    Serological survey of anti-group A rotavirus IgM in UK adults

    Rotaviral associated disease of infants in the UK is seasonal and infection in adults not uncommon but the relationship between these has been little explored. Adult sera collected monthly for one year from routine hospital samples were screened for the presence of anti-group A rotavirus immunoglobulin M class antibodies as a marker of recent infection. Anti-rotavirus IgM was seen in all age groups throughout the year with little obvious seasonal variation in the distribution of antibody levels. IgM concentrations and the proportion seropositive above a threshold both increased with age with high concentrations consistently observed in the elderly. Results suggest either high infection rates of rotavirus in adults, irrespective of seasonal disease incidence in infants, IgM persistence or IgM cross-reactivity. These results support recent evidence of differences between infant and adult rotavirus epidemiology and highlight the need for more extensive surveys to investigate age and time related infection and transmission of rotavirus

    Porcine reproductive and respiratory syndrome virus (PRRSV) in GB pig herds : farm characteristics associated with heterogeneity in seroprevalence

    Background: The between- and within-herd variability of porcine reproductive and respiratory syndrome virus (PRRSV) antibodies were investigated in a cross-sectional study of 103 British pig herds conducted 2003ā€“2004. Fifty pigs from each farm were tested for anti-PRRSV antibodies using ELISA. A binomial logistic model was used to investigate management risks for farms with and without pigs with PRRSV antibodies and multilevel statistical models were used to investigate variability in pigs' log ELISA IRPC (relative index Ɨ 100) in positive herds. Results: Thirty-five herds (34.0%) were seronegative, 41 (39.8%) were seropositive and 27 (26.2%) were vaccinated. Herds were more likely to be seronegative if they had < 250 sows (OR 3.86 (95% CI 1.46, 10.19)) and if the nearest pig herd was ā‰„ 2 miles away (OR 3.42 (95% CI 1.29, 9.12)). The mean log IRPC in seropositive herds was 3.02 (range, 0.83 ā€“ 5.58). Sixteen seropositive herds had only seropositive adult pigs. In these herds, pigs had -0.06 (95% CI -0.10, -0.01) lower log IRPC for every mile increase in distance to the nearest pig unit, and -0.56 (95% CI -1.02, -0.10) lower log IRPC when quarantine facilities were present. For 25 herds with seropositive young stock and adults, lower log IRPC were associated with isolating purchased stock for ā‰„ 6 days (coefficient - 0.46, 95% CI -0.81, -0.11), requesting ā‰„ 48 hours 'pig-free time' from humans (coefficient -0.44, 95% CI -0.79, -0.10) and purchasing gilts (coefficient -0.61, 95% CI -0.92, -0.29). Conclusion: These patterns are consistent with PRRSV failing to persist indefinitely on some infected farms, with fadeout more likely in smaller herds with little/no reintroduction of infectious stock. Persistence of infection may be associated with large herds in pig-dense regions with repeated reintroduction

    The control of Corynebacterium pseudotuberculosis infection in sheep flocks : a mathematical model of the impact of vaccination, serological testing, clinical examination and lancing of abscesses

    A mathematical model of Corynebacterium pseudotuberculosis infection in sheep flocks was used to evaluate strategies for control and elimination of caseous lymphadenitis (CIA). Control strategies tested were vaccination, serological testing and removal of seropositives, clinical examination and removal of sheep with abscesses, lancing abscesses, and appropriate combinations. Three different infection rates with and without replacement of culled ewes were used to evaluate the control options. Controls were either implemented immediately after infection was detected in a flock or once CIA was at endemic equilibrium, and with different frequencies of examination or testing. Elimination of infection was defined as 99% confidence that no sheep were infected with C. pseudo tuberculosis. The control strategies were evaluated by estimating the reduction in infection or probability of elimination and the number of ewes culled from the flock. Lancing abscesses reduced the prevalence of infection when the initial prevalence was 0.90, but vaccination combined with clinical examination reduced infection rapidly with little impact on lamb productivity. Further research is required to develop a diagnostic test with at least 0.90 specificity and sensitivity under field conditions before any methods of control can be recommended with confidence

    TARGETED POLYMERIC BIOMATERIALS FOR THE PREVENTION OF POST SURGICAL ADHESIONS

    Despite recent advances in surgical technique and the development of numerous therapeutic agents, the formation post surgical adhesions (PSA) continues to cause complications for many patients. In this research, we have employed a rational system to develop a novel treatment to address this clinical need. Based on an understanding of the biochemical events that lead to PSA formation, a series of targeted polymeric biomaterials was designed to interrupt the fibrin gel matrix propagation and suppress PSA formation. Using group transfer polymerization, a series of well controlled block copolymers of polyacrylic acid and poly(ethylene glycol-methacrylate) based materials was synthesized. Subsequent functionalization with the pentapeptide Cys-Arg-Glu-Lys-Ala (CREKA) was employed to target the materials to fibrin as a marker of pro-adhesive sites. While preliminary testing of the untargeted materials verified their ability to suppress non-specific protein adsorption to model surfaces, numerous in vitro tests were conducted to study the ability to inhibit fibrin gel propagation. The ability to inhibit both the rate and quantity of fibrinogen deposition to a fibrin coated surface has been demonstrated. In addition, the rate of fibrin gel propagation and the degree of cellular attachment can modulated. Taking advantage of the systematic variation in structure facilitated by the robust synthetic methodology employed, statistical analysis was used to elucidate the structureproperty relationships governing the performance of these materials. The most important factors that lead to enhanced performance in in vitro tests are the length of PEG chain and number of peptide units conjugated to the polymer: increasing PEG chain length and increasing the number of peptides conjugated to the polymer both improve performance in all tests. The synthetic methods that have been developed, in conjunction with the experimental results, will be used to direct future studies, including cytotoxicity and animal studies

