    Selected Hydrologic Applications of LANDSAT-2 Data: an Evaluation

    The author has identified the following significant results. Estimates of soil moisture were obtained from visible, near-IR gamma ray and microwave data. Attempts using GOES thermal-IR were unsuccessful due to resolutions (8 km). Microwaves were the most effective at soil moisture estimates, with and without vegetative cover. Gamma rays provided only one value for the test site, produced by many data points obtained from overlapping 150 meter diameter circles. Even though the resulting averaged value was near the averaged field moisture value, this method suffers from atmospheric contaminants, the need to fly at low altitudes, and the necessity of prior calibration of a given site. Visible and near-IR relationships are present for bare fields but appear to be limited to soil moisture levels between 5 and 20%. The densely vegetated alfalfa fields correlated with near-IR reflectance only; soil moisture values from wheat fields showed no relation to either or near-IR MSS data

    A Partially Functional DNA Helicase II Mutant Defective in Forming Stable Binary Complexes with ATP and DNA: A ROLE FOR HELICASE MOTIF III

    To address the functional significance of motif III in Escherichia coli DNA helicase II, the conserved aspartic acid at position 248 was changed to asparagine. UvrDD248N failed to form stable binary complexes with either DNA or ATP. However, UvrDD248N was capable of forming an active ternary complex when both ATP and single-stranded DNA were present. The DNA-stimulated ATPase activity of UvrDD248N was reduced relative to that of wild-type UvrD with no significant change in the apparent Km for ATP. The mutant protein also demonstrated a reduced DNA unwinding activity. The requirement for high concentrations of UvrDD248N to achieve unwinding of long duplex substrates likely reflects the reduced stability of various binary and ternary complexes that must exist in the catalytic cycle of a helicase. The data suggest that motif III may act as an interface between the ATP binding and DNA binding domains of a helicase. The uvrDD248N allele was also characterized in genetic assays. The D248N protein complemented the UV-sensitive phenotype of a uvrD deletion strain to levels nearly equivalent to wild-type helicase II. In contrast, the mutant protein only partially complemented the mutator phenotype. A correlation between the level of genetic complementation and the helicase activity of UvrDD248N is discussed

    A Point Mutation in Escherichia coli DNA Helicase II Renders the Enzyme Nonfunctional in Two DNA Repair Pathways: EVIDENCE FOR INITIATION OF UNWINDING FROM A NICKIN VIVO

    Biosynthetic errors and DNA damage introduce mismatches and lesions in DNA that can lead to mutations. These abnormalities are susceptible to correction by a number of DNA repair mechanisms, each of which requires a distinct set of proteins. Escherichia coli DNA helicase II has been demonstrated to function in two DNA repair pathways, methyl-directed mismatch repair and UvrABC-mediated nucleotide excision repair. To define further the role of UvrD in DNA repair a site-specific mutant was characterized. The mutation, uvrDQ251E, resides within helicase motif III, a conserved segment of amino acid homology found in a superfamily of prokaryotic and eukaryotic DNA helicases. The UvrD-Q251E protein failed to complement the mutator and ultraviolet light-sensitive phenotypes of a uvrD deletion strain indicating that the mutant protein is inactive in both mismatch repair and excision repair. Biochemical characterization revealed a significant defect in the ability of the mutant enzyme to initiate unwinding at a nick. The elongation phase of the unwinding reaction was nearly normal. Together, the biochemical and genetic data provide evidence that UvrD-Q251E is dysfunctional because the mutant protein fails to initiate unwinding at the nick(s) used to initiate excision and subsequent repair synthesis. These results provide direct evidence to support the notion that helicase II initiates unwinding from a nick in vivo in mismatch repair and excision repair

