    Rosacea and perioral dermatitis: a single‐center retrospective analysis of the clinical presentation of 1032 patients

    Background Rosacea is a common chronic inflammatory cutaneous disorder affecting nearly 5.5 % of the adult population. Our aim was to evaluate the prevalence and epidemiology of rosacea and perioral dermatitis (POD) in an ambulatory care setting. Methods We retrospectively analyzed medical data of patients with a confirmed diagnosis of rosacea or perioral dermatitis (POD) presenting at our university hospital outpatient clinic during a 3‐year period. Results Out of 1032 patients, 81.5 % were diagnosed with rosacea and 18.5 % with POD. Overall prevalence was 1.4 % for rosacea and 0.3 % for POD. 69.3 % of the analyzed patients were female. Overall mean age was 49.3 ± 7.7 (1–92) years; the women’s average age was less than the men’s. Patients with POD were younger and predominantly female, whereas patients with phymatous rosacea were older and predominantly male. The most common phenotypes were papulopustular rosacea (68.4 %), erythematotelangiectatic rosacea (22.5 %), and phymatous rosacea (8.0 %). Special forms of rosacea were diagnosed in 15.8 % of the patients; the most frequent were ocular rosacea (6.9 %) and steroid‐induced rosacea (5.4 %). Conclusions The large patient cohort analyzed in our study provides a good estimate of the frequency of the rosacea subtypes, special forms and of perioral dermatitis in a hospital‐based outpatient care setting

    Retrospective analysis of alpha‐human papillomavirus (HPV) types in tissue samples from anogenital dysplasias – introduction of the RICH (Risk of HPV‐related Carcinoma in HIV+/− patients) score

    Background Chronic viral infections caused by highly contagious human papillomaviruses (HPVs) from the alpha genus are a substantial risk factor for tumour diseases. Objectives The goal of this study was to compare the HPV infection pattern with histology in a patient group of immunocompromised HIV+ and non‐immunocompromised patients with anal intraepithelial neoplasia. Materials and Methods Tissue samples (n = 210) from the anogenital area of 121 patients underwent retrospective histological and molecular examination for HPV DNA prevalence by chip analysis. The study was part of a cancer screening from the Dermatology Department of the LMU Munich, Germany. All data were collected and processed anonymously. Results HPV 6 or 11 are more abundant in tissue samples from histologically diagnosed condylomata acuminata (47.7%) compared to grade 1, 2, and 3 intraepithelial neoplasias (IN 1‐3). Detection of high‐risk (hr) alpha‐HPV DNA was significantly higher in tissue samples from IN 3 (67.5%) compared to IN 1 and 2 (12.9%), and compared to condylomata acuminata (29.5%). No HPV types were detected in histologically unremarkable tissue samples. There was a significant association between the prevalence of HPV 16 and the classifications IN 1 to IN 3 (χ2 (2) = 13.62, P = 0.001). We identified a significant correlation between the prevalence of high‐risk and low‐risk (lr) HPV types and HIV, especially mixed infections of different HPV types correlated with high‐grade IN. Based on the present data, we suggest the risk of carcinoma in HIV+/− patients (RICH) score and test it in the 121 patients. Conclusions hr alpha‐HPVs, mainly HPV 16, are associated with increased oncogenic potential of premalignant lesions (IN 1‐3), especially in HIV+ patients. Based on the combination of HIV/HPV‐testing and histological analysis, we identified correlations that could potentially forecast the risk of malignant transformation and summarized them in the form of RICH score

    Comparative analysis of the phototoxicity induced by BRAF inhibitors and alleviation through antioxidants

    Background Small molecules tackling mutated BRAF (BRAFi) are an important mainstay of targeted therapy in a variety of cancers including melanoma. Albeit commonly reported as side effect, the phototoxic potential of many BRAFi is poorly characterized. In this study, we evaluated the phototoxicity of 17 distinct agents and investigated whether BRAFi‐induced phototoxicity can be alleviated by antioxidants. Methods The ultraviolet (UV) light absorbance of 17 BRAFi was determined. Their phototoxic potential was investigated independently with a reactive oxygen species (ROS) and the 3T3 neutral red uptake (NRU) assay in vitro. To test for a possible phototoxicity alleviation by antioxidants, vitamin C, vitamin E phosphate, trolox, and glutathione (GSH) were added to the 3T3 assay of selected inhibitors. Results The highest cumulative absorbance for both UVA and UVB was detected for vemurafenib. The formation of ROS was more pronounced for all compounds after irradiation with UVA than with UVB. In the 3T3 NRU assay, 8 agents were classified as phototoxic, including vemurafenib, dabrafenib, and encorafenib. There was a significant correlation between the formation of singlet oxygen (P = .026) and superoxide anion (P < .001) and the phototoxicity observed in the 3T3 NRU assay. The phototoxicity of vemurafenib was fully rescued in the 3T3 NRU assay after GSH was added at different concentrations. Conclusion Our study confirms that most of the BRAF inhibitors exhibited a considerable phototoxic potential, predominantly after exposure to UVA. GSH may help treat and prevent the phototoxicity induced by vemurafenib

