    Targeted nasal vaccination provides antibody-independent protection against Staphylococcus aureus.

    Despite showing promise in preclinical models, anti-Staphylococcus aureus vaccines have failed in clinical trials. To date, approaches have focused on neutralizing/opsonizing antibodies; however, vaccines exclusively inducing cellular immunity have not been studied to formally test whether a cellular-only response can protect against infection. We demonstrate that nasal vaccination with targeted nanoparticles loaded with Staphylococcus aureus antigen protects against acute systemic S. aureus infection in the absence of any antigen-specific antibodies. These findings can help inform future developments in staphylococcal vaccine development and studies into the requirements for protective immunity against S. aureus

    Memory Th1 Cells Are Protective in Invasive Staphylococcus aureus Infection

    Mechanisms of protective immunity to Staphylococcus aureus infection in humans remain elusive. While the importance of cellular immunity has been shown in mice, T cell responses in humans have not been characterised. Using a murine model of recurrent S. aureus peritonitis, we demonstrated that prior exposure to S. aureus enhanced IFNγ responses upon subsequent infection, while adoptive transfer of S. aureus antigen-specific Th1 cells was protective in naïve mice. Translating these findings, we found that S. aureus antigen-specific Th1 cells were also significantly expanded during human S. aureus bloodstream infection (BSI). These Th1 cells were CD45RO+, indicative of a memory phenotype. Thus, exposure to S. aureus induces memory Th1 cells in mice and humans, identifying Th1 cells as potential S. aureus vaccine targets. Consequently, we developed a model vaccine comprising staphylococcal clumping factor A, which we demonstrate to be an effective human T cell antigen, combined with the Th1-driving adjuvant CpG. This novel Th1-inducing vaccine conferred significant protection during S. aureus infection in mice. This study notably advances our understanding of S. aureus cellular immunity, and demonstrates for the first time that a correlate of S. aureus protective immunity identified in mice may be relevant in humans

    Liquid–liquid phase separation morphologies in ultra-white beetle scales and a synthetic equivalent

    Cyphochilus beetle scales are amongst the brightest structural whites in nature, being highly opacifying whilst extremely thin. However, the formation mechanism for the voided intra-scale structure is unknown. Here we report 3D x-ray nanotomography data for the voided chitin networks of intact white scales of Cyphochilus and Lepidiota stigma. Chitin-filling fractions are found to be 31 ± 2% for Cyphochilus and 34 ± 1% for Lepidiota stigma, indicating previous measurements overestimated their density. Optical simulations using finite-difference time domain for the chitin morphologies and simulated Cahn-Hilliard spinodal structures show excellent agreement. Reflectance curves spanning filling fraction of 5-95% for simulated spinodal structures, pinpoint optimal whiteness for 25% chitin filling. We make a simulacrum from a polymer undergoing a strong solvent quench, resulting in highly reflective (~94%) white films. In-situ X-ray scattering confirms the nanostructure is formed through spinodal decomposition phase separation. We conclude that the ultra-white beetle scale nanostructure is made via liquid–liquid phase separation

    ‘We Are Not As They Think About Us’: Exploring Omani EFL Learners’ ‘Selves’ in Digital Social Spaces

    This paper reports a research study of Omani EFL learners’ motivation to engage in social technologies through the medium of English, adopting Dörnyei’s (2009) L2 Motivational Self System as the main theoretical framework, whilst exploring other emergent context-sensitive motivational driving forces. The purpose of the research study was to explore identities and self-perceptions of Omani nationals using social media to learn English, an under-researched context. Reflective focused group discussions were conducted with 14 university-age students, along with individually composed language learning histories. The data of the research lends substantial support to the relevance of the L2 Motivational Self System in the Omani context, highlighting, in particular, the emerging collective national and religious identities of young Omani nationals. The paper argues for the need to deepen and broaden our understanding of the association of English social technologies and the national and religious affiliations of learners

    Siluro-Devonian trace fossils from the Mereenie Sandstone, Kings Canyon, Watarrka National Park, Amadeus Basin, Northern Territory, Australia

    Nine trackways referable to the ichnogenus Diplichnites are preserved in the upper Silurian to Lower Devonian Mereenie Sandstone at Kings Canyon, Watarrka National Park, Northern Territory, Australia. Eight trackways are consistent with earlier descriptions of D. gouldi, and one trackway could not be assigned to an ichnospecies. The trackways are co-preserved with a range of sub-horizontal burrows referable to Beaconites and Taenidium, and several vertical burrows, surficial circular traces and a horizontal trail of uncertain identities. The ichnofossil assemblage highlights the diversity of animals present in the late Silurian to Lower Devonian paralic to fluvial environments of central Australia at the time of early colonization of the land’s surface. The assemblage is similar to ichnofaunas from coeval strata elsewhere in Australia and throughout Gondwana, and it highlights the potential of this region for further ichnological studies to elucidate the early stages of terrestrialization in the palaeoequatorial belt
