    Shuttle Planning for Link Closures in Urban Public Transport Networks

    Urban Public Transport systems must periodically close certain links for main- tenance, which can have significant effects on the service provided to passengers. In practice, the effects of closures are mitigated by replacing the link with a simple shuttle service. However, alternative shuttle services could reduce inconvenience at lower op- erating cost. This paper proposes a model to select shuttle lines and frequencies under budget constraints. A new formulation is proposed that allows a minimal frequency restriction on any line that is operated, and minimizes passenger inconvenience cost, including transfers and frequency-dependent waiting time. This model is applied to a shuttle design problem based on a real world case study of the MBTA network of Boston (USA). The results show that additional shuttle routes can reduce passenger delay in comparison to the standard industry practice, while also distributing delay more equally over passengers, at the same operating budget. The results are robust under different assumptions about passenger route choice behavior. Computational experiments show that the proposed formulation, coupled with a preprocessing step, can be solved faster than prior formulations

    Revising mtDNA haplotypes of the ancient Hungarian conquerors with next generation sequencing

    As part of the effort to create a high resolution representative sequence database of the medieval Hungarian conquerors we have resequenced the entire mtDNA genome of 24 published ancient samples with Next Generation Sequencing, whose haplotypes had been previously determined with traditional PCR based methods. We show that PCR based methods are prone to erroneous haplotype or haplogroup determination due to ambiguous sequence reads, and many of the resequenced samples had been classified inaccurately. The SNaPshot method applied with published ancient DNA authenticity criteria is the most straightforward and cheapest PCR based approach for testing a large number of coding region SNP-s, which greatly facilitates correct haplogroup determination

    Heterologous Expression of Alteromonas macleodii and Thiocapsa roseopersicina [NiFe] Hydrogenases in Synechococcus elongatus

    Oxygen-tolerant [NiFe] hydrogenases may be used in future photobiological hydrogen production systems once the enzymes can be heterologously expressed in host organisms of interest. To achieve heterologous expression of [NiFe] hydrogenases in cyanobacteria, the two hydrogenase structural genes from Alteromonas macleodii Deep ecotype (AltDE), hynS and hynL, along with the surrounding genes in the gene operon of HynSL were cloned in a vector with an IPTG-inducible promoter and introduced into Synechococcus elongatus PCC7942. The hydrogenase protein was expressed at the correct size upon induction with IPTG. The heterologously-expressed HynSL hydrogenase was active when tested by in vitro H2 evolution assay, indicating the correct assembly of the catalytic center in the cyanobacterial host. Using a similar expression system, the hydrogenase structural genes from Thiocapsa roseopersicina (hynSL) and the entire set of known accessory genes were transferred to S. elongatus. A protein of the correct size was expressed but had no activity. However, when the 11 accessory genes from AltDE were co-expressed with hynSL, the T. roseopersicina hydrogenase was found to be active by in vitro assay.