    Dermal fillers do not induce upregulation of NLRP3 inflammasomes or expression of inflammatory cytokines in granulomas

    Background: Filling materials have increasingly been used in aesthetics over the last decades. Understanding the pathophysiology of granuloma formation as a very relevant unwanted side effect of filler application may be essential to help avoid these adverse events. Aims: Our aim was to investigate the role of the inflammasome in the formation of filler granuloma, as a central column of the innate immune response. Methods RPMI 1640 medium was used for growth of THP-1 cells and the induction of THP-1 macrophages. Sonication was applied in order to crush the acrylic particles of the filler. ELISA was the method of analysis for the specific cytokines. Biopsy specimens of filler granuloma were analyzed by various immunohistochemical methods. GraphPad Prism 5 software was used for the statistical data analysis. Results: Neither was the sensor NALP3 overexpressed, nor could an elevated expression of cleaved IL-1 beta, IL-18, or IFN-gamma be detected. Furthermore, no increased expression of IL-8 or IL-1 beta was detectable in vitro. Conclusion No increased inflammasome activation could be observed;however, filler granulomas were infiltrated with granulocytes and macrophages. Therefore, we speculate that an unspecific immune response might be the key player in the formation of filler granuloma

    El empleo de productos vegetales como fuente de compuestos bioactivos presenta algunos inconvenientes que pueden poner en riesgo la salud del consumidor. La información respaldada científicamente sobre su actividad biológica es escasa y es difícil asegurar la identidad de las plantas y detectar adulteraciones. La principal limitante en el control es la falta de métodos analíticos adecuados. Este trabajo presenta el desarrollo de una plataforma analítica basada en LC-MS para determinar compuestos polifenólicos en extractos de cáscara y semillas de uva. Se estudiaron extractos acuosos de semilla y cáscara de uva. Se optimizaron la separación cromatográfica y la detección, evaluando métodos de análisis dirigido y no dirigido, combinando el barrido completo, con la fragmentación utilizando análisis dependiente e independiente de datos. Para la identificación de los compuestos, los datos se analizaron con ayuda de una base de datos de espectros de masas y datos de la literatura. El uso de análisis dependiente de datos con y sin energía escalonada produce cromatogramas y espectros similares; ya que el uso de energía escalonada puede abarcar distintas energías de fragmentación fue el modo de trabajo seleccionado. Los compuestos identificados coinciden con los reportados por otros autores para el mismo tipo de muestras

Aplikasi layanannyapun semakin canggih dan mampu melayani banyak kebutuhan dan keinginan manusia. Para remaja di desa sudah memakai handphone yang canggih, hampir semua tersambung ke internet. Kalau remaja di desa saja sudah sampai sejauh itu, apalagi remaja yang di kota, sudah akrab dengan internet dan penggunaannya. Kemajuannya yang sangat pesat, tidak ditunjang edukasi penggunaan tentang keuntungan dan bahaya penggunaan handphone dengan banyak aplikasi. Sejalan dengan hal ini orang tua memiliki keterbatasan pengetahuan mengenai handphone dan penggunaannya. Motivasi remaja menggunakan handphone terutama bukan kebutuhan tetapi keinginan dan kesenangan seperti untuk game online. Sedangkan kegunaan untuk menunjang kerja dan tugas tidak menjadi urutan pertama dalam jawaban para remaja. Orang tua hanya menegur dan mengeluh saja, hal ini tidak menjawab persoalan para remaja. Penyimpangan yang dilakukan para remaja dalam penggunaan handphone menyebabkan perilaku remaja yang destruktif. Perilaku positif dalam menggunakan handphone merupakan keinginan dan harapan yang besar karena sangat konstruktif untuk mendukung kehidupan remaja di masa mendatang. Hal-hal yang konstruktif itu pada waktunya akan tampak perilaku remaja yang produktif dan kompetitif di lingkungan sosial kehidupanny

