    Main properties of sands hydrophobized by alkoxysilane emulsions

    The results of laboratory tests and geotechnical view on sands hydrophobized by alkoxysilane emulsions are presented. For the soils subjected to this process, very low permeability and no capillary elevation was observed. Laboratory tests also indicate that other physical and mechanical properties of hydrophobized sands remain nearly untouched, i.e., the considered hydrophobisation process does not reduce the strength of soils. Properly composed alkoxysilane emulsions can also solidify in pores to produce a stabilizing silicate binder. The filtration barriers in ground and soil stabilization are thus considered as possible applications of the hydrophobized soils.     Automated measurement of foot deformities : flatfoot, high arch, calcaneal fracture

    Radiographic measurements of foot deformities are used to determine, among other things, such conditions as flatfoot, high arch, or calcaneal fracture. Those measurements are achieved by estimating four angles. Manual assessment of those angles is time-consuming not to mention inevitable errors of such approximation. To the best of the authors knowledge, currently there is no research focusing on finding those four angles. In this paper an algorithm for automatic assessment of those angles, based on extremely randomized trees, is being proposed. Moreover this diagnostic assisting system was intended to be as generic as possible and could be applied, to some degree, to other similar problems. To demonstrate usefulness of this method, correlations of automated measurements with manual ones against correlations of manual measurements with manual ones are being compared. The significance level for manual-manual measurements comparison is less than 0.001 in case of all four angles. The significance level for automated-manual measurements comparison is also less than 0.001 in all cases. The results show that the search for the aforementioned angles can be automated. Even with the use of a generic algorithm a high degree of precision can be achieved, allowing for a more efficient diagnosis

    This article is devoted to the issues of assessing the operational functionalities of the vehicle supercharging system on the turbocharge. The article presents assessment of turbocharger technical state verification procedure includes supercharging parameters, damage analysis, turbine blade strength analysis for chosen vehicles, and turbocharger impeller stand tests. The most frequently occurring failures and defects of supercharging devices are presented included their reasons. The main part of the work includes the strength analysis of the impeller blades, the verification of their deviations, and the stand tests by using specialized equipment

    Characterization of exceptionally thermostable single-stranded DNA-binding proteins from Thermotoga maritima and Thermotoga neapolitana

    <p>Abstract</p> <p>Background</p> <p>In recent years, there has been an increasing interest in SSBs because they find numerous applications in diverse molecular biology and analytical methods.</p> <p>Results</p> <p>We report the characterization of single-stranded DNA binding proteins (SSBs) from the thermophilic bacteria <it>Thermotoga maritima </it>(<it>Tma</it>SSB) and <it>Thermotoga neapolitana </it>(<it>Tne</it>SSB). They are the smallest known bacterial SSB proteins, consisting of 141 and 142 amino acid residues with a calculated molecular mass of 16.30 and 16.58 kDa, respectively. The similarity between amino acid sequences of these proteins is very high: 90% identity and 95% similarity. Surprisingly, both <it>Tma</it>SSB and <it>Tne</it>SSB possess a quite low sequence similarity to <it>Escherichia coli </it>SSB (36 and 35% identity, 55 and 56% similarity, respectively). They are functional as homotetramers containing one single-stranded DNA binding domain (OB-fold) in each monomer. Agarose mobility assays indicated that the ssDNA-binding site for both proteins is salt independent, and fluorescence spectroscopy resulted in a size of 68 ± 2 nucleotides. The half-lives of <it>Tma</it>SSB and <it>Tne</it>SSB were 10 h and 12 h at 100°C, respectively. When analysed by differential scanning microcalorimetry (DSC) the melting temperature (<it>T</it><sub>m</sub>) was 109.3°C and 112.5°C for <it>Tma</it>SSB and <it>Tne</it>SSB, respectively.</p> <p>Conclusion</p> <p>The results showed that <it>Tma</it>SSB and <it>Tne</it>SSB are the most thermostable SSB proteins identified to date, offering an attractive alternative to <it>Taq</it>SSB and <it>Tth</it>SSB in molecular biology applications, especially with using high temperature e. g. polymerase chain reaction (PCR).</p
