    Characterization of Sirt2 using conditional RNAi in mice

    Within the past eight years, RNA interference (RNAi) has emerged as a powerful experimental tool for gene function analysis in mice. Reversible control of shRNA mediated RNAi has been achieved by using a tetracycline (tet)-inducible promoter. In the presence of the inductor doxycycline (dox), shRNA mediated gene silencing is initiated, whereas RNAi mechanism is blocked in the absence of dox. To achieve spatially and temporally regulated RNAi, the tet inducible system was combined with a Cre/loxP based strategy for tissue specific activation of shRNA constructs. To this end, a loxP-flanked "promoter inhibitory element" (PIE) was placed between the proximal (PSE) and distal sequence element (DSE) of a dox inducible promoter such that promoter function is completely blocked. Re-activation can be achieved through Cre mediated excision of PIE. To allow for gene silencing in a selected tissue, Cre expression can be regulated by a tissue-specific promoter. In mouse ES cells, the system mediated tight regulation of shRNA expression upon Cre mediated activation and dox administration, reaching knockdown efficiencies of >80%. Unexpectedly, the system showed a limited activity in transgenic mice when applied for conditional silencing of two different targets, LacZ and Sirt2. Sirt2 is a member of the sirtuin family which has considerably gained attention in vitro for its possible role in many physiological processes, including adipogenesis and neurodegenerative diseases. To investigate the function of Sirt2 in vivo, the unmodified dox-responsive and tet-inducible promoter was further used for conditional RNAi in transgenic mice. Inducible shRNA expression resulted in efficient silencing of Sirt2 (>90%) in all tissues which have been analyzed. Suppression of Sirt2 during embryogenesis resulted in offspring consisting of equal ratios of wild type and transgenic pups, indicating that Sirt2 is not indispensable for development. In adult animals, glucose metabolism, insulin sensitivity and energy balance appeared to be unaffected by Sirt2 deficiency. Likewise, expression of PPARγ, a downstream target of Sirt2, was not found to be altered upon Sirt2 inhibition. Finally, Sirt2 silencing was induced in an experimental model of Parkinson disease (PD). Data from Rotarod performances to study motor behaviour did not provide any evidence for a role of Sirt2 in PD pathogenesis as suggested by previous in vitro studies. Taken together, conditional Sirt2 silencing in vivo does not support speculation concerning a central role of Sirt2 in physiological processes, embryogenesis and in a mouse model of Parkinson disease

    Ethnic differences in adverse iron status in early pregnancy: a cross-sectional population-based study

    We studied ethnic differences in terms of iron status during pregnancy between Dutch women and other ethnicities and explore to what extent these differences can be explained by environmental factors. This cross-sectional population-based study (2002–2006) was embedded in the Generation R study and included a total of 4737 pregnant women from seven ethnic groups (Dutch, Turkish, Moroccan, Cape Verdean, Surinamese-Hindustani, Surinamese-Creole and Antillean). Ethnicity was defined according to the Dutch classification of ethnic background. Ferritin, iron and transferrin were measured in early pregnancy. The overall prevalence of iron deficiency was 7 %, ranging from 4 % in both Dutch and Surinamese-Creoles, to 18 % in Turkish, Moroccan and Surinamese-Hindustani women. Iron overload was most prevalent in Surinamese-Creole (11 %) and Dutch (9 %) women. Socioeconomic factors accounted for 5–36 % of the differences. Income was the strongest socioeconomic factor in the Cape Verdean and Surinamese-Hindustani groups and parity for the Turkish and Moroccan groups. Lifestyle determinants accounted for 8–14 % of the differences. In all groups, the strongest lifestyle factor was folic acid use, being associated with higher iron status. In conclusion, in our population, both iron deficiency and iron overload were common in early pregnancy. Our data suggest that ethnic differences in terms of socioeconomic and lifestyle factors only partly drive the large ethnic differences in iron status. Our data support the development of more specific prevention programmes based on further exploration of socioeconomic inequities, modifiable risk and genetic factors in specific ethnic subgroups, as well as the need for individual screening of iron status before supplementation

    Cosmology with a long range repulsive force

    We consider a class of cosmological models in which the universe is filled with a (non-electric) charge density that repels itself by means of a force carried by a vector boson with a tiny mass. When the vector's mass depends upon other fields, the repulsive interaction gives rise to an electromagnetic barrier which prevents these fields from driving the mass to zero. This can modify the cosmology dramatically. We present a very simple realization of this idea in which the vector's mass arises from a scalar field.     Studies of dust devils, and their impact on society, are reviewed. Dust devils have been noted since antiquity, and have been documented in many countries, as well as on the planet Mars. As time-variable vortex entities, they have become a cultural motif. Three major stimuli of dust devil research are identified, nuclear testing, terrestrial climate studies, and perhaps most significantly, Mars research. Dust devils present an occasional safety hazard to light structures and have caused several deaths

    Leftward Lateralization of Auditory Cortex Underlies Holistic Sound Perception in Williams Syndrome

    BACKGROUND: Individuals with the rare genetic disorder Williams-Beuren syndrome (WS) are known for their characteristic auditory phenotype including strong affinity to music and sounds. In this work we attempted to pinpoint a neural substrate for the characteristic musicality in WS individuals by studying the structure-function relationship of their auditory cortex. Since WS subjects had only minor musical training due to psychomotor constraints we hypothesized that any changes compared to the control group would reflect the contribution of genetic factors to auditory processing and musicality. METHODOLOGY/PRINCIPAL FINDINGS: Using psychoacoustics, magnetoencephalography and magnetic resonance imaging, we show that WS individuals exhibit extreme and almost exclusive holistic sound perception, which stands in marked contrast to the even distribution of this trait in the general population. Functionally, this was reflected by increased amplitudes of left auditory evoked fields. On the structural level, volume of the left auditory cortex was 2.2-fold increased in WS subjects as compared to control subjects. Equivalent volumes of the auditory cortex have been previously reported for professional musicians. CONCLUSIONS/SIGNIFICANCE: There has been an ongoing debate in the neuroscience community as to whether increased gray matter of the auditory cortex in musicians is attributable to the amount of training or innate disposition. In this study musical education of WS subjects was negligible and control subjects were carefully matched for this parameter. Therefore our results not only unravel the neural substrate for this particular auditory phenotype, but in addition propose WS as a unique genetic model for training-independent auditory system properties

    Dust Devil Sediment Transport: From Lab to Field to Global Impact

    The impact of dust aerosols on the climate and environment of Earth and Mars is complex and forms a major area of research. A difficulty arises in estimating the contribution of small-scale dust devils to the total dust aerosol. This difficulty is due to uncertainties in the amount of dust lifted by individual dust devils, the frequency of dust devil occurrence, and the lack of statistical generality of individual experiments and observations. In this paper, we review results of observational, laboratory, and modeling studies and provide an overview of dust devil dust transport on various spatio-temporal scales as obtained with the different research approaches. Methods used for the investigation of dust devils on Earth and Mars vary. For example, while the use of imagery for the investigation of dust devil occurrence frequency is common practice for Mars, this is less so the case for Earth. Modeling approaches for Earth and Mars are similar in that they are based on the same underlying theory, but they are applied in different ways. Insights into the benefits and limitations of each approach suggest potential future research focuses, which can further reduce the uncertainty associated with dust devil dust entrainment. The potential impacts of dust devils on the climates of Earth and Mars are discussed on the basis of the presented research results

We hypothesize that this is mediated by a disturbed vascular development of the placenta, umbilical cord and fetus.
