Under the motto “The best schools are those that improve”, the Regional Directorate of School Administration (DRAE), an organization of the Regional Secretary for Education, Science and Technology of the Autonomous Region of Madeira, has encouraged and supported schools to implement the Regional Education System Quality Assessment process in what self-assessment is concerned. This process has demanded a huge investment from both authorities and schools (in terms of human resources, training and time) whose ultimate objective is to improve the quality of educational services. Considering the relevance that schools' structuring documents assume in strategic planning, it seems clear to us that their strategy must reflect their intention to improve, that is why it was decided to observe the impact of the work carried out through the analysis of schools' strategic documents. Therefore, this study aimed to assess the coherence between the diagnosed situation, the strategy outlined for improvement and its respective operationalization; to observe the consistency and sustainability of the proposals prepared; to gather information in order to reinforce/reformulate support to schools in their improvement process. For the purpose, a set of analysis criteria was established for each document and their articulation and documents from 16 schools were analyzed. This communication intends to report the conclusions gathered through the study. A presente comunicação pretende relatar as conclusões obtidas a partir do estudo realizado. &nbsp

Socio-economic characterization of olive groves in a region in the south of Portugal

Serpa and Moura region, in the South of Portugal, have important agricultural potential as well as important agricultural resources that can and should be used and developed, in spite of economic and social depression signs reinforced by demographic difficulties that can lead to desertification in some areas. The main farms' Technical-Economic Orientation (TEO) is olives production, to produce olive oil, since this region has excellent conditions for the activity development. Some mobilizations traditionally made in olive groves lead to soil erosion and make the farm vehicles mobility much more difficult, namely those used in cultural operations. The maintenance of a soil cover in the olive grove space between lines is good, both because it favours the vehicles mobility, it promotes the rain water infiltration and, last but not the least, the soil airing. The objective of this work has been to make a socio-economic characterization of a zone of Serpa and Moura Councils in which a project of soil covers under olive trees is being developed. At the same time cultural accounts were made for the different situations under the study - irrigated olive grove with spontaneous vegetation in the space between lines, irrigated olive grove with seeded vegetation in the space between lines, irrigated olive grove with herbicide application in the space between lines, dry olive grove with spontaneous vegetation in the space between lines, dry olive grove with seeded vegetation in the space between lines and dry olive grove with herbicide application in the space between lines. The survey shows farmers in this area are younger then usual, although they don't have a high level of formal education, general or specific in agriculture. Farm areas are usually high, which can be determinant for the proposed technology adoption. Installation and operation costs for the olive grove are higher on the irrigated olive grove, but of course expected productions are also higher. For this production technology profits coming from olives production are higher then the operation costs. However, in the dry olive grove technology costs are always higher then the profits, being profitability only due to subsidies. The importance of soil cover maintenance, in this region, beside the benefits in what concerns erosion, infiltration and soil airing, is also due to its capacity to minimize the risk farmer's face

PURPOSE: To identify an immunohistochemical pattern of epithelial markers in granular, lattice and Avellino corneal dystrophies. METHODS: Twenty-two corneal buttons, diagnosed as lattice (17), Avellino (4) and granular (1) underwent immunohistochemical studies of cytokeratins (CKs) on pa- raffin-embedded sections (group I). Monoclonal antibodies for pan-CK (AE1/AE3) and CKs 3/12, 5/6, 8, 18 and 19 were used. Twenty-two normal corneas were used as the control (group II). RESULTS: Six lattice and 2 Avellino cases of group I stained positively with anti-CK 3/12 in corneal epithelium and areas of corneal stroma deposits. One of these cases of lattice was positive for anti-pan-CK (AE1/AE3) also in the epithelium and areas of corneal stroma deposits with a similar pattern. None of the controls (group II) revealed any staining in corneal stroma. All disease and control cases (groups I and II) revealed positive staining in corneal epithelium. CONCLUSION: AE1/AE3 and CK 3/12 anti-CK positive markers in the stromal deposits of lattice and Avellino dystrophies may suggest an epithelial genesis of the disease

