = 32. The disconnected light quark chiral susceptibility, quark number susceptibility and the Polyakov loop suggest a chiral and deconfining crossover transition lying between 155 and 185 MeV for our choice of quark mass and lattice spacing. In this region the lattice scale deduced from the Sommer parameter r0r_0 is a−1≈1.3a^{-1} \approx 1.3 GeV, the pion mass is ≈300\approx 300 MeV and the kaon mass is approximately physical. The peak in the chiral susceptibility implies a pseudo critical temperature Tc=171(10)(17)T_c = 171(10)(17) MeV where the first error is associated with determining the peak location and the second with our unphysical light quark mass and non-zero lattice spacing. The effects of residual chiral symmetry breaking on the chiral condensate and disconnected chiral susceptibility are studied using several values of the valence LsL_s.Comment: 41 pages, 10 tables, 13 figure

    Transcriptome and proteomic analysis of mpox virus F3L-expressing cells

    BackgroundMonkeypox or mpox virus (mpox) is a double-stranded DNA virus that poses a significant threat to global public health security. The F3 protein, encoded by mpox, is an apoenzyme believed to possess a double-stranded RNA-binding domain (dsRBD). However, limited research has been conducted on its function. In this study, we present data on the transcriptomics and proteomics of F3L-transfected HEK293T cells, aiming to enhance our comprehension of F3L.MethodsThe gene expression profiles of pCAGGS-HA-F3L transfected HEK293T cells were analyzed using RNA-seq. Proteomics was used to identify and study proteins that interact with F3L. Real-time PCR was used to detect mRNA levels of several differentially expressed genes (DEGs) in HEK293T cells (or Vero cells) after the expression of F3 protein.ResultsA total of 14,822 genes were obtained in cells by RNA-Seq and 1,672 DEGs were identified, including 1,156 up-regulated genes and 516 down-regulated genes. A total of 27 cellular proteins interacting with F3 proteins were identified by liquid chromatography-tandem mass spectrometry (LC-MS/MS), and 19 cellular proteins with large differences in abundance ratios were considered to be candidate cellular proteins. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses showed that the DEGs were significantly enriched in immune-related pathways, including type I interferon signaling pathway, response to virus, RIG-I-like receptor signaling pathway, NOD-like receptor signaling pathway, etc. Moreover, some selected DEGs were further confirmed by real-time PCR and the results were consistent with the transcriptome data. Proteomics data show that cellular proteins interacting with F3 proteins are mainly related to RNA splicing and protein translation.ConclusionsOur analysis of transcriptomic and proteomic data showed that (1) F3L up-regulates the transcript levels of key genes in the innate immune signaling pathway, such as RIGI, MDA5, IRF5, IRF7, IRF9, ISG15, IFNA14, and elicits a broad spectrum of antiviral immune responses in the host. F3L also increases the expression of the FOS and JNK genes while decreasing the expression of TNFR2, these factors may ultimately induce apoptosis. (2) F3 protein interacts with host proteins involved in RNA splicing and protein translation, such as SNRNP70, POLR2H, HNRNPA1, DDX17, etc. The findings of this study shed light on the function of the F3 protein

    Meso-scale mixed-mode fracture modelling of reinforced concrete structures subjected to non-uniform corrosion

    Corrosion-induced concrete cracking is a significant problem affecting the durability of reinforced concrete structures. Considerable research has been carried out in addressing this problem but few have considered the cracking process of concrete as a mixed-mode fracture and the concrete as a multi-phase material. This paper develops a meso-scale mixed-mode fracture model for the cracking of concrete structures under non-uniform corrosion of reinforcement. Concrete is treated as a three-phase heterogeneous material, consisting of aggregates, mortar and interfaces. An example is worked out to demonstrate the application of the derived model and is then partially verified against previously published experimental results. In agreement with experimental results, the new model reproduces the observation that microcracks tend to form first at the interfaces before they connect to generate a discrete crack. Toughening mechanisms, e.g., microcrack shielding, crack deflection, aggregate bridging and crack overlap, have been captured in the model. Further, effects of aggregate randomness on the crack width development of concrete structures, differences between uniform and non-uniform corrosion and a comprehensive parametric study have been investigated and presented

    The Fish-Specific Protein Kinase (PKZ) Initiates Innate Immune Responses via IRF3- and ISGF3-Like Mediated Pathways

    PKZ is a fish-specific protein kinase containing Zα domains. PKZ is known to induce apoptosis through phosphorylating eukaryotic initiation factor 2α kinase (eIF2α) in the same way as double-stranded RNA-dependent protein kinase (PKR), but its exact role in detecting pathogens remains to be fully elucidated. Herein, we have found that PKZ acts as a fish-specific DNA sensor by initiating IFN expression through IRF3- or ISGF3-like mediated pathways. The expression pattern of PKZ is similar to those of innate immunity mediators stimulated by poly (dA:dT) and poly (dG:dC). DNA-PKZ interaction can enhance PKZ phosphorylation and dimerization in vitro. These findings indicate that PKZ participates in cytoplasmic DNA-mediated signaling. Subcellular localization assays have also shown that PKZ is located in the cytoplasm, which suggests that PKZ acts as a cytoplasmic PRR. Meanwhile, co-IP assays have shown that PKZ can separately interact with IRF3, STING, ZDHHC1, eIF2α, IRF9, and STAT2. Further investigations have revealed that PKZ can activate IRF3 and STAT2; and that IRF3-dependent and ISGF3-like dependent mediators are critical for PKZ-induced IFN expression. These results demonstrate that PKZ acts as a special DNA pattern-recognition receptor, and that PKZ can trigger immune responses through IRF3-mediated or ISGF3-like mediated pathways in fish

    An Alternative to Current Therapies of Functional Dyspepsia: Self-Administrated Transcutaneous Electroacupuncture Improves Dyspeptic Symptoms

    Functional dyspepsia is of high prevalence with little treatment options. The aim of this study was to develop a new treatment method using self-management transcutaneous electroacupuncture (TEA) for functional dyspepsia (FD). Twenty-eight patients with FD were enrolled and underwent a crossover clinical trial with 2-week TEA at ST36 and PC6 and 2-week sham-TEA at nonacupuncture sham-points. Questionnaires were used to assess symptoms of dyspepsia and quality of life. Physiological testing included gastric emptying and electrogastrography. It was found that (1) TEA but not sham-TEA significantly improved dyspeptic symptoms and 4 domains in quality of life; improvement was also noted in self-rated anxiety and depression scores; (2) gastric emptying was significantly and substantially increased with 2-week TEA but not sham-TEA; and (3) gastric accommodation was also improved with TEA but not sham-TEA, reflected as increased ingested nutrient volumes at the levels of satiety and maximum tolerance. These findings suggest a therapeutic potential of self-administrated TEA method for functional dyspepsia, possibly attributed to improvement in gastric motility
