    Potential control of forest diseases by solutions of chitosan oligomers, propolis and nanosilver

    Producción CientíficaThere is a growing necessity to replace chemical agents with ecofriendly materials, arising from the impact on the environment and/or human health, which calls for the design of new broad-spectrum fungicides. In this work, chitosan oligomers (COs), propolis (Ps) and silver nanoparticles (AgNPs) mixtures in solution were assessed to control the growth of different phytopathogenic fungi and oomycetes in vitro. Binary solutions of COs-Ps and COs-AgNPs evinced the highest antifungal effect against Fusarium circinatum and Diplodia pinea fungi, respectively, with a ca. 80% reduction in their mycelial growth. The COs solution by itself also proved to be greatly effective against Gremmeniella abietina, Cryphonectria parasitica and Heterobasidion annosum fungi, causing a reduction of 78%, 86% and 93% in their growth rate, respectively. Likewise, COs also attained a 100% growth inhibition on the oomycete Phytophthora cambivora. On the other hand, Ps inhibited totally the growth of Phytophthora ×alni and Phytophthora plurivora. The application of AgNPs reduced the mycelial growth of F. circinatum and D. pinea. However, the AgNPs in some binary and ternary mixtures had a counter-productive effect on the anti-fungal/oomycete activity.     Application of bioactive coatings based on chitosan and propolis for Pinus spp. protection against Fusarium circinatum

    Pine pitch canker (PPC) is a major threat to pine forests worldwide because of the extensive tree deaths, reduced growth, and degradation of timber quality caused by it. Furthermore, the aggressive fungus responsible for this disease (Fusarium circinatum) can also infect pine seeds, causing damping-off in young seedlings. This study proposes an approach based on coating treatments consisting of natural products to ensure seed protection. Seeds from two pine species (the most sensitive to this disease, Pinus radiata D. Don, and a more resistant one, Pinus sylvestris L.) were coated with single and binary mixtures of low and medium molecular weight chitosan and/or ethanolic-propolis extract. The germination rate, pre- and post-emergence mortality, total phenolic content, and radical scavenging activity were assessed. All treatments, and especially the one based on chitosan oligomers, had a beneficial impact on P. sylvestris seedlings, significantly enhancing survival rates and displaying a positive influence on the total phenolic content and on the seedlings’ radical scavenging activity. Conversely, non-significant negative effects on germination percentages were observed in the case of P. radiata seeds.     Producción CientíficaSimple Summary: This study explored the chemical components of the egg glue used by the Western Conifer Seed Bug (Leptoglossus occidentalis Heidemann, 1910) to agglutinate eggs and adhere to pine needles. Results showed that the adhesive secretion includes plasticizers and thermoplastic elastomer resins with semiochemical properties in an oily matrix containing proteins. This knowledge of the egg glue composition can be used to develop new control strategies for L. occidentalis, potentially limiting the economic impact caused by this pest insect that reduces the production of pine nuts by up to 25%.The western conifer seed bug (Leptoglossus occidentalis Heidemann, 1910, Heteroptera: Coreidae) has a significant economic impact due to the reduction in the quality and viability of conifer seed crops; it can feed on up to 40 different species of conifers, showing a clear predilection for Pinus pinea L. in Europe. Elemental analysis pointed to the presence of significant amounts of compounds with high nitrogen content. Functional groups identified by infrared spectroscopy were compatible with the presence of chitin, scleroproteins, LNSP-like and gelatin proteins, shellac wax analogs, and policosanol. Regarding the chemical species identified by GC–MS, eggs and glue hydromethanolic extracts shared constituents such as butyl citrate, dibutyl itaconate, tributyl aconitate, oleic acid, oleamide, erucamide, and palmitic acid, while eggs also showed stearic and linoleic acid-related compounds.     EcoCyc: a comprehensive database resource for Escherichia coli

    The EcoCyc database (http://EcoCyc.org/) is a comprehensive source of information on the biology of the prototypical model organism Escherichia coli K12. The mission for EcoCyc is to contain both computable descriptions of, and detailed comments describing, all genes, proteins, pathways and molecular interactions in E.coli. Through ongoing manual curation, extensive information such as summary comments, regulatory information, literature citations and evidence types has been extracted from 8862 publications and added to Version 8.5 of the EcoCyc database. The EcoCyc database can be accessed through a World Wide Web interface, while the downloadable Pathway Tools software and data files enable computational exploration of the data and provide enhanced querying capabilities that web interfaces cannot support. For example, EcoCyc contains carefully curated information that can be used as training sets for bioinformatics prediction of entities such as promoters, operons, genetic networks, transcription factor binding sites, metabolic pathways, functionally related genes, protein complexes and protein–ligand interactions

