    The conservation and uniqueness of the caspase family in the basal chordate, amphioxus

    <p>Abstract</p> <p>Background</p> <p>The caspase family, which plays a central role in apoptosis in metazoans, has undergone an expansion in amphioxus, increasing to 45 members through domain recombination and shuffling.</p> <p>Results</p> <p>In order to shed light on the conservation and uniqueness of this family in amphioxus, we cloned three representative caspase genes, designated as <it>bbtCaspase-8, bbtCaspase-1/2 </it>and <it>bbtCaspase3</it>-like, from the amphioxus <it>Branchiostoma belcheri tsingtauense</it>. We found that <it>bbtCaspase-8 </it>with conserved protein architecture is involved in the Fas-associated death domain-Caspase-8 mediated pro-apoptotic extrinsic pathway, while <it>bbtCaspase3</it>-like may mediate a nuclear apoptotic pathway in amphioxus. Also, <it>bbtCaspase-1/2 </it>can co-localize with <it>bbtFADD2 </it>in the nucleus, and be recruited to the cytoplasm by amphioxus apoptosis associated speck-like proteins containing a caspase recruitment domain, indicating that <it>bbtCaspase-1/2 </it>may serve as a switch between apoptosis and caspase-dependent innate immune response in invertebrates. Finally, amphioxus extrinsic apoptotic pathway related caspases played important roles in early embryogenesis.</p> <p>Conclusions</p> <p>Our study not only demonstrates the conservation of <it>bbtCaspase-8 </it>in apoptosis, but also reveals the unique features of several amphioxus caspases with novel domain architectures arose some 500 million years ago.</p

    Visible Light Enhanced Extracellular Electron Transfer between a Hematite Photoanode and Pseudomonas aeruginosa

    Exploring the interplay between sunlight, semiconducting minerals, and microorganisms in nature has attracted great attention in recent years. Here we report for the first time the investigation of the interaction between a hematite photoelectrode and Pseudomonas aeruginosa PAO1 under visible light irradiation. Hematite is the most abundant mineral on earth, with a band gap of 2.0 eV. A hematite electrode was electrochemically deposited on fluorine-doped tin oxide (FTO). It was thoroughly characterized by environmental scanning electron microscopy (ESEM), Raman, and UV–Vis spectroscopy, and its prompt response to visible light was determined by linear sweep voltammetry (LSV). Notably, under light illumination, the hematite electrode immersed in a live cell culture was able to produce 240% more photocurrent density than that in the abiotic control of the medium, suggesting a photoenhanced extracellular electron transfer process occurring between hematite and PAO1. Different temperatures of LSV measurements showed bioelectrochemical activity in the system. Furthermore, I–t curves under various conditions demonstrated that both a direct and an indirect electron transferring process occurred between the hematite photoanode and PAO1. Moreover, the indirect electron transferring route was more dominant, which may be mainly attributed to the pyocyanin biosynthesized by PAO1. Our results have expanded our understanding in that in addition to Geobacter and Shewanella it has been shown that more microorganisms are able to perform enhanced extracellular electron transfer with semiconducting minerals under sunlight in nature

    Increased Serum Concentrations of TNF-Like Weak Inducer of Apoptosis Predict Higher 28-Day Mortality in Patients with Sepsis

    We performed the current study to explore potential predictive value of serum Tumor Necrosis Factor- (TNF-) like weak inducer of apoptosis (TWEAK) concentrations for 28-day mortality in patients with sepsis. Adult septic patients (age≥18 years) admitted to a general ICU between November 2016 and October 2017 were consecutively included in our prospective observational study. TWEAK concentrations were detected in septic patients and healthy controls. Dynamic changes of TWEAK concentrations between 1st day and 3rd day of admission to ICU (ΔTWEAK concentrations) were also measured. A total of 79 septic patients were included and 19 of them (24.1%) died after a follow-up period of 28 days. We identified arterial lactate, NT-proBNP, and male gender as independent factors for 28-day mortality of patients with sepsis. The serum levels of TWEAK were significantly lower in septic patients compared to controls (417.4 ± 196.7 pg/ml versus 1243.8 ± 174.3 pg/ml, p<0.001). We found a positive correlation between TWEAK concentrations and SOFA score (Spearman correlation coefficient 0.235, p=0.037). Area under the receiver operating characteristic curve (AUROC) of ΔTWEAK concentrations for 28-day mortality was 0.754 (95% CI 0.645–0.844). We also evaluated the diagnostic performance of combinative index (ΔTWEAK concentrations and lactate) and obtained an AUROC of 0.860 (95% CI 0.763-0.928). In conclusion, our study found lower TWEAK concentrations in septic patients than those in healthy controls. Furthermore, the increased TWEAK concentrations during disease process predict higher 28-day mortality in septic patients. Dynamic changes of TWEAK should be an important supplement for current prognostic markers