    Masih banyak masyarakat saat ini yang belum memahami sepenuhnya bagaimana memilih minyak pelumas yang terbaik untuk sepeda motor matic, khususnya Yamaha Mio. Mereka hanya memilih minyak pelumas berdasarkan rekomendasi dari pabrikan sepeda motor atau dengan melihat merk yang terkenal saja dan tidak memahami bagaimana karakteristik dan jenis dari pelumas tersebut. Dampak salah memilih oli mesin dapat mengalami penurunan performa mesin sepeda motor. Untuk itu, diperlukan diperlukan pengetahuan mengenai kriteria yang menjadi prioritas pertimbangan dalam pemillihan oli mesin serta memudahkan proses pemilihan agar tidak membutuhkan waktu yang panjang dalam pemilihan. Dengan menggunakan metode Analytical Hierarchy Process (AHP) dan dengan teknik pengumpulan data menggunakan observasi, wawancara, kuesioner serta studi pustaka maka dapat membantu dalam pemilihan oli mesin yang sesuai dengan jenis kendaraan matic khususnya Yamaha Mio

    Impact of allergic reactions and urticaria on mental health and quality of life

    Background: Allergic diseases represent a major global health issue with more than one-third of the global population affected by at least one allergic condition. Allergic conditions can not only cause life-threatening anaphylactic reactions but also impact daily life with a significant influence on mental health and the quality of life (QoL). Objectives: This study aims to evaluate the health-related QoL and depression severity among patients presenting in a tertiary care allergy center. Methods: A cross-sectional study was conducted among 596 patients presenting with allergic symptoms or previously diagnosed allergies between October 2018 and April 2019. Patients were screened for depression and the QoL impairment by using three validated scales: the Beck Depression Inventory (BDI), the Dermatologic Life Quality Index (DLQI), and the three-level version of the EuroQol 5-Dimensional (EQ-5D-3L) scale. Results: One-third (34.8%) of the study population was male and two-thirds (65.2%) were female. About 73.7% (n = 427/579) of the patients suffered from at least one previously diagnosed allergic disease, most frequently to pollen (37.0%, n = 214/579) and food (27.3%, n = 158/579), and 20.0 % (n = 116/579) suffered from urticaria. About 19.3% of the total population suffered from depression. Urticaria, as well as insect venom, food/food additives, and drug allergies significantly affected the QoL and depression severity (p < 0.001), reflected by higher DLQI and BDI scores, and lower scores in the EQ5D-3L index. Conclusion: Our results provide evidence for a possible correlation of allergies (e.g. against insect venom, food/food additives, and drugs) and/or urticaria with a reduced QoL and a higher depression rate. Patients particularly indicated restrictions for the dimensions, pain/discomfort as well as anxiety/depression. It might be beneficial to implement a standardized questionnaire as a regular screening method for evaluating the mental health status of patients with allergies and/or urticaria.     Preliminary Study of the Antimicrobial, Anticoagulant, Antioxidant, Cytotoxic, and Anti-Inflammatory Activity of Five Selected Plants with Therapeutic Application in Dentistry

    The usefulness of traditional plants in Mexico to treat human ailments has been known since ancient times. This work evaluated the antimicrobial, anticoagulant, antioxidant, cytotoxic, and anti-inflammatory potential of ethanolic extracts of Aloe vera, Equisetum arvense, Mimosa tenuiflora, Lippia graveolens, and Syzygium aromaticum. The antimicrobial activity of the extracts was evaluated against Streptococcus mutans and Streptococcus sorbinus; a significant inhibitory effect of the L. graveolens extract on both bacteria was observed at concentration levels of 250 &micro;g/mL and greater. The anticoagulant activity was evaluated in terms of prothrombin time (PT) and activated partial thromboplastin time (APTT), A. vera and M. tenuiflora extracts showed no significant difference (p &#706; 0.05) in PT compared with the control, and for APTT the extracts of A. vera, L. graveolens, and S. aromaticum decreased the APTT significantly (p &#706; 0.05) compared with the control. The antioxidant potential by DPPH assay indicated that the E. arvense extract behaved statistically the same as the control. The cytotoxic activity was evaluated in HGF-1 cells using the fluorometric microculture cytotoxicity assay technique, and none of the extracts was toxic at 125 and 250 &micro;g/mL concentrations. Finally, the anti-inflammatory activity was evaluated using ELISA, where the A. vera extract showed the best anti-inflammatory capacity. Further research on the search for bioactive metabolites and elucidation of action mechanisms of the most promising extracts will be carried out
