    A Brief Review of Addictive Tendencies Related to Technology Use: Conceptualization, Treatment, and Future Directions

    Addictive tendencies relating to technology use entail the overuse and misuse of physical and digital devices to the point of maladjustment. Such tendencies, which can arise from video game use, mobile phone use, Internet use, and media streaming/television use, have major implications regarding people’s physiological and psychological states. Although prior research helped with the conceptual and empirical understanding of technology use, these had major limitations. Such limitations included inconsistent terminology (e.g., technology use disorder versus technological addictions), a lack of standardized criteria to diagnose or recognize addictive tendencies of technology use, differences in methodology (e.g., longitudinal studies, experimental studies, case studies, correlational studies), and construct proliferation (e.g., smartphone addiction, young adult attachment to phone). In addition, little research has been conducted regarding the effectiveness of treatments (e.g., psychopharmacological treatments) for tendencies of technology use. Studies regarding treatment efficacy have primarily been limited to case studies with small sample sizes. Ultimately, it is recommended that researchers form a unified front to address these addictive tendencies through consistent theoretical models, research, and criteria

    Safety evaluation of substituted thiophenes used as flavoring ingredients

    AbstractThis publication is the second in a series by the Expert Panel of the Flavor and Extract Manufacturers Association summarizing the conclusions of its third systematic re-evaluation of the safety of flavorings previously considered to be generally recognized as safe (GRAS) under conditions of intended use. Re-evaluation of GRAS status for flavorings is based on updated considerations of exposure, structural analogy, metabolism, pharmacokinetics and toxicology and includes a comprehensive review of the scientific information on the flavorings and structurally related substances. Of the 12 substituted thiophenes reviewed here, 11 were reaffirmed as GRAS based on their rapid absorption, metabolism and excretion in humans and animals; the low estimated dietary exposure from flavor use; the wide margins of safety between the conservative estimates of intake and the no-observed-adverse effect levels; and the lack of significant genotoxic and mutagenic potential. For one of the substituted thiophenes, 3-acetyl-2,5-dimethylthiophene, it was concluded that more detailed exposure information, comparative metabolism studies and comprehensive toxicity data, including an in-depth evaluation of the mechanism of action for any adverse effects observed, are required for continuation of its FEMA GRAS™ status. In the absence of these data, the compound was removed from the FEMA GRAS list

    Teacher Tasks for Mathematical Insight and Reorganization of What it Means to Learn Mathematics

    The mathematics-for-teachers tasks we discuss in this chapter have two qualities: (1) they offer teachers opportunities to experience the pleasure of mathematical insight; and (2) they aim to disrupt and reorganize teachers\u27 views of what it means to do and learn mathematics. Given that many future and inservice elementary teachers fear and dislike mathematics, it is perhaps not too far-fetched to suggest that there is a need for “math therapy.” We believe that a form of mathematics therapy may involve new and different experiences with mathematics. Such experiences, considered broadly to include questions or prompts for mathematical exploration, draw attention to deep mathematical ideas and offer the potential of experiencing the pleasure of significant mathematical insight. In our work with teachers we have developed and used a variety of mathematics tasks as opportunities for experiential therapy. The tasks aim to challenge some of the mathematical myths that future teachers believe to be true and are typically assumed by them in mathematics classrooms. The tasks have potential to disrupt teachers\u27 view of mathematics, and to start the process for reorganizing their thinking about what mathematics is and what it means to do and learn mathematics. In this chapter we describe and discuss four of the mathematics tasks which involve non-routine mathematics problems that we use in our mathematics-for-teachers program. This program is offered annually to our 440 future elementary school (K-8) teachers, who generally lack confidence in mathematics and often fear and/or dislike the subject. It is also offered to inservice teachers through a series of mathematics-for-teachers courses. A student response summarizes the effects of our approach

    Maximum likelihood models and algorithms for gene tree evolution with duplications and losses