    据统计,2020年我国芝麻进口量达到历史高位,为101.61万 t,全国芝麻消费量保持增长态势,但在市场对芝麻的需求量扩大、市场前景广阔的背景下,广西芝麻的种植面积不增反降。为发挥广西芝麻种植优势,实现广西芝麻产业的高质量发展,详细论述了广西芝麻产业发展现状,分析了广西芝麻产业发展中存在的问题,并提出了发展对策。目前广西芝麻产业种植规模趋于稳定、种植品种类型多样、种植效益稳中有升、龙头企业引领市场,但仍然存在高产稳产品种不足,机械化生产能力薄弱,加工技术发展缓慢,产品销售渠道狭窄等问题。可通过加强良种技术攻关,提升器械应用水平,提高加工技术投入,拓宽产业市场渠道等对策,推动广西芝麻产业健康发展。 According to statistics,the import volume of sesame in China reached a historical high in 2020, with 1 016 100 t, and the national sesame consumption maintained a growth trend, but in the context of expanding market demand for sesame seeds and broad market prospects, the planting area of sesame in Guangxi decreased rather than increased. In order to give full play to the advantages of Guangxi′ sesame planting and realize the high-quality development of Guangxi′ sesame industry, the development status of Guangxi′ sesame industry was discussed in detail, the problems existing in the development of Guangxi′ sesame industry were analyzed, and countermeasures for development of Guangxi′ sesame industry were proposed. At present, the planting scale of Guangxi′ sesame industry tends to be stable, the types of planting varieties are diversified, the planting benefits are steadily rising, and leading enterprises lead the market, but there are still problems such as insufficient high-yielding and stable-yielding varieties, weak mechanized production capacity, slow development of processing technology, and narrow product sales channels. The healthy development of Guangxi′ sesame industry can be promoted by strengthening the technical research of improved variery, improving the application level of instruments,improving the investment in processing technology and broadening the market channels of the industry

    Study of Zuojin Pill in Treating Chronic Atrophic Gastritis by UPLC-Q-TOF/MS Based on Serum and Urine Metabolomics Combined with Network Pharmacology

    Zuojin Pill (ZJP) is widely used for the treatment of gastrointestinal diseases, while its specific mechanism has not been systematically investigated. The aim of this study was to explore the therapeutic effects and potential mechanism of ZJP in chronic atrophic gastritis (CAG) through UPLC-Q-TOF/MS-based metabolomics combined with network pharmacology. ZJP and omeprazole significantly reduce contents of IL-1β, IL-6, IL-10, and iNOS and improve pathological characteristics. Metabolomic results indicated that the therapeutic effects of ZJP were mainly related to ten metabolites, namely, choline, L-threonine, hydroxypyruvic acid, creatine, taurine, succinic acid, cis-aconitic acid, citric acid, succinic acid semialdehyde, and uric acid. Pathway analysis showed that the treatment of CAG by ZJP was associated with taurine and hypotaurine metabolism; glyoxylate and dicarboxylate metabolism; glycine, serine, and threonine metabolism; glycerophospholipid metabolism; citrate cycle (TCA cycle), alanine, aspartate, and glutamate metabolism; butanoate metabolism; and purine metabolism. Validation of metabolic markers and key targets of network pharmacology through RT-PCR analysis showed that ZJP significantly downregulated a series of inflammatory markers, such as MAPK1, PKIA, RB1, SCN5A, RXRA, E2F1, PTGS1, IGF2, ADRB1, ADRA1B, PTGS2, and GABRA1. This study was the first to use a combination of metabolomics and network pharmacology to clarify the therapeutic effects of ZJP on CAG and the regulation of multiple metabolic pathways