<p>Abstract</p> <p>Background</p> <p>Telomeres are specialized structures at the end of chromosomes essential for maintaining genome stability and cell viability. The importance of telomeric proteins for telomere maintenance has increased our interest in the identification of homologues within the genus <it>Leishmania</it>. The mammalian TRF1 and TRF2 proteins, for example, bind double-stranded telomeres via a Myb-like DNA-binding domain and are involved with telomere length regulation and chromosome end protection. In addition, TRF2 can modulate the activity of several enzymes and influence the conformation of telomeric DNA. In this work, we identified and characterized a <it>Leishmania </it>protein (LaTRF) homologous to both mammalian TRF1 and TRF2.</p> <p>Results</p> <p>LaTRF was cloned using a PCR-based strategy. ClustalW and bl2seq sequence analysis showed that LaTRF shared sequence identity with the <it>Trypanosoma brucei </it>TRF (TbTRF) protein and had the same degree of sequence similarities with the dimerization (TRFH) and the canonical DNA-binding Myb-like domains of both mammalian TRFs. LaTRF was predicted to be an 82.5 kDa protein, indicating that it is double the size of the trypanosome TRF homologues. Western blot and indirect immunofluorescence combined with fluorescence <it>in situ </it>hybridization showed that LaTRF, similarly to hTRF2, is a nuclear protein that also associates with parasite telomeres. Native and full length LaTRF and a mutant bearing the putative Myb-like domain expressed in bacteria bound double-stranded telomeric DNA <it>in vitro</it>. Chromatin immunoprecipitation showed that LaTRF interacted specifically with telomeres <it>in vivo</it>.</p> <p>Conclusion</p> <p>The nuclear localization of LaTRF, its association and co-localization with parasite telomeres and its high identity with TbTRF protein, support the hypothesis that LaTRF is a <it>Leishmania </it>telomeric protein.</p

Uma resposta conservacionista: o impacto do sistema plantio direto.

    Get PDF

Xylooligosaccharides (XOS) are widely used in the food industry as prebiotic components. XOS with high purity are required for practical prebiotic function and other biological benefits, such as antioxidant and inflammatory properties. In this work, we immobilized the recombinant endo-1,4-β-xylanase of Malbranchea pulchella (MpXyn10) in various chemical supports and evaluated its potential to produce xylooligosaccharides (XOS) from hydrothermal liquor of eucalyptus wood chips. Values >90% of immobilization yields were achieved from amino-activated supports for 120 min. The highest recovery values were found on Purolite (142%) and MANAE-MpXyn10 (137%) derivatives, which maintained more than 90% residual activity for 24 h at 70 °C, while the free-MpXyn10 maintained only 11%. In addition, active MpXyn10 derivatives were stable in the range of pH 4.0–6.0 and the presence of the furfural and HMF compounds. MpXyn10 derivatives were tested to produce XOS from xylan of various sources. Maximum values were observed for birchwood xylan at 8.6 mg mL−1 and wheat arabinoxylan at 8.9 mg mL−1, using Purolite-MpXyn10. Its derivative was also successfully applied in the hydrolysis of soluble xylan present in hydrothermal liquor, with 0.9 mg mL−1 of XOS after 3 h at 50 °C. This derivative maintained more than 80% XOS yield after six cycles of the assay. The results obtained provide a basis for the application of immobilized MpXyn10 to produce XOS with high purity and other high-value-added products in the lignocellulosic biorefinery field. The authors gratefully acknowledge FAPESP (São Paulo Research Foundation, grants No: 2018/07522-6) and FCT (POCI-01-0145-FEDER-032206)—transnational cooperation project EcoTech, and National Institute of Science and Technology of Bioethanol, INCT, CNPq 465319/2014-9/FAPESP n ◦ 2014/50884- 5) for financial support.
