Promoter trapping in microalgae using the antibiotic paromomycin as selective agent
The lack of highly active endogenous promoters to drive the expression of transgenes is one of the main drawbacks to achieving efficient transformation of many microalgal species. Using the model chlorophyte Chlamydomonas reinhardtii and the paromomycin resistance APHVIII gene from Streptomyces rimosus as a marker, we have demonstrated that random insertion of the promoterless marker gene and subsequent isolation of the most robust transformants allows for the identification of novel strong promoter sequences in microalgae. Digestion of the genomic DNA with an enzyme that has a unique restriction site inside the marker gene and a high number of target sites in the genome of the microalga, followed by inverse PCR, allows for easy determination of the genomic region, which precedes the APHVIII marker gene. In most of the transformants analyzed, the marker gene is inserted in intragenic regions and its expression relies on its adequate insertion in frame with native genes. As an example, one of the new promoters identified was used to direct the expression of the APHVIII marker gene in C. reinhardtii, showing high transformation efficiencies.Junta de Andalucía (P09-CVI-5053)Junta de Andalucía (BIO-214
Envasado sostenible

Near-Earth asteroids spectroscopic survey at Isaac Newton Telescope
The population of near-Earth asteroids (NEAs) shows a large variety of
objects in terms of physical and dynamical properties. They are subject to
planetary encounters and to strong solar wind and radiation effects. Their
study is also motivated by practical reasons regarding space exploration and
long-term probability of impact with the Earth. We aim to spectrally
characterize a significant sample of NEAs with sizes in the range of 0.25
- 5.5 km (categorized as large), and search for connections between their
spectral types and the orbital parameters. Optical spectra of NEAs were
obtained using the Isaac Newton Telescope (INT) equipped with the IDS
spectrograph. These observations are analyzed using taxonomic classification
and by comparison with laboratory spectra of meteorites. A total number of 76
NEAs were observed. We classified 44 of them as Q/S-complex, 16 as B/C-complex,
eight as V-types, and another eight belong to the remaining taxonomic classes.
Our sample contains 27 asteroids categorized as potentially hazardous and 31
possible targets for space missions including (459872) 2014 EK24, (436724) 2011
UW158, and (67367) 2000 LY27. The spectral data corresponding to (276049) 2002
CE26 and (385186) 1994 AW1 shows the 0.7 m feature which indicates the
presence of hydrated minerals on their surface. We report that Q-types have the
lowest perihelia (a median value and absolute deviation of AU)
and are systematically larger than the S-type asteroids observed in our sample.
We explain these observational evidences by thermal fatigue fragmentation as
the main process for the rejuvenation of NEA surfaces. In general terms, the
taxonomic distribution of our sample is similar to the previous studies and
matches the broad groups of the inner main belt asteroids. Nevertheless, we
found a wide diversity of spectra compared to the standard taxonomic types.Comment: Accepted in Astronomy & Astrophysics (A&A

Exposure to Mixtures of Pollutants in Mexican Children from Marginalized Urban Areas
Background: Exposure to contaminant mixtures in developing countries is an important public health issue. Children are identified as the most susceptible group to adverse health effects due to the exposure. Objective: The aim of this study was to conduct a screening for mixture pollutants in Mexican children in urban marginalized communities. Methods: We analyzed children (aged 6–12 years old) who resided in four urban marginalized communities in San Luis Potosi, Mexico: i) Bellas Lomas (BEL), a site with vehicular traffic; ii) Tercera Chica (TC), a site with brick kilns; Iii) Rincon de San Jose (SJR), a site with a hazardous waste landfill; and (iv) Morales (MOR) a metallurgical zone with copper-arsenic and electrolytic zinc smelters. Polycyclic Aromatic Hydrocarbons (1-hydroxypyrene (1-OHP)), benzene (trans, trans-muconic acid (t,t-MA), manganese, arsenic and fluoride were quantified in urine and lead in blood samples. Findings: Our results indicate that median exposures to manganese were 4.4, 5.2, 5.8 and 6.3 μg/L for BEL, TC, SJR and MOR, respectively. For BEL, fluoride was present at a higher concentration with 2.3 mg/L followed by MOR, TC and SJR with 1.7, 1.5 and 1.2 mg/L respectively. The highest concentrations of arsenic that were found were 11 μg/L in MOR and lead concentration was reported between 4.2 and 6.8 μg/dL, in BEL, TC and MOR. 1-OHP and t,t-MA were higher in TC (0.23 μmol/mol creatinine (cr), 429.7 μg/g cr, respectively) followed by SJR (0.09 μmol/mol cr, 427.4 μg/g cr), MOR (0.03 μmol/mol cr, 258.6 μg/g cr) and BEL (0.06 μmol/mol cr, 220.6 μg/g cr). Conclusion: Considering the large number of people, especially children, exposed to multiple pollutants, it is important to design effective intervention programs that reduce exposure and the resultant risk in the numerous urban marginalized communities in Mexico
