    Improving a pavement-watering method on the basis of pavement surface temperature measurements

    Pavement-watering has been studied since the 1990's and is currently considered a promising tool for urban heat island reduction and climate change adaptation. However, possible future water resource availability problems require that water consumption be optimized. Although pavement heat flux can be studied to improve pavement-watering methods (frequency and water consumption), these measurements are costly and require invasive construction work to install appropriate sensors in a dense urban environment. Therefore, we analyzed infrared camera measurements of pavement surface temperatures in search of alternative information relevant to this goal. Firstly, surface temperature reductions of up to 4{\textdegree}C during shading and 13{\textdegree}C during insolation were found. Secondly, the infrared camera successfully detected temperature spikes indicative of surface drying and can therefore be used to optimize the watering frequency. Measurements made every 5 min or less are recommended to minimize relevant data loss. Finally, if the water retaining capacity of the studied pavement is known, optimization of total water consumption is possible on the sole basis of surface temperature measurements.Comment: Published in Urban Climat

    Collision of Two Rotating Hayward Black Holes

    We investigate the spin interaction and the gravitational radiation thermally allowed in a head-on collision of two rotating Hayward black holes. The Hayward black hole is a regular black hole in a modified Einstein equation, and hence it can be an appropriate model to describe the extent to which the regularity effect in the near-horizon region affects the interaction and the radiation. If one black hole is assumed to be considerably smaller than the other, the potential of the spin interaction can be analytically obtained and is dependent on the alignment of angular momenta of the black holes. For the collision of massive black holes, the gravitational radiation is numerically obtained as the upper bound by using the laws of thermodynamics. The effect of the Hayward black hole tends to increase the radiation energy, but we can limit the effect by comparing the radiation energy with the gravitational waves GW150914 and GW151226.Comment: 25 pages, 43 figures, published version in EPJ

    Les populations mondiales d’éléphants d’Afrique et d’Asie sont en déclin. Cette disparition progressive des éléphants est multifactorielle, aussi bien liée à la destruction de l’habitat naturel qu’au braconnage. Des actions sont menées dans le milieu naturel afin de préserver les dernières populations d’éléphants. La reproduction des éléphants en captivité semble être également une alternative intéressante pour faire perdurer l’espèce. Cependant, l’éléphant possède des caractéristiques uniques rendant la maîtrise de la reproduction difficile en captivité : il s’agit du plus gros mammifère terrestre, il possède la gestation la plus longue parmi les mammifères terrestres et son système social est complexe et très hiérarchisé. Bien que les naissances d’éléphanteaux en parcs zoologiques soient rares, elles ont contribué à améliorer les connaissances concernant la reproduction des éléphants depuis les années 1950. L’auteur décrit dans cette ouvrage la situation actuelle des éléphants dans le monde, les cycles biologiques reproducteurs des éléphants mâles et femelles et enfin, il énumère les techniques de maîtrise de reproduction en captivité, de l’accouplement au sevrage de l’éléphanteau

    RNA Control of HIV-1 Particle Size Polydispersity

    HIV-1, an enveloped RNA virus, produces viral particles that are known to be much more heterogeneous in size than is typical of non-enveloped viruses. We present here a novel strategy to study HIV-1 Viral Like Particles (VLP) assembly by measuring the size distribution of these purified VLPs and subsequent viral cores thanks to Atomic Force Microscopy imaging and statistical analysis. This strategy allowed us to identify whether the presence of viral RNA acts as a modulator for VLPs and cores size heterogeneity in a large population of particles. These results are analyzed in the light of a recently proposed statistical physics model for the self-assembly process. In particular, our results reveal that the modulation of size distribution by the presence of viral RNA is qualitatively reproduced, suggesting therefore an entropic origin for the modulation of RNA uptake by the nascent VLP

    Single base mismatches in the mRNA target site allow specific seed region-mediated off-target binding of siRNA targeting human coagulation factor 7

    We have analyzed the off-target activity of two siRNAs (F7-1, F7-2) that knock-down human blood coagulation factor 7 mRNA. F7-1 modulates a significant number of non-target transcripts while F7-2 shows high selectivity for the target transcript under various experimental conditions. The 3′-UTRs of all F7-1 off-target genes show statistically significant enrichment of the reverse complement of the F7-1 siRNA seed region located in the guide strand. Seed region enrichment was confirmed in off-target transcripts modulated by siRNA targeting the glucocorticoid receptor. To investigate how these sites contribute to off-target recognition of F7-1, we employed CXCL5 transcript as model system because it contains five F7-1 seed sequence motifs with single base mismatches. We show by transient transfection of reporter gene constructs into HEK293 cells that three out of five sites located in the 3′-UTR region are required for F7-1 off-target activity. For further mechanistic dissection, the sequences of these sites were synthesized and inserted either individually or joined in dimeric or trimeric constructs. Only the fusion constructs were silenced by F7-1 while the individual sites had no off-target activity. Based on F7-1 as a model, a single mismatch between the siRNA seed region and mRNA target sites is tolerated for target recognition and the CXCL5 data suggest a requirement for binding to multiple target sites in off-target transcripts

