    Environmental Conditions along Tuna Larval Dispersion: Insights on the Spawning Habitat and Impact on Their Development Stages

    Estimated larval backward trajectories of three Tuna species, namely, Atlantic Bluefin Tuna (Thunnus thynnus, Linnaeus, 1758), Bullet Tuna (Auxis Rochei, Risso, 1801) and Albacore Tuna (Thunnus alalunga, Bonnaterre, 1788) in the central Mediterranean Sea, were used to characterize their spawning habitats, and to assess the impact of changes due to the major environmental parameters (i.e., sea surface temperature and chlorophyll-a concentration) on larval development during their advection by surface currents. We assumed that the environmental variability experienced by larvae along their paths may have influenced their development, also affecting their survival. Our results showed that the Tuna larvae underwent an accelerated growth in favorable environmental conditions, impacting on the notochord development. In addition, further updated information on spawning and larval retention habitats of Atlantic Bluefin Tuna, Bullet and Albacore Tunas in the central Mediterranean Sea were delivered

    DNA Barcoding as a tool for Zoological Taxonomy: Identification of bony fish in the Mediterranean Sea

    The description of all the species present in nature is a vast task to be fulfilled by using the classical approach of morphological description of the organisms. In recent years, the traditional taxonomy, based primarily on identification keys of species, has shown a number of limitations in the use of the distinctive features in many animal taxa and inconsistencies with the genetic data. Furthermore, the increasing need to get a true estimate of biodiversity has led Zoological Taxonomy to seek new approaches and methodologies to support the traditional methods. The classification procedure has added modern criteriasuch as the evolutionary relationships and the genetic, biochemical and morphological characteristics of the organisms.Until now the Linnean binomial was the only abbreviated code associated with the description of the morphology of a species. The new technologies aim to achieve a short nucleotide sequence of the DNA to be used as an unique and solely label for a particular species, a specific genetic barcode. For both morphological and genetic approaches, skills and experience are required. Taxonomy is one of zoological disciplines that has been benefited from the achievements reached by modern molecular biotechnology. Using a molecular approach it is possible to identify cryptic species, to establish a family relationship between species and their membership of taxonomic categories or to reconstruct the evolutionary history of a taxon

    Biochemical and neuropathological findings in a Creutzfeldt-Jakob Disease patient with the rare Val180Ile-129Val haplotype in the prion protein gene

    Genetic Creutzfeldt-Jakob disease (gCJD) associated with the V180I mutation in the prion protein (PrP) gene (PRNP) in phase with residue 129M is the most frequent cause of gCJD in East Asia, whereas it is quite uncommon in Caucasians. We report on a gCJD patient with the rare V180I-129V haplotype, showing an unusually long duration of the disease and a characteristic pathological PrP (PrPSc) glycotype. Family members carrying the mutation were fully asymptomatic, as commonly observed with this mutation. Neuropathological examination showed a lesion pattern corresponding to that commonly reported in Japanese V180I cases with vacuolization and gliosis of the cerebral cortexes, olfactory areas, hippocampus and amygdala. PrP was deposited with a punctate, synaptic-like pattern in the cerebral cortex, amygdala and olfactory tract. Western blot analyses of proteinase-K-resistant PrP showed the characteristic two-banding pattern of V180I gCJD, composed of mono- and un-glycosylated isoforms. In line with reports on other V180I cases in the literature, Real-Time Quaking Induced Conversion (RT-QuIC) analyses did not demonstrate the presence of seeding activity in the cerebrospinal fluid and olfactory mucosa, suggesting that this haplotype also may result in a reduced seeding efficiency of the pathological PrP. Further studies are required to understand the origin, penetrance, disease phenotype and transmissibility of 180I-129V haplotype in Caucasians

    Valutazione dello stato della biodiversità in aree antropizzate: applicazione della metodologia BRUV (Baited Remote Underwater Video) per lo studio della componente nectonica dell'ecosistema

    Uno studio di valutazione dello stato della biodiversità focalizzato sulla componente nectonica e finalizzato alla determinazione dell’abbondanza e della composizione in specie del popolamento marino costiero si può avvalere di diversi strumenti e metodologie, le quali possono essere più o meno invasive sull’ecosistema oggetto dello studio. È possibile ad esempio applicare delle metodologie di pesca sperimentale, i.e. di prelievo degli organismi dal loro ambiente al fine di riconoscere e quantificare la presenza delle specie nectoniche nell’area di studio. Esistono tuttavia metodologie meno distruttive che permettono la raccolta di dati attraverso una stima visiva dell’abbondanza e della composizione delle specie direttamente nel loro ambiente naturale. Di queste metodologie fanno parte i censimenti visivi in situ (Samoilys M.A. 1997), i quali hanno il vantaggio di permettere la raccolta dati nel rispetto della biodiversità, evitando eventi mortali e/o fortemente stressanti agli individui oggetto dello studio. Questo fa si che tali metodologie risultino particolarmente utili in ambienti vulnerabili e/o soggetti a particolari vincoli protezionistici volti alla conservazione dell’ambiente marino, come le Aree Marine Protette (AMP), le quali in questo studio rappresentano delle zone di confronto caratterizzate da un limitato impatto antropico. Per tali ragioni, nell’ambito del progetto CISAS è stato scelto di portare avanti la valutazione dello stato del popolamento ittico attraverso una particolare tecnica di censimento visivo, la quale viene effettuata attraverso il sistema BRUV (Baited Remote Underwater Video)

