    The Effect Nanocapsule of Turmeric Extracts in Rations on Nutrient Digestibility of Broiler Chickens

    Abstract. The use of turmeric is restricted by its low solubility in water, therefore it has low bioavailability. This obstacle can be solved by the development of nanoparticle technology to improve drug delivery profile. This study aimed to develop nanoparticle formulation using turmeric extract and industrial chitosan as the matrix and sodium-tripolyphosphate as cross linker, to study its ability to improve feed digestibility. Method used in the formulation of nanoparticle in this study was by ionic gelation followed by oven drying at 50°C. Method used to evaluate the digestibility  was total collection. One hundred and twenty broiler chickens with an average body weight of 900 g, were randomly divided into 20 treatments (one treatment was fasted and 19 were treated with the ration plus feed additive), and six replicates were performed on each test. Chickens were fasted one day before and after they were treated with rations. Nutrient levels and the amount of feed consumed and excreta released were weighed to calculate the digestibility of the ration. It was found that the basal ration had dry matter digestibility of 70.48% significantly lower compared to the basal ration plus nanocapsule turmeric extract: NP level at 0.4% for in EE and EA were 73.11 and 75.90%. The results of this study concluded that formulation of nanocapsule using turmeric extract and industrial chitosan as the matrix and sodium tripolyphosphate as cross linker was potential to increase nutrient digestibility, therefore, it can be an alternative for feed additive in broiler chicken diet. Key words: Digestibility, nanocapsule, turmeric extract, broiler chicken Abstrak. Penggunaan kunyit/kurkumin terbatas karena kelarutannya yang rendah dalam air sehingga bioavailabilitasnya juga rendah. Masalah ini dapat diatasi dengan teknologi nano yang dikembangkan sebagai alternatif penghantaran obat bagi bahan kimia yang mempunyai bioavailabilitas rendah. Penelitian ini bertujuan untuk mengembangkan formulasi nanokapsul menggunakan ekstrak kunyit dan kitosan industri sebagai matrik atau polimer serta sodium tripolifosfat sebagai cross linker, untuk dipelajari kemampuannya dalam meningkatkan kecernaan ransum. Metode yang digunakan dalam formulasi nanokapsul adalah gelasi ionik dilanjutkan pengeringan dengan oven pada suhu 50°C. Sedangkan metode untuk mengevaluasi kecernaan menggunakan total koleksi. Seratus dua puluh ekor ayam broiler dengan bobot badan rata-rata 900 g, dibagi secara acak ke dalam 20 perlakuan (satu perlakuan dipuasakan dan 19 diberi ransum perlakuan dengan penambahan feed additive), masing-masing enam ulangan dan satu ekor untuk tiap ulangan. Ayam dipuasakan sehari pada sebelum dan sesudah ayam diperlakukan dengan ransum, kadar nutrien dan jumlah pakan yang dikonsumsi serta ekskreta yang dikeluarkan ditimbang beratnya untuk menghitung kecernaan ransum. Telah ditemukan bahwa kecernaan bahan kering ransum basal (70,48%) nyata lebih kecil dibanding ransum basal yang ditambah nanokapsul ekstrak kunyit: pada  NP level 0,4% sebesar 73,11% pada EE dan 75,90% pada EA. Dapat disimpulkan bahwa formulasi nanokapsul menggunakan ekstrak kunyit dan kitosan industri sebagai matrik dan sodiumtripolifosfat sebagai cross-linker berpotensi sebagai feed additive alternatif untuk meningkatkan kecernaan nutrien pada ransum ayam broiler. Kata kunci: Kecernaan, nanokapsul, ekstrak kunyit, ayam broile

    Metabolizable Energy of Ration Added with Nanocapsule of Turmeric Extract on Broiler Chicken

