To supplement or not to supplement? The rationale of vitamin D supplementation in systemic lupus erythematosus

Background: Systemic Lupus Erythematosus (SLE) is a systemic autoimmune disease characterised by abnormal activation of the immune system, chronic inflammation and organ damage. Lupus patients are more prone to be vitamin D deficient. However, current evidence is not conclusive with regards to the role played by vitamin D in SLE development, progression, and clinical manifestations. Objective: Here, we will summarise the current knowledge about vitamin D deficiency prevalence, risk factors, molecular effects, and potential pathogenic role in SLE. We will focus on the link between vitamin D deficiency and lupus clinical manifestations, and on the clinical trials assessing the effects of vitamin D supplementation in SLE. Method: A detailed literature search was performed exploiting the available databases, using “vitamin D and lupus/SLE” as keywords. The relevant interventional trials published over the last decade have been considered and the results are reported here. Conclusion: Several immune cells express vitamin D receptors. Thus, an immunomodulatory role for vitamin D in lupus is plausible. Numerous observational studies have investigated the relationship between vitamin D levels and clinical/serological manifestations of SLE with contrasting results. Negative correlations between vitamin D levels and disease activity, fatigue, renal and cardiovascular disease, and anti-dsDNA titres have been described but not conclusively accepted. In experimental models of lupus, vitamin D supplementation can improve the disease. Interventional trials have assessed the potential therapeutic value of vitamin D in SLE, but further larger studies are needed
Impact of Age and Body Site on Adult Female Skin Surface pH
Background: pH is known as an important parameter in epidermal barrier function and homeostasis. Aim: The impact of age and body site on skin surface pH (pH(SS)) of women was evaluated in vivo. Methods: Time domain dual lifetime referencing with luminescent sensor foils was used for pH(SS) measurements. pH(SS) was measured on the forehead, the temple, and the volar forearm of adult females (n = 97, 52.87 +/- 18.58 years, 20-97 years). Every single measurement contained 2,500 pH values due to the luminescence imaging technique used. Results: pH(SS) slightly increases with age on all three investigated body sites. There are no significant differences in pH(SS) between the three investigated body sites. Conclusion: Adult pH(SS) on the forehead, the temple and the volar forearm increases slightly with age. Anomalous Couplings in Double Higgs Production
The process of gluon-initiated double Higgs production is sensitive to
non-linear interactions of the Higgs boson. In the context of the Standard
Model, studies of this process focused on the extraction of the Higgs trilinear
coupling. In a general parametrization of New Physics effects, however, an even
more interesting interaction that can be tested through this channel is the
(ttbar hh) coupling. This interaction vanishes in the Standard Model and is a
genuine signature of theories in which the Higgs boson emerges from a
strongly-interacting sector. In this paper we perform a model-independent
estimate of the LHC potential to detect anomalous Higgs couplings in
gluon-fusion double Higgs production. We find that while the sensitivity to the
trilinear is poor, the perspectives of measuring the new (ttbar hh) coupling

Eradication of chronic myeloid leukemia stem cells: a novel mathematical model predicts no therapeutic benefit of adding G-CSF to imatinib
Imatinib mesylate induces complete cytogenetic responses in patients with chronic myeloid leukemia (CML), yet many patients have detectable BCR-ABL transcripts in peripheral blood even after prolonged therapy. Bone marrow studies have shown that this residual disease resides within the stem cell compartment. Quiescence of leukemic stem cells has been suggested as a mechanism conferring insensitivity to imatinib, and exposure to the Granulocyte-Colony Stimulating Factor (G-CSF), together with imatinib, has led to a significant reduction in leukemic stem cells in vitro. In this paper, we design a novel mathematical model of stem cell quiescence to investigate the treatment response to imatinib and G-CSF. We find that the addition of G-CSF to an imatinib treatment protocol leads to observable effects only if the majority of leukemic stem cells are quiescent; otherwise it does not modulate the leukemic cell burden. The latter scenario is in agreement with clinical findings in a pilot study administering imatinib continuously or intermittently, with or without G-CSF (GIMI trial). Furthermore, our model predicts that the addition of G-CSF leads to a higher risk of resistance since it increases the production of cycling leukemic stem cells. Although the pilot study did not include enough patients to draw any conclusion with statistical significance, there were more cases of progression in the experimental arms as compared to continuous imatinib. Our results suggest that the additional use of G-CSF may be detrimental to patients in the clinic

Formyl Peptide Receptor as a Novel Therapeutic Target for Anxiety-Related Disorders
Formyl peptide receptors (FPR) belong to a family of sensors of the immune system that detect microbe-associated molecules and inform various cellular and sensorial mechanisms to the presence of pathogens in the host. Here we demonstrate that Fpr2/3-deficient mice show a distinct profile of behaviour characterised by reduced anxiety in the marble burying and light-dark box paradigms, increased exploratory behaviour in an open-field, together with superior performance on a novel object recognition test. Pharmacological blockade with a formyl peptide receptor antagonist, Boc2, in wild type mice reproduced most of the behavioural changes observed in the Fpr2/3(-/-) mice, including a significant improvement in novel object discrimination and reduced anxiety in a light/dark shuttle test. These effects were associated with reduced FPR signalling in the gut as shown by the significant reduction in the levels of p-p38. Collectively, these findings suggest that homeostatic FPR signalling exerts a modulatory effect on anxiety-like behaviours. These findings thus suggest that therapies targeting FPRs may be a novel approach to ameliorate behavioural abnormalities present in neuropsychiatric disorders at the cognitive-emotional interface
Strong Double Higgs Production at the LHC
The hierarchy problem and the electroweak data, together, provide a plausible
motivation for considering a light Higgs emerging as a pseudo-Goldstone boson
from a strongly-coupled sector. In that scenario, the rates for Higgs
production and decay differ significantly from those in the Standard Model.
However, one genuine strong coupling signature is the growth with energy of the
scattering amplitudes among the Goldstone bosons, the longitudinally polarized
vector bosons as well as the Higgs boson itself. The rate for double Higgs
production in vector boson fusion is thus enhanced with respect to its
negligible rate in the SM. We study that reaction in pp collisions, where the
production of two Higgs bosons at high pT is associated with the emission of
two forward jets. We concentrate on the decay mode hh -> WW^(*)WW^(*) and study
the semi-leptonic decay chains of the W's with 2, 3 or 4 leptons in the final
states. While the 3 lepton final states are the most relevant and can lead to a
3 sigma signal significance with 300 fb^{-1} collected at a 14 TeV LHC, the two
same-sign lepton final states provide complementary information. We also
comment on the prospects for improving the detectability of double Higgs