    The comprehensive updated regulatory network of Escherichia coli K-12

    BACKGROUND: Escherichia coli is the model organism for which our knowledge of its regulatory network is the most extensive. Over the last few years, our project has been collecting and curating the literature concerning E. coli transcription initiation and operons, providing in both the RegulonDB and EcoCyc databases the largest electronically encoded network available. A paper published recently by Ma et al. (2004) showed several differences in the versions of the network present in these two databases. Discrepancies have been corrected, annotations from this and other groups (Shen-Orr et al., 2002) have been added, making the RegulonDB and EcoCyc databases the largest comprehensive and constantly curated regulatory network of E. coli K-12. RESULTS: Several groups have been using these curated data as part of their bioinformatics and systems biology projects, in combination with external data obtained from other sources, thus enlarging the dataset initially obtained from either RegulonDB or EcoCyc of the E. coli K12 regulatory network. We kindly obtained from the groups of Uri Alon and Hong-Wu Ma the interactions they have added to enrich their public versions of the E. coli regulatory network. These were used to search for original references and curate them with the same standards we use regularly, adding in several cases the original references (instead of reviews or missing references), as well as adding the corresponding experimental evidence codes. We also corrected all discrepancies in the two databases available as explained below. CONCLUSION: One hundred and fifty new interactions have been added to our databases as a result of this specific curation effort, in addition to those added as a result of our continuous curation work. RegulonDB gene names are now based on those of EcoCyc to avoid confusion due to gene names and synonyms, and the public releases of RegulonDB and EcoCyc are henceforth synchronized to avoid confusion due to different versions. Public flat files are available providing direct access to the regulatory network interactions thus avoiding errors due to differences in database modelling and representation. The regulatory network available in RegulonDB and EcoCyc is the most comprehensive and regularly updated electronically-encoded regulatory network of E. coli K-12

    Actualidad genética y clínica en las Polineuropatías Sensorimotoras Hereditarias

    Como objetivo hemos tenido el ampliar el conocimiento en el tema de las polineuropatías sensorimotoras hereditarias y mostrar los aportes de la genética molecular en la clasificación de esta enfermedad. En el desarrollo, revisamos el tema de las polineuropatías sensorimotoras hereditarias partiendo desde sus primeras descripciones. Se hace énfasis en la generalidad del cuadro clínico, seleccionando inicialmente la clasificación de Dyck &amp; Lambert para luego detallar el gran paso generado por los avances de la genética molecular en la clasificación exacta de este grupo de desórdenes de los nervios periféricos. Se establece como regla para clasificarlo la naturaleza de la polineuropatía, dividiéndola en Desmielinizante y Axonal. En cada uno de estos dos grupos se explica claramente el patrón de herencia (A. Dominante, A recesiva, Ligada al Cromosoma X) con sus respectivas mutaciones y productos proteicos alterados.Conclusiones: Las neuropatías sensorimotoras son un gran síndrome, en el cual la existencia de múltiples mutaciones explica la gran variabilidad en el cuadro clínico y en la expresión de esta enfermedad. El diagnóstico exacto de estas neuropatías parece estar muy ligado a la genética molecular, por lo que la clasificación de Dyck y Lambert es la de mayor utilidad para una correcta interpretación clínica de este síndrome.Palabras clave: CMT, Polineuropatías Hereditarias sensorimotoras, Charcot Marie Tooth, NHMS.</p

    The effect of cold waves on mortality in urban and rural areas of Madrid

    While many studies analyze the effect of extreme thermal events on health, little has been written about the effects of extreme cold on mortality. This scarcity of papers is particularly relevant when we search studies about extreme cold on the health of rural population. Therefore, we tried to analyze the effect of cold waves on urban areas and rural areas from Madrid and to test whether differentiated effects exist between both population classes. For this purpose, we analyzed data from the municipalities with over 10,000 inhabitants for the period from January 1, 2000 through December 31, 2013. Municipalities were classified as urban or rural (Eurostat), and they were grouped into similar climatological zones: Urban Metropolitan Centre (UMC), Rural Northern Mountains (RNM), Rural Centre (RC) and Southern Rural (SR). The dependent variable was the daily mortality rate due to natural causes per million inhabitants (CIE-X: A00-R99) that occurred between the months of November and March for the period. The independent variable was minimum daily temperature (ºC) (Tmin). Social and demographic contextual variables were used, including: population > age 64 (%), deprivation index and housing indicators. The analysis was carried out in three phases: (1) determination of the threshold temperature (Tthreshold) which defines the cold waves; (2) determination of the relative risk (RR) for cold waves using Poisson linear regression (GLM); and (3) using GLM of the binomial family, Odds Ratios (OR) were calculated to analyze the relationship between the frequency of the appearance of cold waves and the socioeconomic variables.The authors gratefully acknowledge the grants for projects ENPY376/18; ENPY470/19 and ENPY 107/18 from the Carlos III Institute of Health