    Beyond cAMP: The Regulation of Akt and GSK3 by Dopamine Receptors

    Brain dopamine receptors have been preferred targets for numerous pharmacological compounds developed for the treatment of various neuropsychiatric disorders. Recent discovery that D2 dopamine receptors, in addition to cAMP pathways, can engage also in Akt/GSK3 signaling cascade provided a new framework to understand intracellular signaling mechanisms involved in dopamine-related behaviors and pathologies. Here we review a recent progress in understanding the role of Akt, GSK3, and related signaling molecules in dopamine receptor signaling and functions. Particularly, we focus on the molecular mechanisms involved, interacting partners, role of these signaling events in the action of antipsychotics, psychostimulants, and antidepressants as well as involvement in pathophysiology of schizophrenia, bipolar disorder, and Parkinson’s disease. Further understanding of the role of Akt/GSK3 signaling in dopamine receptor functions could provide novel targets for pharmacological interventions in dopamine-related disorders

    The defensive response of the honeybee Apis mellifera

    Honeybees (Apis mellifera) are insects living in colonies with a complex social organization. Their nest contains food stores in the form of honey and pollen, as well as the brood, the queen and the bees themselves. These resources have to be defended against a wide range of predators and parasites, a task that is performed by specialized workers, called guard bees. Guards tune their response to both the nature of the threat and the environmental conditions, in order to achieve an efficient trade-off between defence and loss of foraging workforce. By releasing alarm pheromones, they are able to recruit other bees to help them handle large predators. These chemicals trigger both rapid and longer-term changes in the behaviour of nearby bees, thus priming them for defence. Here, we review our current understanding on how this sequence of events is performed and regulated depending on a variety of factors that are both extrinsic and intrinsic to the colony. We present our current knowledge on the neural bases of honeybee aggression and highlight research avenues for future studies in this area. We present a brief overview of the techniques used to study honeybee aggression, and discuss how these could be used to gain further insights into the mechanisms of this behaviour

    HIV-1 tropism determination using a phenotypic Env recombinant viral assay highlights overestimation of CXCR4-usage by genotypic prediction algorithms for CRRF01_AE and CRF02_AG

    Background: Human Immunodeficiency virus type-1 (HIV) entry into target cells involves binding of the viral envelope (Env) to CD4 and a coreceptor, mainly CCR5 or CXCR4. The only currently licensed HIV entry inhibitor, maraviroc, targets CCR5, and the presence of CXCX4-using strains must be excluded prior to treatment. Co-receptor usage can be assessed by phenotypic assays or through genotypic prediction. Here we compared the performance of a phenotypic Env-Recombinant Viral Assay (RVA) to the two most widely used genotypic prediction algorithms, Geno2Pheno([coreceptor]) and webPSSM. Methods: Co-receptor tropism of samples from 73 subtype B and 219 non-B infections was measured phenotypically using a luciferase-tagged, NL4-3-based, RVA targeting Env. In parallel, tropism was inferred genotypically from the corresponding V3-loop sequences using Geno2Pheno([coreceptor]) (5-20% FPR) and webPSSM-R5X4. For discordant samples, phenotypic outcome was retested using co-receptor antagonists or the validated Trofile (R) Enhanced-Sensitivity-Tropism-Assay. Results: The lower detection limit of the RVA was 2.5% and 5% for X4 and R5 minority variants respectively. A phenotype/genotype result was obtained for 210 samples. Overall, concordance of phenotypic results with Geno2Pheno([coreceptor]) was 85.2% and concordance with webPSSM was 79.5%. For subtype B, concordance with Geno2pheno([coreceptor]) was 94.4% and concordance with webPSSM was 79.6%. High concordance of genotypic tools with phenotypic outcome was seen for subtype C (90% for both tools). Main discordances involved CRF01_AE and CRF02_AG for both algorithms (CRF01_AE: 35.9% discordances with Geno2Pheno([coreceptor]) and 28.2% with webPSSM; CRF02_AG: 20.7% for both algorithms). Genotypic prediction overestimated CXCR4-usage for both CRFs. For webPSSM, 40% discordance was observed for subtype A. Conclusions: Phenotypic assays remain the most accurate for most non-B subtypes and new subtype-specific rules should be developed for non-B subtypes, as research studies more and more draw conclusions from genotypically-inferred tropism, and to avoid unnecessarily precluding patients with limited treatment options from receiving maraviroc or other entry inhibitors

    Measurement of the Cooling Efficiency of Pavement-watering as an Urban Heat Island Mitigation Technique

    The Paris region (Île-de-France) was amongst the hardest hit by the August 2003 heat wave, due in part to subsequent amplification of its urban heat island. This has created high heat-wave awareness in climate change adaptation studies for the city of Paris. Over the summer of 2013, pavement watering was studied experimentally in two locations as a climate change adaptation method. Pavement watering was found to lower pavement surface temperatures by several degrees for several hours after watering, while also strongly reducing its cooling rate a few hours before and after sunset. Heat flux and storage at a depth of 5 cm in the pavement were also found to have been significantly reduced, especially during direct sunlight exposure, but also when the pavement was in the shade. Uninterrupted watering appears necessary during direct sunlight exposure of the pavement to maximize efficiency
