    Evaluation of intramuscular lateral distribution profile of topically administered acetaminophen in rats

    &#65279;To clarify to what extent topically administered drug molecules horizontally permeate into tissues surrounding the administration site, the intramuscular lateral concentration profile of acetaminophen was investigated in vivo using the microdialysis method in rats. When acetaminophen was intramuscularly administered for 6 hr in a pinpoint manner at a constant rate of 3 &#956;g/min, it was clearly detected in the muscle surrounding the administration site, being 17.5 &#956;g/ml when measured at a 2 mm distance from the administration site. The concentration in the muscle was decreased as the distance increased, and those measured at 5 mm and 40 mm were 0.35 &#956;g/ml and 0.09 &#956;g/ml, respectively. In addition, it was shown that the concentration in the muscle at 40 mm reflected the compoundā€™s concentration in plasma, but not the compoundā€™s horizontal permeation from the administration site. With these observations, the intramuscular distribution profile of acetaminophen was numerically characterized according to Fickā€™s law. As a result, it was revealed that horizontal permeation is the primary process accountable for the increased intramuscular concentration only in the area adjacent to the administration site, and the radius of the adjacent area was calculated to be 5.80 mm for acetaminophen. </p

    Restoration of Corneal Transparency by Mesenchymal Stem Cells

    Summary Transparency of the cornea is indispensable for optimal vision. Ocular trauma is a leading cause of corneal opacity, leading to 25 million cases of blindness annually. Recently, mesenchymal stem cells (MSCs) have gained prominence due to their inflammation-suppressing and tissue repair functions. Here, we investigate the potential of MSCs to restore corneal transparency following ocular injury. Using an in vivo mouse model of ocular injury, we report that MSCs have the capacity to restore corneal transparency by secreting high levels of hepatocyte growth factor (HGF). Interestingly, our data also show that HGF alone can restore corneal transparency, an observation that has translational implications for the development of HGF-based therapy

To compare the effects of post-penetrating keratoplasty (PK) and post-keratoprosthesis (KPro) surgery-related inflammation on the posterior segment of the eye and to assess inhibition of tumor necrosis factor alpha (TNFa) and interleukin-1 beta (IL-1b) on these effects. METHODS. BALB/C (syngeneic) or C57BL/6 (allogeneic) corneas were transplanted onto BALB/ C host beds as part of PK or miniature KPro (m-KPro) implantation. Intraocular pressure (IOP) was measured via an intracameral pressure sensor; tissues were harvested and analyzed 8 weeks after surgery. Expression of TNFa and IL-1b in the retina was analyzed using real-time quantitative (q)PCR. Optic nerve degeneration (axon count, circularity, and area) was assessed quantitatively using ImageJ software. After m-KPro implantation, mice were treated with saline, anti-TNFa, or anti-IL-1b antibody, and axonal loss was assessed after 10 weeks. RESULTS. Mean IOP was within normal limits in the operated and fellow eyes in all groups. The mRNA expression of TNFa and IL-1b was highest in m-KPro groups with either syngeneic or an allogeneic carrier. We observed optic nerve degeneration in both allogeneic PK and mKPro implanted eyes with an allogeneic carrier. However, TNFa blockade significantly reduced axonal loss by 35%. CONCLUSIONS. Allogeneic PK and m-KPro implants with an allogeneic carrier lead to chronic inflammation in the posterior segment of the eye, resulting in optic nerve degeneration. In addition, blockade of TNFa prevents axonal degeneration in this preclinical model of allogeneic m-KPro (alloKPro) implantation

    Immunohistochemical study of vascular endothelial growth factorā€‘C/vascular endothelial growth factor receptorā€‘3 expression in oral tongue squamous cell carcinoma: Correlation with the induction of lymphangiogenesis

    The aim of the present study was to elucidate the associations between the expression of the vascular endothelial growth factor-C (VEGF-C)/VEGF receptor-3 (VEGFR-3) axis and lymphangiogenesis, regional lymph node metastasis and clinicopathological factors in oral tongue squamous cell carcinoma (OTSCC) using immunohistochemistry. The expression of VEGF-C, VEGFR-3 and podoplanin was immunohistochemically evaluated in specimens obtained from 65 patients with OTSCC (T1-2, N0) who had undergone radical surgery alone. The associations between the expression of VEGF-C, VEGFR-3 and podoplanin, and lymphangiogenesis, regional lymph node metastasis and clinocopathological factors were determined by immunohistochemical analysis. VEGF-C, VEGFR-3 and combined VEGF-C/VEGFR-3 expression was significantly higher in cases with regional recurrence compared with those without lymph node involvement (P<0.001). As regards lymphangiogenesis, a significant correlation was observed between podoplanin expression and VEGF-C, VEGFR-3 and combined VEGF-C/VEGFR-3 expression (P<0.001). Therefore, lymphangiogenesis in the peritumoral stroma was associated with lymph node metastasis. However, podoplanin expression did not exhibit a significant correlation with the progression of lymph node metastasis. The results of the present study suggest that the VEGF-C/VEGFR-3 axis may be associated with lymph node metastasis through lymphangiogenesis. Determining the VEGF-C/VEGFR-3 expression status may help predict which patients will develop regional recurrence and provide novel targets for therapies to suppress lymph node metastasis in the treatment of OTSCC

    Response of laboratory plasma influenced by a feedback loop to external force

    This study experimentally examines the sensitivity of laboratory plasma influenced by a feedback loop to applied external force, with the objective of gaining insight into chaos control in plasma. The ionization waves in laboratory plasma are adopted as the medium of a typical nonlinear system. In the case of a system without the influence of feedback (defined as system A), the orbit of the chaotic system changes to a periodic one as the intensity of the applied external force increases. In the case of a system with the influence of feedback (defined as system B), the chaotic orbit changes to a periodic one by the application of external force with lower intensity (approximately two-thirds) compared to system A. This reveals that the feedback loop enhances the response of the system to external forces
