    Rigidity and volume preserving deformation on degenerate simplices

    Given a degenerate (n+1)(n+1)-simplex in a dd-dimensional space MdM^d (Euclidean, spherical or hyperbolic space, and dā‰„nd\geq n), for each kk, 1ā‰¤kā‰¤n1\leq k\leq n, Radon's theorem induces a partition of the set of kk-faces into two subsets. We prove that if the vertices of the simplex vary smoothly in MdM^d for d=nd=n, and the volumes of kk-faces in one subset are constrained only to decrease while in the other subset only to increase, then any sufficiently small motion must preserve the volumes of all kk-faces; and this property still holds in MdM^d for dā‰„n+1d\geq n+1 if an invariant ckāˆ’1(Ī±kāˆ’1)c_{k-1}(\alpha^{k-1}) of the degenerate simplex has the desired sign. This answers a question posed by the author, and the proof relies on an invariant ck(Ļ‰)c_k(\omega) we discovered for any kk-stress Ļ‰\omega on a cell complex in MdM^d. We introduce a characteristic polynomial of the degenerate simplex by defining f(x)=āˆ‘i=0n+1(āˆ’1)ici(Ī±i)xn+1āˆ’if(x)=\sum_{i=0}^{n+1}(-1)^{i}c_i(\alpha^i)x^{n+1-i}, and prove that the roots of f(x)f(x) are real for the Euclidean case.     When good bugs go bad: Epidemiology and antimicrobial resistance profiles of Corynebacterium striatum, an emerging multidrug-resistant, opportunistic pathogen

    ABSTRACT Infections with Corynebacterium striatum have been described in the literature over the last 2 decades, with the majority being bacteremia, central line infections, and occasionally, endocarditis. In recent years, the frequency of C. striatum infections appears to be increasing; a factor likely contributing to this is the increased ease and accuracy of the identification of Corynebacterium spp., including C. striatum , from clinical cultures. The objective of this study was to retrospectively characterize C. striatum isolates recovered from specimens submitted as part of routine patient care at a 1,250-bed, tertiary-care academic medical center. Multiple strain types were recovered, as demonstrated by repetitive-sequence-based PCR. Most of the strains of C. striatum characterized were resistant to antimicrobials commonly used to treat Gram-positive organisms, such as penicillin, ceftriaxone, meropenem, clindamycin, and tetracycline. The MIC 50 for ceftaroline was &gt;32 Ī¼g/ml. Although there are no interpretive criteria for susceptibility with telavancin, it appeared to have potent in vitro efficacy against this species, with MIC 50 and MIC 90 values of 0.064 and 0.125 Ī¼g/ml, respectively. Finally, as previously reported in case studies, we demonstrated rapid in vitro development of daptomycin resistance in 100% of the isolates tested ( n = 50), indicating that caution should be exhibited when using daptomycin for the treatment of C. striatum infections. C. striatum is an emerging, multidrug-resistant pathogen that can be associated with a variety of infection types. </jats:p

    Diseased myocardium from humans and experimental animal models shows heightened expression and activity of a specific subtype of phospholipase C (PLC), the splice variant PLCĪ²1b. Previous studies from our group showed that increasing PLCĪ²1b expression in adult mouse hearts by viral transduction was sufficient to cause sustained contractile dysfunction of rapid onset, which was maintained indefinitely in the absence of other pathological changes in the myocardium. We hypothesized that impaired contractility alone would be sufficient to induce a compensatory transcriptional response. Unbiased, comprehensive mRNA-sequencing was performed on 6 biological replicates of rAAV6-treated blank, PLCĪ²1b and PLCĪ²1a (closely related but inactive splice variant) hearts 8 weeks after injection, when reduced contractility was manifest in PLCĪ²1b hearts without evidence of induced hypertrophy. Expression of PLCĪ²1b resulted in expression changes in only 9 genes at FDR<0.1 when compared with control and these genes appeared unrelated to contractility. Importantly, PLCĪ²1a caused similar mild expression changes to PLCĪ²1b, despite a complete lack of effect of this isoform on cardiac contractility. We conclude that contractile depression caused by PLCĪ²1b activation is largely independent of changes in the transcriptome, and thus that lowered contractility is not sufficient in itself to provoke measurable transcriptomic alterations. In addition, our data stress the importance of a stringent control group to filter out transcriptional changes unrelated to cardiac function

    Comparison of sample preparation methods, instrumentation platforms, and contemporary commercial databases for identification of clinically relevant mycobacteria by matrix-assisted laser desorption ionization - Time of flight mass spectrometry

    When mycobacteria are recovered in clinical specimens, timely species-level identification is required to establish the clinical significance of the isolate and facilitate optimization of antimicrobial therapy. Matrix-assisted laser desorption ionizationā€“time of flight mass spectrometry (MALDI-TOF MS) has recently been reported to be a reliable and expedited method for identification of mycobacteria, although various specimen preparation techniques and databases for analysis are reported across studies. Here we compared two MALDI-TOF MS instrumentation platforms and three databases: Bruker Biotyper Real Time Classification 3.1 (Biotyper), Vitek MS Plus Saramis Premium (Saramis), and Vitek MS v3.0. We evaluated two sample preparation techniques and demonstrate that extraction methods are not interchangeable across different platforms or databases. Once testing parameters were established, a panel of 157 mycobacterial isolates (including 16 Mycobacterium tuberculosis isolates) was evaluated, demonstrating that with the appropriate specimen preparation, all three methods provide reliable identification for most species. Using a score cutoff value of ā‰„1.8, the Biotyper correctly identified 133 (84.7%) isolates with no misidentifications. Using a confidence value of ā‰„90%, Saramis correctly identified 134 (85.4%) isolates with one misidentification and Vitek MS v3.0 correctly identified 140 (89.2%) isolates with one misidentification. The levels of accuracy were not significantly different across the three platforms (P = 0.14). In addition, we show that Vitek MS v3.0 requires modestly fewer repeat analyses than the Biotyper and Saramis methods (P = 0.04), which may have implications for laboratory workflow
