    Studies on the biodistribution of dextrin nanoparticles

    The characterization of biodistribution is a central requirement in the development of biomedical applications based on the use of nanoparticles, in particular for controlled drug delivery. The blood circulation time, organ biodistribution and rate of excretion must be well characterized in the process of product development. In this work, the biodistribution of recently developed self-assembled dextrin nanoparticles is addressed. Functionalization of the dextrin nanoparticles with a DOTA-monoamide-type metal chelator, via click chemistry, is described. The metal chelator-functionalized nanoparticles were labeled with the ᵧ-emitting 153Sm3+ radioisotope and the blood clearance rate and organ biodistribution of the nanoparticles were obtained. The effect of PEG surface coating on the blood clearance rate and organ biodistribution of the nanoparticles was also studied.The authors wish to acknowledge funding through the FCT/POCTI programme (project PTDC/QUI/70063/2006)

    Ga[NO2A-N-(alfa-amino)propionate] chelates: synthesis and evaluation as potential tracers for 68Ga3+ PET

    The availability of commercial 68Ge/68Ga cyclotron-independent 68Ga3+ generators is making Positron Emission Tomography (PET) accessible to most hospitals, which is generating a surge of interest in the design and synthesis of bi-functional chelators for Ga3+. In this work we introduce the NO2A-N-(alfa-amino)propionic acid family of chelators based on the triazacyclononane scaffold. Complexation of the parent NO2A-N-(alfa-amino)propionic acid chelator and of a low molecular weight (model) amide conjugate with Ga3+ was studied by 1H and 71Ga NMR. The Ga3+ chelate of the amide conjugate shows pH-independent N3O3 coordination in the pH range 3-10 involving the carboxylate group of the pendant propionate arm in a 6 member chelate. For the Ga[NO2A-N-(alfa-amino)propionate] chelate, a reversible pH-triggered switch from Ga3+ coordination to the carboxylate group to coordination to the amine group of the propionate arm, was observed upon pH increase/decrease in the pH range 4-6. This phenomenon can conceivably constitute the basis of a physiological pH sensor. Both complexes are stable in the physiological range. The [67Ga][NO2A-N-(alfa-benzoylamido)propionate] chelate was found to be stable in human serum. Biodistribution studies of the 67Ga3+-labeled pyrene butyric acid conjugate NO2A-N-(alfa-pyrenebutanamido)propionic acid revealed that, despite its high lipophilicity and concentration-dependent aggregation properties, the chelate follows mainly renal elimination with very low liver/spleen accumulation and no activity deposition in bones after 24 hours.     Iliac artery false aneurysm twelve years after allograft nephrectomy

    We report a case of a non-infected right external iliac artery false aneurysm. The patient was a 44 year-old woman on chronic peritoneal dialysis had had an allograft nephrectomy 12 years before and who presented with acute abdominal pain. Ultrasound and CT-scan showed a saccular aneurysm arising from the right external iliac artery. A large false aneurysm was identified during surgery, from donor arterial vessel remaining in situ after graft nephrectomy. Resection of the false aneurysm, with ligation of the right external iliac artery and interposition of a femorofemoral graft was successfully performed, with an uneventful post-operative recovery. False aneurysms after renal allograft nephrectomy are very rare. To our knowledge, this is the longest reported period of time between na nephrectomy and clinical evidence of a false aneurysm.info:eu-repo/semantics/publishedVersio

    We report a case of a cerebral venous thrombosis (CVT) in a chronic kidney disease patient with three CVT predisposing conditions. A 53 year-old woman on chronic peritoneal dialysis presented to the emergency department with acute headache and vertigo. The neurological examination and head CT scan performed at the emergency department were normal but, three days later, a lateral gait deviation and a horizontal nystagmus were identified. A brain MRI and MRI-venogram confirmed a left lateral sinus thrombosis. Hormonal replacement therapy (HRT), a positive lupus anticoagulante and a homozygous mutation on the methylenetetrahydrofolate reductase gene, with hyperhomocysteinaemia, were the three well-known prothrombotic conditions identified in this patient. HRT was discontinued, the patient started anticoagulation with warfarin and folic acid supplementation and was discharged, 10 days after admission, complaining of a mild vertigo. After six months of therapy the patient had vertigo improvement and maintained a positive lupus anticoagulant. The head MRI and MRI-venography showed a thrombus reduction.info:eu-repo/semantics/publishedVersio

O conhecimento sobre os principais produtos químicos, as formas de utilização com os devidos cuidados na aplicação e na escolha correta do produto constituem parte das estratégias disponíveis para minimizar os prejuízos causados pelos carrapatos. A situação em termos de disponibilidade de produtos carrapaticidas eficazes para a maioria das populações de carrapatos no Brasil é bastante preocupante, especialmente na região Sudeste. A transferência do conhecimento existente, de forma organizada, pode aglutinar os setores público e privado em torno de uma causa comum: melhorar o controle existente do carrapato no país

    Fungal peritonitis in peritoneal dialysis patients: is previous antibiotic therapy an essential condition?

    The aim of this study was to analyse the clinical and microbiological features of fungal peritonitis, in chronic peritoneal dialysis patients, focusing on non-traditional risk factors for this feared complication. From 2001 to 2004, five episodes of fungal peritonitis were diagnosed in five different patients, accounting for 4.5% of all peritonitis cases seen during this period. Candida spp. were the most frequent isolates. In all cases, peritoneal dialysis catheter removal and switching to haemodialysis were necessary. In these five cases of fungal peritonitis only one was preceded by antibiotic use, within the previous 3 months, the classical risk factor for fungal peritonitis. Identifying predisposing factors usually not taken into account, may lead to an early diagnosis and to a better understanding of fungal peritonitis pathogenesis.info:eu-repo/semantics/publishedVersio
