    Spatial and Temporal Variations of Microplastics within Humboldt Bay, California

    This study aimed to quantify microplastic (MP) concentration and analyze the spatial and temporal variabilities of the concentrations during the tidal cycle in Humboldt Bay, California. To get an approximation of MP concentration, both water and sediment samples were taken at five different stations, twice during one tidal cycle. Sampling was conducted during two different cruises, on the 19th and 21st of September 2020. The samples were processed in the lab using a density separation procedure and filtration. MP concentrations in the different samples were determined using an average optical microscopy count. Comparison of the water column MP concentrations during ebb and flood tides shows higher concentrations during flood tide, 49.0 particles/L ± 32.37 (flood) vs 34.4 particles/L ± 16.32 (ebb), indicating that MPs are brought into Humboldt Bay from the ocean. The comparison of the MP concentrations during lower energy and higher energy conditions indicates that concentrations in the water column were elevated when there was greater tidal kinetic energy, approximated by the covariance of the measured velocity in North Bay Channel. This result was assumed to be caused by the strong tidal currents stirring up both sediments and the settled MPs into the water column. Due to lower tidal kinetic energy on the sediment sampling cruise day, we could not confirm that assumption. Water samples indicated that MPs are heterogeneously distributed in the bay, with higher concentrations found near the Entrance Channel and lower concentrations found further north in the bay. Sediment samples also indicate a heterogeneous distribution of MPs in the bay, with the lowest concentrations near the Entrance Channel, 15 particles/kg, where high tidal currents inhibit settling of particles

    Scientific response to a cluster of shark bites

    1. Shark bites are of high public concern globally. Information on shark occurrence and behaviour, and of the effects of human behaviours, can help understand the drivers of shark-human interactions. In Australia, a number of shark bite clusters occurred over the last decade. One of these took place in Cid Harbour the Whitsundays, Queensland, a region for which little was known about the shark community. Here, we describe and evaluate the research in response to that shark bite cluster. 2. Fishing methods, acoustic and satellite tracking, and baited remote underwater video cameras (BRUVs) were used to identify the shark species using Cid Harbour, estimate relative abundance, and describe habitat use and residency. Side-scan sonar and BRUVs were also used to assess prey availability. Recreational users were surveyed to understand human behaviour and their awareness and perceptions of ‘Shark Smart’ behaviours. This allowed shark occurrence and behaviour to be interpreted in the context of human behaviours in the Harbour. 3. Eleven shark species were identified. Relative abundance was not unusually high, and residency in Cid Harbour was typically low. For example, 79% of acoustically tagged sharks visited the harbour on <10% days at liberty. Shark prey was available year-round. Notably, anchored boats regularly conduct activities that can attract sharks (dumping food scraps, provisioning and cleaning fish). 4. Alone, the methods used in this study had variable success, but combined they provided a large amount of complementary information. Including a social science component in the research response to the shark bite incidents allowed for a more holistic understanding of the Cid Harbour bite incidents. 5. This study did not identify anything unusual about the shark community that could have contributed to the Cid Harbour shark bite cluster. However, the three incidents involved people bitten almost instantly after entering the water, which is unusual and suggests that feeding/attracting sharks to boats could have been a contributor and also that any species capable of biting humans could have been responsible. 6. The eradication of activities that attract sharks to areas where people enter the water may reduce shark bite risk

    1-arcsecond imaging strategy for the LoTSS survey using the International LOFAR Telescope

    We present the first wide area (2.5 x 2.5 square degrees), deep (median noise of approximately 80 microJy per beam) LOFAR High Band Antenna image at a resolution of 1.2 arcseconds by 2 arcseconds. It was generated from an 8-hour International LOFAR Telescope (ILT) observation of the ELAIS-N1 field at frequencies ranging from 120 to 168 MHz with the most up-to-date ILT imaging strategy. This intermediate resolution falls between the highest possible resolution (0.3 arcseconds) achievable by using all International LOFAR Telescope (ILT) baselines and the standard 6-arcsecond resolution in the LoTSS (LOFAR Two-meter Sky Survey) image products utilizing the LOFAR Dutch baselines only. This is the first demonstration of the feasibility of approximately 1 arcsecond imaging using the ILT, providing unique information on source morphology at scales below the surface brightness limits of higher resolutions. The total calibration and imaging time is approximately 52,000 core hours, nearly five times more than producing a 6-arcsecond image. We also present a radio source catalog containing 2263 sources detected over the 2.5 x 2.5 square degrees image of the ELAIS-N1 field, with a peak intensity threshold of 5.5 sigma. The catalog has been cross-matched with the LoTSS deep ELAIS-N1 field radio catalog, and its flux density and positional accuracy have been investigated and corrected accordingly. We find that approximately 80% of sources that we expect to be detectable based on their peak brightness in the LoTSS 6-arcsecond image are detected in this image, which is approximately a factor of two higher than for 0.3 arcsecond imaging in the Lockman Hole, implying there is a wealth of information on these intermediate scales.Comment: Submitted to A&

    Antibiotic Cocktail for Pediatric Acute Severe Colitis and the Microbiome : The PRASCO Randomized Controlled Trial

