    Variação intra e interpessoal como fatores determinantes para o cálculo do número de observações para estimar a ingestão dietética habitual de adolescentes

    This cross-sectional study describes the variation in the dietary intake of energy and macronutrients based on estimated coefficients of within- and between-person variation and intends to calculate the number of days required to evaluate the dietary intake of adolescents in Piracicaba, Brazil. The sample was made up by ninety-two adolescents aged between eleven and sixteen years. Interviews were performed to assess demographic, anthropometric and dietary variables, which were evaluated by the answers obtained through two 24-hour recalls. Descriptive analyses were performed and variances and Coefficients of variation within- and between-person were obtained by results of one-way ANOVA. The mean energy intake observed was 2,326.2 kilocalories; mean macronutrient intake came to 89.0 grams of fatty acids, 305.3 grams of carbohydrates and 82.2 grams of proteins. Coefficients of within-person variation ranged from 36% to 45%, while between-person coefficients varied from 26% to 31%. Variance ratios ranged from 1.35 (carbohydrate) to 2.62 (protein). The lowest number of observations required to correctly evaluate the usual diet calculated (r = 0.90) was six days, for carbohydrates while the highest was eleven, for proteins. Coefficients of variation were similar to those observed in international studies. We conclude that, in this study, two observations were sufficient to obtain reasonable correlations. However, six evaluations are necessary for these adolescents in order to obtain excellent correlations.Este estudo transversal descreve a variabilidade do consumo alimentar de energia e macronutrientes a partir da estimativa dos coeficientes de variação intra e interpessoal e calcula o número de dias necessários para se avaliar a ingestão dietética de adolescentes de Piracicaba, Brasil. A amostra foi constituída por 92 adolescentes com idade entre 11 e 16 anos. Por meio de entrevistas, foram avaliadas variáveis demográficas, antropométricas e de ingestão dietética, levantada por meio de dois Recordatórios de 24h. Foram realizadas análises descritivas, sendo que as variâncias e os coeficientes de variação intra e interpessoal foram obtidos a partir dos resultados da ANOVA com um fator de classificação. O consumo médio de energia observado foi de 2.326,2 Kcal, e em relação aos macronutrientes foram observados valores de 89,0g de lipídios, 305,3g de carboidratos e 82,2g de proteínas. Os coeficientes de variação intrapessoal variaram de 36% a 45% enquanto os coeficientes de variação interpessoal variaram de 26% a 31%. A razão entre as variâncias variou de 1,35 (carboidrato) a 2,62 (proteína). O menor número de observações necessárias para avaliar corretamente (r = 0,90) a dieta habitual foi de 6 dias para carboidratos. O maior número foi para proteínas (11 dias). Os coeficientes de variação foram semelhantes aos observados em estudos internacionais. Conclui-se que, para o presente estudo, duas observações foram suficientes para a obtenção de coeficientes de correlação razoáveis.     Handheld near-infrared spectroscopy: state-of-the-art instrumentation and applications in material identification, food authentication, and environmental investigations

    This present review article considers the rapid development of miniaturized handheld near-infrared spectrometers over the last decade and provides an overview of current instrumental developments and exemplary applications in the fields of material and food control as well as environmentally relevant investigations. Care is taken, however, not to fall into the exaggerated and sometimes unrealistic narrative of some direct-to-consumer companies, which has raised unrealistic expectations with full-bodied promises but has harmed the very valuable technology of NIR spectroscopy, rather than promoting its further development. Special attention will also be paid to possible applications that will allow a clientele that is not necessarily scientifically trained to solve quality control and authentication problems with this technology in everyday life.info:eu-repo/semantics/publishedVersio

    An unusual T-cell childhood acute lymphoblastic leukemia harboring a yet unreported near-tetraploid karyotype