    Coupling Lagrangian simulation models and remote sensing to explore the environmental effect on larval growth rate: The Mediterranean case study of round sardinella (Sardinella aurita) early life stages

    The relationship between environmental conditions and early life-history traits of Sardinella aurita are investigated using material collected in two sites of the Central Mediterranean Sea. Individual mean daily growth during the planktonic phase has been determined by using otolith microstructure analysis, while Lagrangian simulation models allowed to estimate the daily position in space and time of each specimen from the hatching to the catch. Generalized Additive Mixed Models (GAMMs) have been implemented to explore the impact of environmental conditions at time t, t-1 day and t-2 days on the mean daily growth rate occurring at time t. Spatial analysis evidenced a wide dispersion of eggs and larvae in the coastal area of both sampling sites in correspondence to relatively warmer and chlorophyll-a enriched waters. Lagrangian simulations detected a complementary larval dispersal pathway able to transport larvae to a known retention area. Temperature at time t was the most important driver affecting the mean daily larval growth, followed by the food availability. On the other hand, models performed on lagged environmental covariates (t-1 and t-2) did not show any significant effect on the growth rate at time t. In addition to the sub-linear positive correlation between temperature and mean daily larval growth, model highlighted a decrease in the otolith core width at higher temperature that can be linked to an earlier stage of ontogeny at hatching. This study provided a useful methodological approach that takes advantage of available remote sensing data to perform ecological studies in support to fisheries management

    La convenzione oggetto del presente rapporto nasce nel contesto delle complesse attività di EduLab, gruppo di divulgazione scientifica che,seppur già attivo dal2008, nasce ufficialmente nel 2015e mette insieme ricercatori di varie discipline che condividono la passione di divulgare la scienza e i risultati da essa prodotti nei propri ambiti di competenza.La comunicazione scientifica, molto sentita e ben strutturata all’interno dell’IAS (ex. IAMC), ha portato a numerosi risultati e ad un numero ragguardevole di progetti e programmi, ed è un’attività ormai talmente ben consolidata da diventare organica all’istituto e alle sue strategie. I prodotti sviluppati sono tanti e anche i successi ottenuti, quali premi sulle attività di divulgazione, la partecipazione ad Expo 2015 ed a numerose edizioni in ambito di manifestazioni nazionali quali: Festival della Scienza, Esperienza insegna, Notte della scienza e Notte dei ricercatori, Illuminiamo la scienza. Grazie ad un curriculum unico e complesso, il gruppo è stato il driver della nascita dell’Osservatorio della Biodiversità della Regione Sicilia, oggi riferimento regionale per la didattica e la divulgazione scientifica sul tema della biodiversità, visitato ad oggi da più di 500 classi di studenti siciliani.     “How a good video can remove ‘barriers’ for a more inclusive science communication”

    There is a burgeoning body of evidence suggesting that technology can enhance learning, and multiple studies have shown that videos represent a very effective tool in science engagement and education. In fact, video media can transform the complexities of science and nature into something more tangible and tractable (Dabylchuk et al., 2018). Documentaries can hence be powerful tools for learning, to raise awareness for important topics, and adding a scientific approach makes it possible to deepen knowledge about the world. According to Dale’s cone of experience (Fig. 1), people memorize 10% of what they read, 20% of what they hear, 30% of what they see and 50% of what they hear and see (Wiman and Mierhenry, 1969). These statistics seems to convey a very clear message: blind or visually impaired people are penalised in the learning compared to sighted ones. However, our experience in the area of inclusive science communication has led us to wonder: are we sure that sight is a fundamental means for learning? Or is it possible that sight is a predominant sense over others, and therefore it can even mislead or limit learning? To answer this question, we held an ad-hoc educational laboratory with the users of the “IstitutodeiCiechiOpereRiunite I. Florio – F. ed A. Salamone” of Palermo, with which we have established a convention on 29/01/2019. Specifically, we involved 20 users with different degrees of visual impairment, in the projection of three science videos, selected from those realised by the EDUlab divulgation group over the years. At the end of the video projection, users were asked to express their opinions and their personal interpretations of scientific issues, allowing us to understand what e how many information have been received and, above all, if sighted usersare really advantaged compared to the visually impaired ones
