    Brown Adipose Tissue Thermogenic Capacity Is Regulated by Elovl6.

    Although many transcriptional pathways regulating BAT have been identified, the role of lipid biosynthetic enzymes in thermogenesis has been less investigated. Whereas cold exposure causes changes in the fatty acid composition of BAT, the functional consequences of this remains relatively unexplored. In this study, we demonstrate that the enzyme Elongation of Very Long Chain fatty acids 6 (Elovl6) is necessary for the thermogenic action of BAT. Elovl6 is responsible for converting C16 non-essential fatty acids into C18 species. Loss of Elovl6 does not modulate traditional BAT markers; instead, it causes reduced expression of mitochondrial electron transport chain components and lower BAT thermogenic capacity. The reduction in BAT activity appears to be counteracted by increased beiging of scWAT. When beige fat is disabled by thermoneutrality or aging, Elovl6 KO mice gain weight and have increased scWAT mass and impaired carbohydrate metabolism.     Third post-Newtonian dynamics of compact binaries: Noetherian conserved quantities and equivalence between the harmonic-coordinate and ADM-Hamiltonian formalisms

    A Lagrangian from which derive the third post-Newtonian (3PN) equations of motion of compact binaries (neglecting the radiation reaction damping) is obtained. The 3PN equations of motion were computed previously by Blanchet and Faye in harmonic coordinates. The Lagrangian depends on the harmonic-coordinate positions, velocities and accelerations of the two bodies. At the 3PN order, the appearance of one undetermined physical parameter \lambda reflects an incompleteness of the point-mass regularization used when deriving the equations of motion. In addition the Lagrangian involves two unphysical (gauge-dependent) constants r'_1 and r'_2 parametrizing some logarithmic terms. The expressions of the ten Noetherian conserved quantities, associated with the invariance of the Lagrangian under the Poincar\'e group, are computed. By performing an infinitesimal ``contact'' transformation of the motion, we prove that the 3PN harmonic-coordinate Lagrangian is physically equivalent to the 3PN Arnowitt-Deser-Misner Hamiltonian obtained recently by Damour, Jaranowski and Sch\"afer.Comment: 30 pages, to appear in Classical and Quantum Gravit

    Days Spent at Home and Mortality After Critical Illness: A Cluster Analysis Using Nationwide Data

    BACKGROUND: Beyond the question of short-term survival, days spent at home could be considered a patient-centered outcome in critical care trials. RESEARCH QUESTION: What are the days spent at home and healthcare trajectories during the year after surviving critical illness? STUDY DESIGN AND METHODS: Data were extracted on adult survivors spending at least two nights in a French intensive care unit (ICU) during 2018 who were treated with invasive mechanical ventilation and/or vasopressors or inotropes. Trauma, burn, organ transplant, stroke and neurosurgical patients were excluded. Stays at home, death, hospitalizations were reported before and after ICU stay, using state sequence analysis. An unsupervised clustering method was performed to identify cohorts based on post-ICU trajectories. RESULTS: Of 77,132 ICU survivors, 89% returned home. In the year post-discharge, these patients spent a median 330 (IQR 283-349) days at home. At one year, 77% of patients were still at home and 17% had died. Fifty-one percent had been re-hospitalized and 10% required a further ICU admission. Forty-eight percent used rehabilitation facilities and 5.7% hospital at home. Three clusters of patients with distinct post-ICU trajectories were identified. Patients in cluster 1 (68% of total) survived and spent most of the year at home (338 (323-354) days). Patients in cluster 2 (18%) had more complex trajectories but most could return home (91%), spending 242 (174-277) days at home. Patients in cluster 3 (14%) died with only 37% returning home for 45 (15-90) days. INTERPRETATION: Many patients had complex healthcare trajectories after surviving critical illness. Wide variations in the ability to return home after ICU discharge was observed between clusters, which represents an important patient-centered outcome

    Developmental characterization of the microRNA-specific C. elegans Argonautes alg-1 and alg-2.