    This study was aimed to develop nanoparticle formulation using turmeric extract and industrialchitosan as the polymer and sodium-tripolyphosphate as cross linker, to evaluate its ability to improvetrue metabolizable energy on broiler chicken. Forty eight of broiler chickens with an average bodyweight of 900 g, were randomly divided into 8 treatments (one treatment was fasted and 7 were fedration plus feed additive), containing six birds per treatment. The formulation of nanoparticle in thisstudy used ionic gelation method. Method used to evaluate the metabolizable energy was totalcollection. Chickens were fasted one day before and after they were treated with rations. Nitrogen, grossenergy and total weight of feed and excreta or ileal digesta released were analyzed to calculate the truemetabolizable energy of the ration. It was found that the basal ration added with nanocapsule 0.1% hadtrue metabolizable energy (3370.66 kcal/kg) significantly higher (P<0.05) than basal ration (3214.85kcal/kg). The result of this study concluded that formulation of nanocapsule using turmeric extract andindustrial chitosan as the polymer and sodium-tripolyphosphate as cross linker was potential to increasetrue metabolizable energy so it can be an alternative for broiler chicken feed additive

    Requirements and operational guidelines for secure and sustainable digital phenotyping:Design and development study

    Background: Digital phenotyping, the measurement of human behavioral phenotypes using personal devices, is rapidly gaining popularity. Novel initiatives, ranging from software prototypes to user-ready research platforms, are innovating the field of biomedical research and health care apps. One example is the BEHAPP project, which offers a fully managed digital phenotyping platform as a service. The innovative potential of digital phenotyping strategies resides among others in their capacity to objectively capture measurable and quantitative components of human behavior, such as diurnal rhythm, movement patterns, and communication, in a real-world setting. The rapid development of this field underscores the importance of reliability and safety of the platforms on which these novel tools are operated. Large-scale studies and regulated research spaces (eg, the pharmaceutical industry) have strict requirements for the software-based solutions they use. Security and sustainability are key to ensuring continuity and trust. However, the majority of behavioral monitoring initiatives have not originated primarily in these regulated research spaces, which may be why these components have been somewhat overlooked, impeding the further development and implementation of such platforms in a secure and sustainable way.Objective: This study aims to provide a primer on the requirements and operational guidelines for the development and operation of a secure behavioral monitoring platform.Methods: We draw from disciplines such as privacy law, information, and computer science to identify a set of requirements and operational guidelines focused on security and sustainability. Taken together, the requirements and guidelines form the foundation of the design and implementation of the BEHAPP behavioral monitoring platform.Results: We present the base BEHAPP data collection and analysis flow and explain how the various concepts from security and sustainability are addressed in the design.Conclusions: Digital phenotyping initiatives are steadily maturing. This study helps the field and surrounding stakeholders to reflect upon and progress toward secure and sustainable operation of digital phenotyping–driven research

    Validation for the Quantification of Andrographolide Isolated From Andrographis Paniculata Nees Plant Using HPLC

    The aim of study was to develop quantitative analysis of isolated andrographolide from Andrographis paniculata and different solvent for prelimenary studies to preperation Self Nano Emulsifying Drug Delivery System (SNEDDS) using HPLC. The separation was acquired on Sunfire C18 column with an isocratic mixture of methanol and water at a ratio of 6:4, v/v as a mobile phase. The method to determine the content of isolated andrographolide showed an adequate precision, with a RSD smaller than 1%. The accuracy was analyzed by adding the standard andrographolide, and good recovery values were obtained for all concentrations used. The HPLC method developed in this study showed specificity and selectivity with linearity in the working range and good precision and accuracy, making it very suitable for the quantification of isolated andrographolide. Compared to the standard, the purity of the isolated andrographolide was 95.74 ± 0.29 %. Prelimenary study to determined the highest solubility of isolated andrographolide in oil, surfactant and co-surfactant phases for preperation of SNEDDS were obtained 1.226 ± 0.009 of Capryol-90, 2.965 ± 0.014 of tween 20, and 6.074 ± 0.101 mg mL-1 of PEG 400, respectively.     Design and development of a low temperature, inductance based high frequency ac susceptometer

    We report on the development of an induction based low temperature high frequency ac susceptometer capable of measuring at frequencies up to 3.5 MHz and at temperatures between 2 K and 300 K. Careful balancing of the detection coils and calibration have allowed a sample magnetic moment resolution of 5×1010Am25\times10^{-10} Am^2 at 1 MHz. We will discuss the design and characterization of the susceptometer, and explain the calibration process. We also include some example measurements on the spin ice material CdEr2_2S4_4 and iron oxide based nanoparticles to illustrate functionality

