    Towards using NMR to screen for spoiled tomatoes stored in 1,000 L, aseptically sealed, metal-lined totes.

    Nuclear magnetic resonance (NMR) spectroscopy is used to track factory relevant tomato paste spoilage. It was found that spoilage in tomato paste test samples leads to longer spin lattice relaxation times T1 using a conventional low magnetic field NMR system. The increase in T1 value for contaminated samples over a five day room temperature exposure period prompted the work to be extended to the study of industry standard, 1,000 L, non-ferrous, metal-lined totes. NMR signals and T1 values were recovered from a large format container with a single-sided NMR sensor. The results of this work suggest that a handheld NMR device can be used to study tomato paste spoilage in factory process environments

    A study of the ozonolysis of isoprene in a cryogenic buffer gas cell by high resolution microwave spectroscopy

    We have developed a method to quantify reaction product ratios using high resolution microwave spectroscopy in a cryogenic buffer gas cell. We demonstrate the power of this method with the study of the ozonolysis of isoprene, CH2=C(CH3)-CH=CH2, the most abundant, non-methane hydrocarbon emitted into the atmosphere by vegetation. Isoprene is an asymmetric diene, and reacts with O3 at the 1,2 position to produce methyl vinyl ketone (MVK), formaldehyde, and a pair of carbonyl oxides: [CH3CO-CH=CH2 + CH2=OO] + [CH2=O + CH3COO-CH=CH2]. Alternatively, O3 could attack at the 3,4 position to produce methacrolein (MACR), formaldehyde, and two carbonyl oxides [CH2=C(CH3)-CHO + CH2=OO] + [CH2=O + CH2=C(CH3)-CHOO]. Purified O3 and isoprene were mixed for approximately 10 seconds under dilute (1.5-4% in argon) continuous flow conditions in an alumina tube held at 298 K and 5 Torr. Products exiting the tube were rapidly slowed and cooled within the buffer gas cell by collisions with cryogenic (4-7 K) He. High resolution chirped pulse microwave detection between 12 and 26 GHz was used to achieve highly sensitive (ppb scale), isomer-specific product quantification. We observed a ratio of MACR to MVK of 2.1 +/- 0.4 under 1:1 ozone to isoprene conditions and 2.1 +/- 0.2 under 2:1 ozone to isoprene conditions, a finding which is consistent with previous experimental results. Additionally, we discuss relative quantities of formic acid (HCOOH), an isomer of CH2=OO, and formaldehyde (CH2=O) under varying experimental conditions, and characterize the spectroscopic parameters of the singly-substituted 13C trans-isoprene and 13C anti-periplanar-methacrolein species. This work has the potential to be extended towards a complete branching ratio analysis, as well towards the ability to isolate, identify, and quantify new reactive intermediates in the ozonolysis of alkenes

    AbstractPrevious studies have shown that the pattern and degree of 3-(trifluoromethyl)-3-(m-[125I]iodophenyl)diazirine ([125I]TID) photoincorporation into the nicotinic acetylcholine receptor (nAChR) can be used as a sensitive measure of nAChR conformation. Upon desensitization by prolonged exposure to agonists, certain drugs and detergents, or reconstitution into desensitizing lipids, the levels of [125I]TID incorporation into the subunits of the nAChR are dramatically reduced. In this study, we characterized the effects of the snake venom proteins α-bungarotoxin and α-cobrotoxin, as well as the smaller antagonists tubocurarine and gallamine, on [125I]TID incorporation into the subunits of both partially-purified nAChR in native lipids, or affinity-purified nAChR reconstituted into different combinations of lipids. Unlike all other compounds previously tested, α-bungarotoxin and α-cobrotoxin reproducibly increased the level of [125I]TID incorporation into all four subunits of nAChR reconstituted into dioleoylphosphatidylcholine, dioleoylphosphatidic acid and cholesterol. Gallamine had little or no effect on [125I]TID incorporation at any concentration tested (0.1 μM—5 mM). Tubocurarine had no effect on [125I]TID incorporation at low concentrations, but at higher concentrations reduced the level of [125I]TID labeling. The snake venom proteins may shift the population of nAChR, which exists as a mixture of resting state and desensitized conformations, entirely to the resting state. However, the binding of the snake venom toxins does not appear sufficient to induce the resting state conformation in nAChR which have been desensitized by other means, such as solubilization in desensitizing detergents or reconstitution in desensitizing lipids