    <p>Abstract</p> <p>Background</p> <p>The abundance of new genomic data provides the opportunity to map the location of gene duplication and loss events on a species phylogeny. The first methods for mapping gene duplications and losses were based on a parsimony criterion, finding the mapping that minimizes the number of duplication and loss events. Probabilistic modeling of gene duplication and loss is relatively new and has largely focused on birth-death processes.</p> <p>Results</p> <p>We introduce a new maximum likelihood model that estimates the speciation and gene duplication and loss events in a gene tree within a species tree with branch lengths. We also provide an, in practice, efficient algorithm that computes optimal evolutionary scenarios for this model. We implemented the algorithm in the program DrML and verified its performance with empirical and simulated data.</p> <p>Conclusions</p> <p>In test data sets, DrML finds optimal gene duplication and loss scenarios within minutes, even when the gene trees contain sequences from several hundred species. In many cases, these optimal scenarios differ from the lca-mapping that results from a parsimony gene tree reconciliation. Thus, DrML provides a new, practical statistical framework on which to study gene duplication.</p

    A supermatrix analysis of genomic, morphological, and paleontological data from crown Cetacea

    <p>Abstract</p> <p>Background</p> <p>Cetacea (dolphins, porpoises, and whales) is a clade of aquatic species that includes the most massive, deepest diving, and largest brained mammals. Understanding the temporal pattern of diversification in the group as well as the evolution of cetacean anatomy and behavior requires a robust and well-resolved phylogenetic hypothesis. Although a large body of molecular data has accumulated over the past 20 years, DNA sequences of cetaceans have not been directly integrated with the rich, cetacean fossil record to reconcile discrepancies among molecular and morphological characters.</p> <p>Results</p> <p>We combined new nuclear DNA sequences, including segments of six genes (~2800 basepairs) from the functionally extinct Yangtze River dolphin, with an expanded morphological matrix and published genomic data. Diverse analyses of these data resolved the relationships of 74 taxa that represent all extant families and 11 extinct families of Cetacea. The resulting supermatrix (61,155 characters) and its sub-partitions were analyzed using parsimony methods. Bayesian and maximum likelihood (ML) searches were conducted on the molecular partition, and a molecular scaffold obtained from these searches was used to constrain a parsimony search of the morphological partition. Based on analysis of the supermatrix and model-based analyses of the molecular partition, we found overwhelming support for 15 extant clades. When extinct taxa are included, we recovered trees that are significantly correlated with the fossil record. These trees were used to reconstruct the timing of cetacean diversification and the evolution of characters shared by "river dolphins," a non-monophyletic set of species according to all of our phylogenetic analyses.</p> <p>Conclusions</p> <p>The parsimony analysis of the supermatrix and the analysis of morphology constrained to fit the ML/Bayesian molecular tree yielded broadly congruent phylogenetic hypotheses. In trees from both analyses, all Oligocene taxa included in our study fell outside crown Mysticeti and crown Odontoceti, suggesting that these two clades radiated in the late Oligocene or later, contra some recent molecular clock studies. Our trees also imply that many character states shared by river dolphins evolved in their oceanic ancestors, contradicting the hypothesis that these characters are convergent adaptations to fluvial habitats.</p

    Mucosal Targeting of a BoNT/A Subunit Vaccine Adjuvanted with a Mast Cell Activator Enhances Induction of BoNT/A Neutralizing Antibodies in Rabbits

    We previously reported that the immunogenicity of Hcβtre, a botulinum neurotoxin A (BoNT/A) immunogen, was enhanced by fusion to an epithelial cell binding domain, Ad2F, when nasally delivered to mice with cholera toxin (CT). This study was performed to determine if Ad2F would enhance the nasal immunogenicity of Hcβtre in rabbits, an animal model with a nasal cavity anatomy similar to humans. Since CT is not safe for human use, we also tested the adjuvant activity of compound 48/80 (C48/80), a mast cell activating compound previously determined to safely exhibit nasal adjuvant activity in mice.New Zealand White or Dutch Belted rabbits were nasally immunized with Hcβtre or Hcβtre-Ad2F alone or combined with CT or C48/80, and serum samples were tested for the presence of Hcβtre-specific binding (ELISA) or BoNT/A neutralizing antibodies.Hcβtre-Ad2F nasally administered with CT induced serum anti-Hcβtre IgG ELISA and BoNT/A neutralizing antibody titers greater than those induced by Hcβtre + CT. C48/80 provided significant nasal adjuvant activity and induced BoNT/A-neutralizing antibodies similar to those induced by CT.Ad2F enhanced the nasal immunogenicity of Hcβtre, and the mast cell activator C48/80 was an effective adjuvant for nasal immunization in rabbits, an animal model with a nasal cavity anatomy similar to that in humans