    A high-throughput screen to identify inhibitors of SOD1 transcription

    Amyotrophic lateral sclerosis (ALS) is a fatal degenerative motor neuron disease. Approximately 20% of familial ALS cases are caused by mutations in the Cu/Zn superoxide dismutase (SOD1) gene. Rodents expressing mutant SOD1 transgenes develop progressive, fatal motor neuron disease and disease onset and progression is dependent on the level of SOD1. We investigated the possibility that a reduction in SOD1 protein may be of therapeutic benefit in ALS and screened 30,000 compounds for inhibition of SOD1 transcription. The most effective inhibitor identified was N-{4-[4-(4-methylbenzoyl)-1-piperazinyl]phenyl}-2-thiophenecarboxamide (Compound ID 7687685), which in PC12 cells showed an EC50 of 10.6 microM for inhibition of SOD1 expression and an LD50 >30 microM. This compound was subsequently shown to reduce endogenous SOD1 levels in HeLa cells and to exhibit a modest reduction of SOD1 protein levels in mouse spinal cord tissue. These data suggest that the efficacy of compound 7687685 as an inhibitor of SOD1 gene expression is not likely to be clinically useful, although the strategy reported could be applied broadly to screening for small molecule inhibitors of gene expression

    Anaerobic Carbon Monoxide Dehydrogenase Diversity in the Homoacetogenic Hindgut Microbial Communities of Lower Termites and the Wood Roach

    Anaerobic carbon monoxide dehydrogenase (CODH) is a key enzyme in the Wood-Ljungdahl (acetyl-CoA) pathway for acetogenesis performed by homoacetogenic bacteria. Acetate generated by gut bacteria via the acetyl-CoA pathway provides considerable nutrition to wood-feeding dictyopteran insects making CODH important to the obligate mutualism occurring between termites and their hindgut microbiota. To investigate CODH diversity in insect gut communities, we developed the first degenerate primers designed to amplify cooS genes, which encode the catalytic (β) subunit of anaerobic CODH enzyme complexes. These primers target over 68 million combinations of potential forward and reverse cooS primer-binding sequences. We used the primers to identify cooS genes in bacterial isolates from the hindgut of a phylogenetically lower termite and to sample cooS diversity present in a variety of insect hindgut microbial communities including those of three phylogenetically-lower termites, Zootermopsis nevadensis, Reticulitermes hesperus, and Incisitermes minor, a wood-feeding cockroach, Cryptocercus punctulatus, and an omnivorous cockroach, Periplaneta americana. In total, we sequenced and analyzed 151 different cooS genes. These genes encode proteins that group within one of three highly divergent CODH phylogenetic clades. Each insect gut community contained CODH variants from all three of these clades. The patterns of CODH diversity in these communities likely reflect differences in enzyme or physiological function, and suggest that a diversity of microbial species participate in homoacetogenesis in these communities

    The purpose of this study was to analyze bone surfaces cut by Er:YAG laser or by diamond points. Five male rats (Rattus norvegicus albinus) were killed by lethal dose of phenobarbital, and selected bones were cut by Er:YAG laser at 300 mJ and 2 Hz, or by diamond points. The samples were submitted to evaluation through SEM at standard magnifications. By analyzing the surfaces, the authors could observe a pattern for each group, with a smoother surface in the laser cut group than in the diamond cut group. In higher magnifications, the images suggest melting and solidifying in the laser group. The authors conclude that the laser was effective in bone cutting. However, it was responsible for morphological changes that suggest a significant temperature increase.O objetivo deste trabalho foi analisar morfologicamente as superfícies ósseas resultantes da secção por pontas diamantadas ou por laser de érbio:YAG. Cinco ratos (Rattus norvegicus albinus foram sacrificados por dose letal de fenobarbital. Após a execução deste procedimento, os ossos predeterminados foram submetidos à secção por pontas diamantadas ou por laser de érbio:YAG em uma energia de 300 mJ por pulso e taxa de repetição de 2 Hz. As amostras foram submetidas a análise em microscópio eletrônico de varredura, revelando a existência de um padrão para as secções obtidas com cada instrumento, sendo verificada uma superfície mais regular nas amostras seccionadas com o laser de érbio:YAG. Em aumentos da ordem de 3000 vezes, pode-se observar indícios de fusão e seqüente solidificação das superfícies seccionadas por meio do laser de érbio:YAG. Conclui-se que o laser de érbio:YAG foi eficaz na remoção de tecido ósseo, mas que, nos parâmetros utilizados neste estudo, foi responsável por alterações morfológicas sugestivas de significativo aumento de temperatura, não devendo ser indicado, nestas condições, para a execução de secções ósseas