    Long Range Anticorrelations and Non-Gaussian Behavior of a Leaky Faucet

    We find that intervals between successive drops from a leaky faucet display scale-invariant, long-range anticorrelations characterized by the same exponents of heart beat-to-beat intervals of healthy subjects. This behavior is also confirmed by numerical simulations on lattice and it is faucet-width- and flow-rate-independent. The histogram for the drop intervals is also well described by a L\'evy distribution with the same index for both histograms of healthy and diseased subjects. This additional result corroborates the evidence for similarities between leaky faucets and healthy hearts underlying dynamics.Comment: Self-extracting uuencoded postscript file.     Heterogeneity of cell surface glutamate and GABA receptor expression in Shank and CNTN4 autism mouse models

    Autism spectrum disorder (ASD) refers to a large set of neurodevelopmental disorders, which have in common both repetitive behavior and abnormalities in social interactions and communication. Interestingly, most forms of ASD have a strong genetic contribution. However, the molecular underpinnings of this disorder remain elusive. The SHANK3 gene (and to a lesser degree SHANK2) which encode for the postsynaptic density (PSD) proteins SHANK3/SHANK2 and the CONTACTIN 4 gene which encodes for the neuronal glycoprotein CONTACTIN4 (CNTN4) exhibit mutated variants which are associated with ASD. Like many of the other genes associated with ASD, both SHANKs and CNTN4 affect synapse formation and function and are therefore related to the proper development and signaling capability of excitatory and inhibitory neuronal networks in the adult mammal brain. In this study we used mutant/knock-out mice of Shank2 (Shank2-/-), Shank3 (Shank3αβ-/-), and Cntn4 (Cntn4-/-) as ASD-models to explore whether these mice share a molecular signature in glutamatergic and GABAergic synaptic transmission in ASD-related brain regions. Using a biotinylation assay and subsequent western blotting we focused our analysis on cell surface expression of classical several ionotropic glutamate and GABA receptor subunits: GluA1, GluA2, and NR1GluN1 were analyzed for excitatory synaptic transmission, and the α1 subunit of the GABAA receptor was analyzed for inhibitory synaptic transmission. We found that both Shank2-/- and Shank3αβ-/- mice exhibit reduced levels of several cell surface glutamate receptors in most of the analyzed brain regions – especially in the striatum and thalamus – when compared to wildtype controls. Interestingly, even though Cntn4-/- mice also show reduced levels of some cell surface glutamate receptors in the cortex and hippocampus, increased levels of cell surface glutamate receptors were found in the striatum. Moreover, Cntn4-/- mice do not only show brain region-specific alterations in cell surface glutamate receptors but also a downregulation of cell surface GABA receptors in several of the analyzed brain regions. The results of this study suggest that even though mutations in defined genes can be associated with ASD this does not necessarily result in a common molecular phenotype in surface expression of glutamatergic and GABAergic receptor subunits in defined brain regions

    Drivers of Tuberculosis Transmission.

    Measuring tuberculosis transmission is exceedingly difficult, given the remarkable variability in the timing of clinical disease after Mycobacterium tuberculosis infection; incident disease can result from either a recent (ie, weeks to months) or a remote (ie, several years to decades) infection event. Although we cannot identify with certainty the timing and location of tuberculosis transmission for individuals, approaches for estimating the individual probability of recent transmission and for estimating the fraction of tuberculosis cases due to recent transmission in populations have been developed. Data used to estimate the probable burden of recent transmission include tuberculosis case notifications in young children and trends in tuberculin skin test and interferon γ-release assays. More recently, M. tuberculosis whole-genome sequencing has been used to estimate population levels of recent transmission, identify the distribution of specific strains within communities, and decipher chains of transmission among culture-positive tuberculosis cases. The factors that drive the transmission of tuberculosis in communities depend on the burden of prevalent tuberculosis; the ways in which individuals live, work, and interact (eg, congregate settings); and the capacity of healthcare and public health systems to identify and effectively treat individuals with infectious forms of tuberculosis. Here we provide an overview of these factors, describe tools for measurement of ongoing transmission, and highlight knowledge gaps that must be addressed