    Background: Alterations in the microbiome have been postulated to drive inflammation in IBD. In this pilot randomized controlled trial, we evaluated the effectiveness of quadruple antibiotic cocktail in addition to intravenous-corticosteroids (IVCSs) in acute severe colitis (ASC). Methods: Hospitalized children with ASC (pediatric ulcerative colitis activity index [PUCAI] >= 65) were randomized into 2 arms: the first received antibiotics in addition to IVCS (amoxicillin, vancomycin, metronidazole, doxycycline/ciprofloxacin [IVCS+AB]), whereas the other received only IVCS for 14 days. The primary outcome was disease activity (PUCAI) at day 5. Microbiome was analyzed using 16S rRNA gene and metagenome. Results: Twenty-eight children were included: 16 in the AB + IVCS arm and 12 in the IVCS arm (mean age 13.9 +/- 4.1 years and 23 [82%] with extensive colitis). The mean day-5 PUCAI was 25 +/- 16.7 vs 40.4 +/- 20.4, respectively (P = 0.037). Only 3 and 2 children, respectively, required colectomy during 1-year follow-up (P = 0.89). Microbiome data at time of admission were analyzed for 25 children, of whom 17 (68%) had a predominant bacterial species (>33% abundance); response was not associated with the specific species, whereas decreased microbiome diversity at admission was associated with day-5 response in the IVCS arm. Conclusion: Patients with ASC have alterations in the microbiome characterized by loss of diversity and presence of predominant bacterial species. Quadruple therapy in addition to IVCS improved disease activity on day 5, but larger studies are needed to determine whether this is associated with improved long-term outcomes.Peer reviewe

    Healthy Cities Phase V evaluation: further synthesizing realism

    In this article we reflect on the quality of a realist synthesis paradigm applied to the evaluation of Phase V of the WHO European Healthy Cities Network. The programmatic application of this approach has led to very high response rates and a wealth of important data. All articles in this Supplement report that cities in the network move from small-scale, time-limited projects predominantly focused on health lifestyles to the significant inclusion of policies and programmes on systems and values for good health governance. The evaluation team felt that, due to time and resource limitations, it was unable to fully exploit the potential of realist synthesis. In particular, the synthetic integration of different strategic foci of Phase V designation areas did not come to full fruition. We recommend better and more sustained integration of realist synthesis in the practice of Healthy Cities in future Phase

    Sexually Dimorphic Serotonergic Dysfunction in a Mouse Model of Huntington's Disease and Depression

    Depression is the most common psychiatric disorder in Huntington's disease (HD) patients. In the general population, women are more prone to develop depression and such susceptibility might be related to serotonergic dysregulation. There is yet to be a study of sexual dimorphism in the development and presentation of depression in HD patients. We investigated whether 8-week-old male and female R6/1 transgenic HD mice display depressive-like endophenotypes associated with serotonergic impairments. We also studied the behavioral effects of acute treatment with sertraline. We found that only female HD mice exhibited a decreased preference for saccharin as well as impaired emotionality-related behaviors when assessed on the novelty-suppressed feeding test (NSFT) and the forced-swimming test (FST). The exaggerated immobility time displayed by female HD in the FST was reduced by acute administration of sertraline. We also report an increased response to the 5-HT1A receptor agonist 8-OH-DPAT in inducing hypothermia and a decreased 5-HT2A receptor function in HD animals. While tissue levels of serotonin were reduced in both male and female HD mice, we found that serotonin concentration and hydroxylase-2 (TPH2) mRNA levels were higher in the hippocampus of males compared to female animals. Finally, the antidepressant-like effects of sertraline in the FST were blunted in male HD animals. This study reveals sex-specific depressive-related behaviors during an early stage of HD prior to any cognitive and motor deficits. Our data suggest a crucial role for disrupted serotonin signaling in mediating the sexually dimorphic depression-like phenotype in HD mice

    A large-scale genome-wide association study meta-analysis of cannabis use disorder

    Summary Background Variation in liability to cannabis use disorder has a strong genetic component (estimated twin and family heritability about 50–70%) and is associated with negative outcomes, including increased risk of psychopathology. The aim of the study was to conduct a large genome-wide association study (GWAS) to identify novel genetic variants associated with cannabis use disorder. Methods To conduct this GWAS meta-analysis of cannabis use disorder and identify associations with genetic loci, we used samples from the Psychiatric Genomics Consortium Substance Use Disorders working group, iPSYCH, and deCODE (20 916 case samples, 363 116 control samples in total), contrasting cannabis use disorder cases with controls. To examine the genetic overlap between cannabis use disorder and 22 traits of interest (chosen because of previously published phenotypic correlations [eg, psychiatric disorders] or hypothesised associations [eg, chronotype] with cannabis use disorder), we used linkage disequilibrium score regression to calculate genetic correlations. Findings We identified two genome-wide significant loci: a novel chromosome 7 locus (FOXP2, lead single-nucleotide polymorphism [SNP] rs7783012; odds ratio [OR] 1·11, 95% CI 1·07–1·15, p=1·84 × 10−9) and the previously identified chromosome 8 locus (near CHRNA2 and EPHX2, lead SNP rs4732724; OR 0·89, 95% CI 0·86–0·93, p=6·46 × 10−9). Cannabis use disorder and cannabis use were genetically correlated (rg 0·50, p=1·50 × 10−21), but they showed significantly different genetic correlations with 12 of the 22 traits we tested, suggesting at least partially different genetic underpinnings of cannabis use and cannabis use disorder. Cannabis use disorder was positively genetically correlated with other psychopathology, including ADHD, major depression, and schizophrenia. Interpretation These findings support the theory that cannabis use disorder has shared genetic liability with other psychopathology, and there is a distinction between genetic liability to cannabis use and cannabis use disorder.