    <p>Abstract</p> <p>Background</p> <p>Near-tetraploid (model #81-103) and near-triploid (model #67-81) karyotypes are found in around 1% of childhood acute lymphoblastic leukemia. Due to its rarity, these two cytogenetic subgroups are generally included in the hyperdiploid group (model # > 51). Therefore separate informations about these two subgroups are limited to a few reports. Some studies found that near-tetraploidy is relatively more frequent in higher median ages and it is associated to Frech-American-British Classification subtype L2. Although the mechanisms by which leukemic blast cells divide is still unclear, studies have suggested that hyperdiploidy, near-triploidy and near-tetraploidy do not seem to share the same mechanism.</p> <p>Findings</p> <p>Herewith, we present a new childhood T-acute lymphoblastic leukemia case of near-tetraploid karyotype with loss of two p53-gene copies, characterized in detail by cytogenetic and molecular studies.</p> <p>Conclusion</p> <p>We suggest that p53 is a good target gene to be screened, once p53 is one of the main effectors of cell cycle checkpoints.</p

    South American Expert Roundtable : increasing adaptive governance capacity for coping with unintended side effects of digital transformation

    This paper presents the main messages of a South American expert roundtable (ERT) on the unintended side effects (unseens) of digital transformation. The input of the ERT comprised 39 propositions from 20 experts representing 11 different perspectives. The two-day ERT discussed the main drivers and challenges as well as vulnerabilities or unseens and provided suggestions for: (i) the mechanisms underlying major unseens; (ii) understanding possible ways in which rebound effects of digital transformation may become the subject of overarching research in three main categories of impact: development factors, society, and individuals; and (iii) a set of potential action domains for transdisciplinary follow-up processes, including a case study in Brazil. A content analysis of the propositions and related mechanisms provided insights in the genesis of unseens by identifying 15 interrelated causal mechanisms related to critical issues/concerns. Additionally, a cluster analysis (CLA) was applied to structure the challenges and critical developments in South America. The discussion elaborated the genesis, dynamics, and impacts of (groups of) unseens such as the digital divide (that affects most countries that are not included in the development of digital business, management, production, etc. tools) or the challenge of restructuring small- and medium-sized enterprises (whose service is digitally substituted by digital devices). We identify specific issues and effects (for most South American countries) such as lack of governmental structure, challenging geographical structures (e.g., inclusion in high-performance transmission power), or the digital readiness of (wide parts) of society. One scientific contribution of the paper is related to the presented methodology that provides insights into the phenomena, the causal chains underlying “wanted/positive” and “unwanted/negative” effects, and the processes and mechanisms of societal changes caused by digitalization

    OBJECTIVE: Adequate isolation of nucleic acids from peripheral blood, fine-needle aspiration cells in stained slides, and fresh and formalin-fixed/paraffin-embedded tissues is crucial to ensure the success of molecular endocrinology techniques, especially when samples are stored for long periods, or when no other samples can be collected from patients who are lost to follow-up. Here, we evaluate several procedures to improve current methodologies for DNA (salting-out) and RNA isolation. MATERIALS AND METHODS: We used proteinase K treatment, heat shock, and other adaptations to increase the amount and quality of the material retrieved from the samples. RESULTS: We successfully isolated DNA and RNA from the samples described above, and this material was suitable for PCR, methylation profiling, real-time PCR and DNA sequencing. CONCLUSION: The techniques herein applied to isolate nucleic acids allowed further reliable molecular analyses. Arq Bras Endocrinol Metab. 2012;56(9):618-26OBJETIVO: O isolamento adequado de ácidos nucleicos a partir de sangue periférico, lâmina corada de punção aspirativa por agulha fina, tecido fixado em formalina e emblocado em parafina e tecido fresco é fundamental para assegurar o sucesso de técnicas aplicadas em endocrinologia molecular, principalmente quando lidamos com amostras estocadas por longos períodos ou quando há impossibilidade de nova coleta de amostra de pacientes que perderam o seguimento. Neste trabalho, objetivamos otimizar as metodologias clássicas para a extração de DNA (salting-out) e RNA. MATERIAIS E MÉTODOS: Utilizamos proteinase K, choque térmico, dentre outras modificações, com o objetivo de aumentar a quantidade e a qualidade do material recuperado a partir das amostras descritas acima. RESULTADOS: Isolamos com sucesso DNA e RNA de tais amostras e o material obtido foi adequado para a realização de PCR, perfil de metilação, PCR em tempo real e sequenciamento de DNA. CONCLUSÃO: As técnicas aplicadas neste estudo para isolar ácidos nucleicos permitiram a realização posterior de análises moleculares consistentes e confiáveis.