    The genes alg-1 and alg-2 (referred to as "alg-1/2") encode the Argonaute proteins affiliated to the microRNA (miRNA) pathway in C. elegans. Bound to miRNAs they form the effector complex that effects post-transcriptional gene silencing. In order to define biological features important to understand the mode of action of these Argonautes, we characterize aspects of these genes during development. We establish that alg-1/2 display an overlapping spatio-temporal expression profile and shared association to a miRNAs set, but with gene-specific predominant expression in various cells and increased relative association to defined miRNAs. Congruent with their spatio-temporal coincidence and regardless of alg-1/2 drastic post-embryonic differences, only loss of both genes leads to embryonic lethality. Embryos without zygotic alg-1/2 predominantly arrest during the morphogenetic process of elongation with defects in the epidermal-muscle attachment structures. Altogether our results highlight similarities and specificities of the alg-1/2 likely to be explained at different cellular and molecular levels

    POLG2 deficiency causes adult-onset syndromic sensory neuropathy, ataxia and parkinsonism

    Objective: Mitochondrial dysfunction plays a key role in the pathophysiology of neurodegenerative disorders such as ataxia and Parkinson's disease. We describe an extended Belgian pedigree where seven individuals presented with adult-onset cerebellar ataxia, axonal peripheral ataxic neuropathy, and tremor, in variable combination with parkinsonism, seizures, cognitive decline, and ophthalmoplegia. We sought to identify the underlying molecular etiology and characterize the mitochondrial pathophysiology of this neurological syndrome. Methods: Clinical, neurophysiological, and neuroradiological evaluations were conducted. Patient muscle and cultured fibroblasts underwent extensive analyses to assess mitochondrial function. Genetic studies including genome-wide sequencing were conducted. Results: Hallmarks of mitochondrial dysfunction were present in patients' tissues including ultrastructural anomalies of mitochondria, mosaic cytochrome c oxidase deficiency, and multiple mtDNA deletions. We identified a splice acceptor variant in POLG2, c.970-1G>C, segregating with disease in this family and associated with a concomitant decrease in levels of POLG2 protein in patient cells. Interpretation: This work extends the clinical spectrum of POLG2 deficiency to include an overwhelming, adult-onset neurological syndrome that includes cerebellar syndrome, peripheral neuropathy, tremor, and parkinsonism. We therefore suggest to include POLG2 sequencing in the evaluation of ataxia and sensory neuropathy in adults, especially when it is accompanied by tremor or parkinsonism with white matter disease. The demonstration that deletions of mtDNA resulting from autosomal-dominant POLG2 variant lead to a monogenic neurodegenerative multicomponent syndrome provides further evidence for a major role of mitochondrial dysfunction in the pathomechanism of nonsyndromic forms of the component neurodegenerative disorders

    Spinal contribution to neuromuscular recovery differs between elbow-flexor and knee-extensor muscles after a maximal sustained fatiguing task

    Data from studies of elbow-flexor (EF) or knee-extensor (KE) muscles suggest that a fatigue-related decrease in motoneuron excitability only occurs in EF. It is unknown how motoneuron excitability changes after sustained fatiguing maximal voluntary isometric contractions (MVICs) in EF and KE in the same participants. In two sessions, eight healthy men performed a 2-min MVIC of EF or KE to induce fatigue with brief MVICs before and six times after the 2-min MVIC. Electromyographic responses elicited by corticospinal tract stimulation at the transmastoid [cervicomedullary motor-evoked potential (CMEP)] or thoracic [thoracic motor-evoked potential (TMEP)] level were recorded from EF and KE, respectively. To account for muscle excitability, CMEPs and TMEPs were normalized to maximal M-wave (Mmax) elicited by peripheral nerve stimulation during each brief MVIC. Immediately after the 2-min MVIC, biceps brachii and brachioradialis CMEP/Mmax were 88% (SD 11%) (P = 0.026) and 87% (SD 12%) (P = 0.029) of pre-MVIC values, respectively, and remained lower than PRE after 5 s of recovery [91% (SD 8%), P = 0.036 and 87% (SD 13%), P = 0.046, respectively]. No subsequent time points differed from PRE (all P ³ 0.253). TMEP/Mmax for rectus femoris and vastus lateralis were not different from PRE at any time during the recovery period (all P > 0.050). A different recovery pattern in motoneuron excitability occurred in EF as it recovered by 60 s whereas KE motoneurons were unaffected by the fatiguing task. The present findings may contribute to better understand muscle-specific neurophysiological differences in spinal excitability