    Reliability of Relative Predictions in Population Viability Analysis

    Despite numerous claims that population viability analysis (PVA) makes reliable predictions of the relative risks of extinction, there is little evidence to support this assertion. To assess the veracity of the claim, we investigated uncertainty in the relative predictions of a PVA model with simulation experiments. We used a stochastic Ricker model to investigate the reliability of predicted changes in risks of decline in response to changes in parameters, the reliability of ranking species in terms of their relative threat, and the reliability of choosing the better of two management decisions. The predicted changes in risks of decline within 100 years were more reliable than absolute predictions. We made useful predictions of relative risks using only 10 years of data. Across 160 different parameter combinations, the rank correlation between the true risks of extinction within 100 years and predicted risks was 0.59 with 10 years of data, increasing to 0.89 with 100 years of data. We identified the better of two management strategies 67 74% of the time using 10 years of data, increasing to 92 93% of the time with 100 years of data. Our results demonstrate that, despite considerable uncertainty in the predicted risks of decline, PVA may reliably contribute to the management of threatened species

We report such a case involving bladder herniation and subsequent entrapment after attempted closed reduction with anterior pelvic external fixation immediately treated with open reduction and internal fixation along with a review of the literature

    Models of the ICM with Heating and Cooling: Explaining the Global and Structural X-ray Properties of Clusters

    (Abridged) Theoretical models that include only gravitationally-driven processes fail to match the observed mean X-ray properties of clusters. As a result, there has recently been increased interest in models in which either radiative cooling or entropy injection play a central role in mediating the properties of the intracluster medium. Both sets of models give reasonable fits to the mean properties of clusters, but cooling only models result in fractions of cold baryons in excess of observationally established limits and the simplest entropy injection models do not treat the "cooling core" structure present in many clusters and cannot account for entropy profiles revealed by recent X-ray observations. We consider models that marry radiative cooling with entropy injection, and confront model predictions for the global and structural properties of massive clusters with the latest X-ray data. The models successfully and simultaneously reproduce the observed L-T and L-M relations, yield detailed entropy, surface brightness, and temperature profiles in excellent agreement with observations, and predict a cooled gas fraction that is consistent with observational constraints. The model also provides a possible explanation for the significant intrinsic scatter present in the L-T and L-M relations and provides a natural way of distinguishing between clusters classically identified as "cooling flow" clusters and dynamically relaxed "non-cooling flow" clusters. The former correspond to systems that had only mild levels (< 300 keV cm^2) of entropy injection, while the latter are identified as systems that had much higher entropy injection.     A Simple Landscape-Scale Test of a Spatially Explicit Population Model: Patch Occupancy in Fragmented South-Eastern Australian Forests

    The results of a landscape-scale test of ALEX, a widely used metapopulation model for Population Viability Analysis (PVA), are described. ALEX was used to predict patch occupancy by the laughing kookaburra and the sacred kingfisher in patches of eucalypt forest in south-eastern Australia. These predictions were compared to field surveys to determine the accuracy of the model. Predictions also were compared to a "naïve" null model assuming no fragmentation effects. The naive null model significantly over-predicted the number of eucalypt patches occupied by the sacred kingfisher, but the observed patch occupancy was not significantly different from that predicted using ALEX. ALEX produced a better fit to the field data than the naive null model for the number of patches occupied by the laughing kookaburra. Nevertheless, ALEX still significantly over-predicted the number of occupied patches, particularly remnants dominated by certain forest types – ribbon gum and narrow-leaved peppermint. The predictions remained significantly different from observations, even when the habitat quality of these patches was reduced to zero. Changing the rate of dispersal improved overall predicted patch occupancy, but occupancy rates for the different forest types remained significantly different from the field observations. The lack of congruence between field data and model predictions could have arisen because the laughing kookaburra may move between an array of patches to access spatially separated food and nesting resources in response to fragmentation. Alternatively, inter-specific competition may be heightened in a fragmented habitat. These types of responses to fragmentation are not incorporated as part of traditionally applied metapopulation models. Assessments of predictions from PVA models are rare but important because they can reveal the types of species for which forecasts are accurate and those for which they are not. This can assist the collection of additional empirical data to identify important factors affecting population dynamics

    Cluster Sunyaev-Zeldovich Effect Scaling Relations

    X-ray observations of an "entropy floor" in nearby groups and clusters of galaxies offer evidence that important non-gravitational processes, such as radiative cooling and/or "preheating", have strongly influenced the evolution of the intracluster medium (ICM). We examine how the presence of an entropy floor modifies the thermal Sunyaev-Zeldovich (SZ) effect. A detailed analysis of scaling relations between X-ray and SZ effect observables and also between the two primary SZ effect observables is presented. We find that relationships between the central Compton parameter and the temperature or mass of a cluster are extremely sensitive to the presence of an entropy floor. The same is true for correlations between the integrated Compton parameter and the X-ray luminosity or the central Compton parameter. In fact, if the entropy floor is as high as inferred in recent analyses of X-ray data, a comparison of these correlations with both current and future SZ effect observations should show a clear signature of this excess entropy. Moreover, because the SZ effect is redshift-independent, the relations can potentially be used to track the evolution of the cluster gas and possibly discriminate between the possible sources of the excess entropy.