    Thermometry and Refrigeration in a Two-Component Mott Insulator of Ultracold Atoms

    Interesting spin Hamiltonians can be realized with ultracold atoms in a two-component Mott insulator (2CMI). It was recently demonstrated that the application of a magnetic field gradient to the 2CMI enables new techniques of thermometry and adiabatic cooling. Here we present a theoretical description which provides quantitative analysis of these two new techniques.     Spin gradient thermometry for ultracold atoms in optical lattices

    We demonstrate spin gradient thermometry, a new general method of measuring the temperature of ultracold atoms in optical lattices. We realize a mixture of spins separated by a magnetic field gradient. Measurement of the width of the transition layer between the two spin domains serves as a new method of thermometry which is observed to work over a broad range of lattice depths and temperatures, including in the Mott insulator regime. We demonstrate the thermometry in a system of ultracold rubidium atoms, and suggest that interesting spin physics can be realized in this system. The lowest measured temperature is 1 nK, indicating that the system has reached the quantum regime, where insulating shells are separated by superfluid layers.Comment: 5 pages, 3 figures, minor edits for clarit

    Approximate Analytical Model for the Squeeze-Film Lubrication of the Human Ankle Joint with Synovial Fluid Filtrated by Articular Cartilage

    The aim of this article is to propose an analytical approximate squeeze-film lubrication model of the human ankle joint for a quick assessment of the synovial pressure field and the load carrying due to the squeeze motion. The model starts from the theory of boosted lubrication for the human articular joints lubrication (Walker et al., Rheum Dis 27:512ā€“520, 1968; Maroudas, Lubrication and wear in joints. Sector, London, 1969) and takes into account the fluid transport across the articular cartilage using Darcyā€™s equation to depict the synovial fluid motion through a porous cartilage matrix. The human ankle joint is assumed to be cylindrical enabling motion in the sagittal plane only. The proposed model is based on a modified Reynolds equation; its integration allows to obtain a quick assessment on the synovial pressure field showing a good agreement with those obtained numerically (Hlavacek, J Biomech 33:1415ā€“1422, 2000). The analytical integration allows the closed form description of the synovial fluid film force and the calculation of the unsteady gap thickness

    Spin gradient demagnetization cooling of ultracold atoms

    A major goal of ultracold atomic physics is quantum simulation of spin Hamiltonians in optical lattices. Progress towards this goal requires the attainment of extremely low temperatures. Here we demonstrate a new cooling method which consists of applying a time-varying magnetic field gradient to a spin mixture of ultracold atoms. We have used this method to prepare isolated spin distributions at positive and negative spin temperatures of +/-50 picokelvin. The spin system can also be used to cool other degrees of freedom, and we have used this coupling to reduce the temperature of an apparently equilibrated sample of rubidium atoms in a Mott insulating state to 350 picokelvin.     Kinetics of the neutralizing antibody response to respiratory syncytial virus infections in a birth cohort

    The kinetics of respiratory syncytial virus (RSV) neutralizing antibodies following birth, primary and secondary infections are poorly defined. The aims of the study were to measure and compare neutralizing antibody responses at different time points in a birth cohort followed-up over three RSV epidemics. Rural Kenyan children, recruited at birth between 2002 and 2003, were monitored for RSV infection over three epidemic seasons. Cord and 3-monthly sera, and acute and convalescent sera following RSV infection, were assayed in 28 children by plaque reduction neutralization test (PRNT). Relative to the neutralizing antibody titers of pre-exposure control sera (1.8ā€‰log10 PRNT), antibody titers following primary infection were (i) no different in sera collected between 0 and 0.4 months post-infection (1.9ā€‰log10 PRNT, Pā€‰=ā€‰0.146), (ii) higher in sera collected between 0.5 and 0.9 (2.8ā€‰log10 PRNT, Pā€‰<ā€‰0.0001), 1.0ā€“1.9 (2.5ā€‰log10 PRNT, Pā€‰<ā€‰0.0001), and 2.0ā€“2.9 (2.3ā€‰log10 PRNT, Pā€‰<ā€‰0.001) months post-infection, and (iii) no different in sera collected at between 3.0 and 3.9 months post-infection (2.0ā€‰log10 PRNT, Pā€‰=ā€‰0.052). The early serum neutralizing response to secondary infection (3.02ā€‰log10 PRNT) was significantly greater than the early primary response (1.9ā€‰log10 PRNT, Pā€‰<ā€‰0.0001). Variation in population-level virus transmission corresponded with changes in the mean cohort-level neutralizing titers. It is concluded that following primary RSV infection the neutralizing antibody response declines to pre-infection levels rapidly (āˆ¼3 months) which may facilitate repeat infection.     A multispecies model for the transmission and control of mastitis in dairy cows

    Mastitis in dairy cows is a significant economic and animal welfare issue in the dairy industry. The bacterial pathogens responsible for infection of the mammary gland may be split into two main categories: major and minor pathogens. Infection with major pathogens generally results in clinical illness or strong inflammatory responses and reduced milk yields, whereas minor pathogen infection is usually subclinical. Previous investigations have considered the transmission of these pathogens independently. Experimental evidence has shown cross-protection between species of pathogens. In this study a mathematical model for the coupled transmission of major and minor pathogens along with their interaction via the host was developed in order to consider various methods for controlling the incidence of major pathogen infection. A stability analysis of the model equilibria provides explanations for observed phenomena and previous decoupled modelling results. This multispecies model structure has provided a basis for quantifying the extent of cross-protection between species and assessing possible control strategies against the disease