    Water Condensation Zones around Main Sequence Stars

    Understanding the set of conditions that allow rocky planets to have liquid water on their surface -- in the form of lakes, seas or oceans -- is a major scientific step to determine the fraction of planets potentially suitable for the emergence and development of life as we know it on Earth. This effort is also necessary to define and refine the so-called "Habitable Zone" (HZ) in order to guide the search for exoplanets likely to harbor remotely detectable life forms. Until now, most numerical climate studies on this topic have focused on the conditions necessary to maintain oceans, but not to form them in the first place. Here we use the three-dimensional Generic Planetary Climate Model (PCM), historically known as the LMD Generic Global Climate Model (GCM), to simulate water-dominated planetary atmospheres around different types of Main-Sequence stars. The simulations are designed to reproduce the conditions of early ocean formation on rocky planets due to the condensation of the primordial water reservoir at the end of the magma ocean phase. We show that the incoming stellar radiation (ISR) required to form oceans by condensation is always drastically lower than that required to vaporize oceans. We introduce a Water Condensation Limit, which lies at significantly lower ISR than the inner edge of the HZ calculated with three-dimensional numerical climate simulations. This difference is due to a behavior change of water clouds, from low-altitude dayside convective clouds to high-altitude nightside stratospheric clouds. Finally, we calculated transit spectra, emission spectra and thermal phase curves of TRAPPIST-1b, c and d with H2O-rich atmospheres, and compared them to CO2 atmospheres and bare rock simulations. We show using these observables that JWST has the capability to probe steam atmospheres on low-mass planets, and could possibly test the existence of nightside water clouds.Comment: Accepted for publication in Astronomy & Astrophysic

    A Bright Submillimeter Source in the Bullet Cluster (1E0657--56) Field Detected with BLAST

    We present the 250, 350, and 500 micron detection of bright submillimeter emission in the direction of the Bullet Cluster measured by the Balloon-borne Large Aperture Submillimeter Telescope (BLAST). The 500 micron centroid is coincident with an AzTEC 1.1 mm point-source detection at a position close to the peak lensing magnification produced by the cluster. However, the 250 micron and 350 micron centroids are elongated and shifted toward the south with a differential shift between bands that cannot be explained by pointing uncertainties. We therefore conclude that the BLAST detection is likely contaminated by emission from foreground galaxies associated with the Bullet Cluster. The submillimeter redshift estimate based on 250-1100 micron photometry at the position of the AzTEC source is z_phot = 2.9 (+0.6 -0.3), consistent with the infrared color redshift estimation of the most likely IRAC counterpart. These flux densities indicate an apparent far-infrared luminosity of L_FIR = 2E13 Lsun. When the amplification due to the gravitational lensing of the cluster is removed, the intrinsic far-infrared luminosity of the source is found to be L_FIR <= 10^12 Lsun, consistent with typical luminous infrared galaxies.Comment: Accepted for publication in the Astrophysical Journal.     All Optical Implementation of Multi-Spin Entanglement in a Semiconductor Quantum Well

    We use ultrafast optical pulses and coherent techniques to create spin entangled states of non-interacting electrons bound to donors (at least three) and at least two Mn2+ ions in a CdTe quantum well. Our method, relying on the exchange interaction between localized excitons and paramagnetic impurities, can in principle be applied to entangle a large number of spins.Comment: 17 pages, 3 figure
