    The Left-To-Die Boat: Review 2

    In March 2011 an inflatable boat carrying 72 asylum seekers from sub-Saharan Africa set out from the coast of Libya hoping to reach the Italian island of Lampedusa. As one Italian official commented, sailing from Libya towards Italy should have been ‘a bit like doing a slalom between military ships’. Yet as, out of fuel, supplies of food and water dwindled to nothing and the people on board began to get sick and die, the boat continued to drift and no help came. Eventually it floated all the way back to the Libyan coast. Of the 50 men, 20 women and two babies who set out to sea, only eleven remained alive. Sharon Davis and Geoff Parish reconstruct this tragedy through interviews with five young men who survived the trip. Their first-person accounts of the journey are augmented by interviews with sympathetic Dutch politician Tineke, Rome-based Eritrean Priest Father Mussie Zerai, who was contacted by the passengers by satellite phobe, human rights investigator Lorenzo Pezzani, who forensically reconstructed the boat’s movements, and dour NATO spokesperson Oana Lungescu. One survivor (Abu Kurke) speaks in halting English; for the others the vernacular is faded under voice-overs. It is raw, simple narrative, expertly selected and edited to move the story seamlessly forward while also allowing listeners time to form mental images of the boat and its passengers and to absorb the emotional impact of what they are hearing. The survivors’ individual accounts overlap and repeat to poetic effect, underscoring the veracity of their testimony. The survivors’ tale floats on a bed of sound – the splash of waves; the hum of a boat motor; restrained, ominous music (with cello predominating), and the occasionally distant chime from an old clock tower marking the relentless passing of time and its deadly impact on the voyage. The radio makers’ approach is dignified, restrained and powerful. Just as the clean lines of minimalist architecture can rely on sophisticated engineering, so the apparent simplicity of a chronological account conceals the artistry, deep thought and sheer hard work of radio production. A strong undercurrent of anger flows through the documentary, and we are given strong encouragement to doubt that NATO has been fully truthful in its response to the tragedy. The Left-To-Die Boat won a United Nations Association of Australia Media Peace Prize and was shortlisted for a Sony Academy award (UK), the New South Wales Premier\u27s Literary Awards and the Australian Human Rights Awards

    Mapping the Galactic Halo with blue horizontal branch stars from the 2dF quasar redshift survey

    We use 666 blue horizontal branch (BHB) stars from the 2Qz redshift survey to map the Galactic halo in four dimensions (position, distance and velocity). We find that the halo extends to at least 100 kpc in Galactocentric distance, and obeys a single power-law density profile of index ~-2.5 in two different directions separated by 150 degrees on the sky. This suggests that the halo is spherical. Our map shows no large kinematically coherent structures (streams, clouds or plumes) and appears homogeneous. However, we find that at least 20% of the stars in the halo reside in substructures and that these substructures are dynamically young. The velocity dispersion profile of the halo appears to increase towards large radii while the stellar velocity distribution is non Gaussian beyond 60 kpc.     Long-term safety and efficacy of teriflunomide : nine-year follow-up of the randomized TEMSO study

    This study was funded by Sanofi Genzyme.OBJECTIVE: To report safety and efficacy outcomes from up to 9 years of treatment with teriflunomide in an extension (NCT00803049) of the pivotal phase 3 Teriflunomide Multiple Sclerosis Oral (TEMSO) trial (NCT00134563). METHODS: A total of 742 patients entered the extension. Teriflunomide-treated patients continued the original dose; those previously receiving placebo were randomized 1:1 to teriflunomide 14 mg or 7 mg. RESULTS: By June 2013, median (maximum) teriflunomide exposure exceeded 190 (325) weeks per patient; 468 patients (63%) remained on treatment. Teriflunomide was well-tolerated with continued exposure. The most common adverse events (AEs) matched those in the core study. In extension year 1, first AEs of transient liver enzyme increases or reversible hair thinning were generally attributable to patients switching from placebo to teriflunomide. Approximately 11% of patients discontinued treatment owing to AEs. Twenty percent of patients experienced serious AEs. There were 3 deaths unrelated to teriflunomide. Soon after the extension started, annualized relapse rates and gadolinium-enhancing T1 lesion counts fell in patients switching from placebo to teriflunomide, remaining low thereafter. Disability remained stable in all treatment groups (median Expanded Disability Status Scale score ≀2.5; probability of 12-week disability progression ≀0.48). CONCLUSIONS: In the TEMSO extension, safety observations were consistent with the core trial, with no new or unexpected AEs in patients receiving teriflunomide for up to 9 years. Disease activity decreased in patients switching from placebo and remained low in patients continuing on teriflunomide. CLASSIFICATION OF EVIDENCE: This study provides Class III evidence that long-term treatment with teriflunomide is well-tolerated and efficacy of teriflunomide is maintained long-term.Publisher PDFPeer reviewe

    Comparative in situ analyses of cell wall matrix polysaccharide dynamics in developing rice and wheat grain

    Cell wall polysaccharides of wheat and rice endosperm are an important source of dietary fibre. Monoclonal antibodies specific to cell wall polysaccharides were used to determine polysaccharide dynamics during the development of both wheat and rice grain. Wheat and rice grain present near synchronous developmental processes and significantly different endosperm cell wall compositions, allowing the localisation of these polysaccharides to be related to developmental changes. Arabinoxylan (AX) and mixed-linkage glucan (MLG) have analogous cellular locations in both species, with deposition of AX and MLG coinciding with the start of grain filling. A glucuronoxylan (GUX) epitope was detected in rice, but not wheat endosperm cell walls. Callose has been reported to be associated with the formation of cell wall outgrowths during endosperm cellularisation and xyloglucan is here shown to be a component of these anticlinal extensions, occurring transiently in both species. Pectic homogalacturonan (HG) was abundant in cell walls of maternal tissues of wheat and rice grain, but only detected in endosperm cell walls of rice in an unesterified HG form. A rhamnogalacturonan-I (RG-I) backbone epitope was observed to be temporally regulated in both species, detected in endosperm cell walls from 12 DAA in rice and 20 DAA in wheat grain. Detection of the LM5 galactan epitope showed a clear distinction between wheat and rice, being detected at the earliest stages of development in rice endosperm cell walls, but not detected in wheat endosperm cell walls, only in maternal tissues. In contrast, the LM6 arabinan epitope was detected in both species around 8 DAA and was transient in wheat grain, but persisted in rice until maturity
