    Non-specific lipid trasnfer protein (nsLTP) : a protein family involved in tomato allergy

La prevalencia de las enfermedades alérgicas ha aumentado drásticamente en las últimas décadas tanto en los países desarrollados como en los países en vías de desarrollo. Entre todas las alergias, las inducidas por alérgenos alimentarios tienen una incidencia que oscila entre el 2% entre los adultos y el 8% en la población infantil. Los síntomas inducidos pueden variar desde gastrointestinales, angioedema, problemas orales o respiratorios, reacciones de urticarias o incluso síntomas sistémicos como la anafilaxia, comprometiendo gravemente la vida de los pacientes.La alergia debe ser reconocida como un problema de salud pública y deben hacerse esfuerzos para su prevención, diagnóstico y óptimo tratamiento. Una adecuada atención al paciente requiere un estudio detallado de su historia clínica, el análisis de los posibles factores ambientales y la realización de pruebas diagnósticas in vivo e in vitro adecuadas. Para ello, las pruebas cutáneas deberán utilizar extractos proteicos estandarizados y con todos los tejidos (semillas, piel y pulpa) bien representados y/o un diagnóstico molecular in vitro con los pertinentes alérgenos purificados...Type I hypersensitivity is a disorder characterized by raised IgE antibody levels against antigens in the environment that are harmless for most people (the so-called allergens). The prevalence of allergic diseases has dramatically increased in both developed and developing countries over the last decades. Among allergies, those induced by food allergens have an incidence that range from 2% among adults to 8% in the child population. The induced symptoms can vary from gastrointestinal, angioedema, oral or respiratory problems, urticarial or even systemic reactions such as anaphylaxis, severely compromising the patient´s life. Allergy should be recognized as a public health problem and efforts should be made towards its prevention, diagnosis and optimal treatment. The optimization of patient care requires a detailed study of the patient’s history, an analysis of the possible environmental exposure factors; and the performance of suitable in vivo and in vitro diagnostic tests.     Direct PCR-free electrochemical biosensing of plant-food derived nucleic acids in genomic DNA extracts. Application to the determination of the key allergen Sola l 7 in tomato seeds

    A novel and disposable electrochemical biosensor for PCR-free and selective detection of Sola l 7, a non-specific lipid transfer protein (nsLTP) found in tomato seeds associated to severe symptoms of tomato-allergic patients, is reported in this work. The methodology involves the formation of DNA/RNA heterohybrids by sandwich hybridization of a specific fragment of the Sola l 7 allergen coding sequence with appropriate RNA probes designed and described for the first time in this work. Labeling was carried out with commercial antibodies specific to the heteroduplexes and secondary antibodies conjugated with HRP onto the surface of magnetic beads. Amperometric transduction was performed upon magnetic capture of the resulting magnetic bioconjugates on screen-printed electrodes using the system H2O2/HQ. A comparison of the sandwich hybridization format with a direct approach as well as between different labeling strategies was performed. The LOD value achieved was 0.2 pM (5 amol in 25 μL).     Quality of life of patients with metastatic pancreatic adenocarcinoma initiating first-line chemotherapy in routine practice

    Despite advances in surgery, radiotherapy, and chemotherapy, pancreatic adenocarcinoma often progresses rapidly and causes death. The physical decline of these patients is expected to impact their quality of life (QoL). Therefore, in addition to objective measures of effectiveness, the evaluation of health-related QoL should be considered a matter of major concern when assessing therapy outcomes. Observational, prospective, multicenter study including patients with metastatic pancreatic adenocarcinoma who started first-line chemotherapy in 12 Spanish centers. Treatment and clinical characteristics were recorded at baseline. Patients' health-related quality of life, ECOG, and Karnofsky index were measured at baseline, at Days 15 and 30, and every four weeks up to 6 months of chemotherapy. Health-related quality of life was measured using the EORTC-QLQ-C30 and EQ-5D questionnaires. Other endpoints included overall survival and progression-free survival. The study sample included 116 patients (median age of 65 years). Mean (SD) scores for the QLQ-C30 global health status scale showed a significant increasing trend throughout the treatment (p = 0.005). Patients with either a Karnofsky index of 70-80 or ECOG 2 showed greater improvement in the QLQ-C30 global health status score than the corresponding groups with better performance status (p ≤ 0.010). Pain, appetite, sleep disturbance, nausea, and constipation significantly improved throughout the treatment (p < 0.005). Patients with QLQ-C30 global health status scores ≥50 at baseline had significantly greater overall survival and progression-free survival (p = 0.005 and p = 0.021, respectively). No significant associations were observed regarding the EQ-5D score. Most metastatic pancreatic adenocarcinoma patients receiving first-line chemotherapy showed an increase in health-related quality of life scores throughout the treatment. Patients with lower performance status and health-related quality of life at baseline tended to greater improvement.     IgE-reactivity pattern of tomato seed and peel nonspecific lipid-transfer proteins after in vitro gastrointestinal digestion

    The influence of gastrointestinal digestion on the immunological properties of three different nonspecific lipid-transfer proteins (nsLTPs) described in tomato fruit has been assessed using an in vitro system mimicking the stomach and intestine digestion conditions. Tomato peel/pulp nsLTP, Sola l 3, was degraded after digestion, although the immunoglobulin E (IgE) recognition of intact protein and a 10 kDa band were still observed after 30 min of duodenal digestion in the presence of phosphatidylcholine. The tomato seed nsLTP, Sola l 7, showed a higher stability than the other seed allergen, Sola l 6, during digestion. Sola l 7 showed an IgE immunoreactive 6.5 kDa band in immunoblotting analysis, retaining up to 7% of its IgE-binding capacity in inhibition ELISA test after 60 min of duodenal digestion and keeping intact its ability to activate basophils after digestion.     En un trabajo previo, publicado en esta misma revista, los autores han estudiado los resultados generados por los proyectos de investigación financiados por el Fondo de Investigación Sanitaria (FIS) a las Instituciones Sanitarias Asistenciales (hospitales) y los procedentes de los cuestionarios de encuestas remitidos a los investigadores responsables de tales proyectos y a los directores gerentes de los hospitales en los que se realizaron los mismos. En el presente trabajo, se continúa el proceso de evaluación del FIS, centrándose la atención en la distribución de los proyectos de investigación concedidos a los centros de investigación, facultades y escuelas, y en los resultados obtenidos a través de los cuestionarios de encuesta cumplimentados por los investigadores principales pertenecientes a estas instituciones o entidades.La realización de este trabajo ha sido posible gracias a la financiación otorgada por el FIS al proyecto de investigación 96/1803.Peer reviewe
