    Comparison of the talk test and percent heart rate reserve for exercise prescription

    Exercise intensity is traditionally prescribed using %HRmax, %HRR, %VO2max, or %VO2R. Recently, the Talk Test (TT) has been proposed as an alternative method to guide exercise intensity. However, it is unknown if prescribing exercise intensity solely using the TT can provoke training responses that are comparable to traditional guidelines. This study compared the responses to training using either the TT or %HRR. Forty-four subjects (17 males and 27 females: age=20.4±3.02 years; body height=170.5±9.79 cm; body weight=71.9±13.63 kg) completed an incremental maximal cycle ergometer test, were stratified by VO2max and gender, and randomly assigned to training groups guided by either %HRR (n=20) or the TT (n=24). Both groups completed 40-minute training sessions three days per week for 10 weeks. In the HRR group, exercise intensity was targeted (per ACSM guidelines) at 40-59% HRR for weeks 1-4, 50-59% HRR for weeks 5-8, and 60-79% HRR for weeks 9-10. In the TT group, exercise intensity was targeted at the highest power output (PO) that still allowed for comfortable speech. Changes in VO2max, peak power output (PPO), VO2 at ventilatory threshold (VT), and PO at VT were compared between the groups using two-way ANOVA with repeated measures. There were significant (p0.05) interaction effect. Guiding exercise prescription using the TT is a simple and effective method for prescribing exercise intensity and elicits improvements in exercise performance that are comparable to the traditional %HRR guidelines

    Estudio de las reacciones de hidratación temprana en el hormigón proyectado

    [ES] El hormigón proyectado es un tipo de hormigón especial utilizado como soporte de rocas durante la construcción de túneles y excavación de minas, entre otras aplicaciones. Sus principales características son el fraguado rápido y el desarrollo de una resistencia mecánica muy alta a edades tempranas, lo cual permite que el hormigón se fije a la base sin necesidad de un soporte extra, endureciéndose en pocos minutos. Las reacciones de hidratación que ocurren en las primeras horas en el hormigón proyectado determinan el desarrollo de las propiedades mecánicas y su estudio es de crucial importancia para entender y optimizar el comportamiento de este tipo de hormigones. El estudio de estas reacciones, sin embargo, resulta relativamente complicado por varias razones: (i) el hormigón proyectado endurece muy rápido, lo cual dificulta su manejo durante los ensayos experimentales, y (ii) composiciones similares mezcladas en el laboratorio no se comportan de la misma forma que las proyectadas.     [lean Production And Psychosocial Risks: The Case Of A Multinational Merger In A Metallurgical Company In Brazil].

    This study focused on the method known as lean production as a work-related psychosocial risk factor in a Brazilian multinational auto parts company after its merger with other multinational companies. The authors conducted a qualitative analysis of two time points: the first using on-site observation and key interviews with managers and workers during implementation of lean production in 1996; the second, 16 years later, comparing data from a document search in labor inspection records from the Ministry of Labor and Employment and legal proceedings initiated by the Office of the Public Prosecutor for Labor Affairs. The merger led to layoffs, replacements, and an increase in the workday. A class action suit was filed on grounds of aggravated working conditions. The new production model led to psychosocial risks that increased the need for workers' health precautions when changes in the production process introduced new and increased risks of physical and mental illnesses.301765-7

    This study provides the proof of principle that probing the host and the microbe transcriptomes simultaneously is a valuable means to accessing unique information on host pathogen interactions. Our results also underline the extraordinary plasticity of host cell and pathogen responses to infection, and provide a solid framework to further understand the complex mechanisms involved in immunity to M. tuberculosis and in mycobacterial adaptation to different intracellular environments

    Attenuation of Toll-Like Receptor Expression and Function in Latent Tuberculosis by Coexistent Filarial Infection with Restoration Following Antifilarial Chemotherapy

    Mycobacterium tuberculosis (Mtb) and filarial coinfection is highly prevalent, and the presence of filarial infections may regulate the Toll-like receptor (TLR)-dependent immune response needed to control Mtb infection. By analyzing the baseline and mycobacterial antigen–stimulated expression of TLR1, 2, 4, and 9 (in individuals with latent tuberculosis [TB] with or without filarial infection), we were able to demonstrate that filarial infection, coincident with Mtb, significantly diminishes both baseline and Mtb antigen-specific TLR2 and TLR9 expression. In addition, pro-inflammatory cytokine responses to TLR2 and 9 ligands are significantly diminished in filaria/TB-coinfected individuals. Definitive treatment of lymphatic filariasis significantly restores the pro-inflammatory cytokine responses in individuals with latent TB. Coincident filarial infection exerted a profound inhibitory effect on protective mycobacteria-specific TLR-mediated immune responses in latent tuberculosis and suggests a novel mechanism by which concomitant filarial infections predispose to the development of active tuberculosis in humans

    Molecular Blocking of CD23 Supports Its Role in the Pathogenesis of Arthritis

    BACKGROUND: CD23 is a differentiation/activation antigen expressed by a variety of hematopoietic and epithelial cells. It can also be detected in soluble forms in biological fluids. Initially known as the low-affinity receptor for immunoglobulin E (Fc epsilonRII), CD23 displays various other physiologic ligands such as CD21, CD11b/c, CD47-vitronectin, and mannose-containing proteins. CD23 mediates numerous immune responses by enhancing IgE-specific antigen presentation, regulating IgE synthesis, influencing cell differentiation and growth of both B- and T-cells. CD23-crosslinking promotes the secretion of pro-inflammatory mediators from human monocytes/macrophages, eosinophils and epithelial cells. Increased CD23 expression is found in patients during allergic reactions and rheumatoid arthritis while its physiopathologic role in these diseases remains to be clarified. METHODOLOGY/PRINCIPAL FINDINGS: We previously generated heptapeptidic countrestructures of human CD23. Based on in vitro studies on healthy and arthritic patients' cells, we showed that CD23-specific peptide addition to human macrophages greatly diminished the transcription of genes encoding inflammatory cytokines. This was also confirmed by significant reduction of mediator levels in cell supernatants. We also show that CD23 peptide decreased IgE-mediated activation of both human and rat CD23(+) macrophages. In vivo studies in rat model of arthritis showed that CD23-blocking peptide ameliorates clinical scores and prevent bone destruction in a dose dependent manner. Ex-vivo analysis of rat macrophages further confirmed the inhibitory effect of peptides on their activation. Taken together our results support the role of CD23 activation and subsequent inflammatory response in arthritis. CONCLUSION: CD23-blocking peptide (p30A) prevents the activation of monocytes/macrophages without cell toxicity. Thus, targeting CD23 by antagonistic peptide decreases inflammatory markers and may have clinical value in the treatment of human arthritis and allergic reactions involving CD23
