    Porfyriny jsou organické látky důležité pro život. Mezi nejdůležitější zástupce patří chlorofyl a hem. Chlorofyl je zelené barvivo, které se vyskytuje především u rostlin, hem naproti tomu nenacházíme u rostlin, ale u živočichů. Jeho syntéza je přirozený biologický proces, který probíhá způsobem, který jsem se pokusila ve své práci popsat. Dále jsem se pak zaměřila na poruchy této biosyntézy a také onemocnění, která tím vznikají - porfyrie. Nechybí zde ani jejich charakteristika a v některých případech i léčba, která je prozatím pouze symptomatická.Porphyrins are organic substances important for life. The most important representatives include chlorophyll and heme. Chlorophyll is the green pigment that is found primarily in plants, on the other hand heme we do not find in plants, but in animals. Its synthesis is a natural biological process that takes place in a way that I tried to describe in my work. I have also paid attention to the failure of the biosynthesis and diseases that arises - porphyria.     The subject of this diploma thesis was to identify changes in a visit rate of spas in Czech Republic that have been caused mainly after adoption of a new Indication list in 2012. Its impacts were examined by comparison and analysis between two time periods, by year 2013 and from year 2013. Essential indicators of spas, such as trend of treatment days, numbers of clients, accommodation capacity and staffing, were examined in more detail in the diploma thesis. Based on the results and findings were then estimated further trends and development of spas in Czech Republic

    Candida albicans a Pseudomonas aeruginosa patří mezi oportunistické patogeny, které mimo jiné způsobují řadu onemocnění u imunokompromitovaných pacientů. C. albicans je dimorfická houba, která je schopna reverzibilně měnit svou morfologickou formu z kvasinkové na vláknitou a zvyšovat tak svou patogenitu. Mimoto je schopná v lidském hostiteli interagovat s bakteriemi, např. s P. aeruginosa a ovlivňovat tak zdraví jedince. P. aeruginosa je gramnegativní bakterie, která bývá často spojována s infekcemi respiračního systému, nejčastěji u pacientů s cystickou fibrózou. Tyto dva mikroorganismy spolu v kokultuře vzájemně komunikují na molekulární úrovni a ovlivňují tak své chování. V této práci se snažíme zjistit, zda kokultivace C. albicans a P. aeruginosa ovlivní senzitivitu C. albicans vůči azolovým antimykotikům.Candida albicans and Pseudomonas aeruginosa belong to the oportunistic pathogens, which cause many diseases in immunocompromised pacients. C. albicans is the dimorpic yeast, which can reversibly change its morphological form (yeast hyphae pseudohyphae) and thus increase its pathogenity. Furthmore, in human hosts C. albicans is able to interact with bacteria, .e.g. with P. aeruginosa, and affect the health of individuals. P. aeruginosa is a common Gram-negative bacterium, which is often associated with infections of respiratory tract, most often in patients with cystic fibrosis. In coculture these two microorganisms communicate at the molecular level and affect the behavior of each other.     Development of a spectrofluorometric method for detection of nuclear condensation and fragmentation in cells

    Disertační práce se zaměřuje na popis apoptózy a na přehled bioanalytických metod, které se využívají při její detekci a hodnocení. V rámci disertační práce jsme s využitím sondy Hoechst 33258 vyvinuli spektrofluorimetrickou metodu pro detekci nukleární kondenzace a fragmentace. Pomocí této metody jsme popsali vliv modelových toxinů na proces nukleární kondenzace in vitro. Následně jsme výsledky získané novou metodou porovnali s výstupy dalších metod určených k detekci apoptózy, tj. detekce aktivity kaspáz, JNK a PARP aktivace, a metody TUNEL a DNA žebřík. V závěrečné části práce jsme naměřené výsledky shrnuli, diskutovali a porovnali s literaturou. Nově vyvinutá metoda využívající Hoechst 33258 je plně porovnatelná s metodou TUNEL a má navíc řadu výhod (např. rychlost, kvantifikace, nízká cena). Další velkou výhodou je možnost analyzovat velké množství vzorků najednou.This doctoral thesis focuses on explaining the process of apoptosis and on an overview and description of bioanalytical methods used in its evaluation and detection. In the experimental part of this thesis, we developed the spectrofluorometric method Hoechst 33258 for the detection of nuclear condensation with subsequent evaluation. Using this method, we detected the effect of selected toxins on the process of nuclear condensation in vitro. Subsequently, we compared the results obtained by the new method with the outputs of other methods designed to detect apoptosis, ie detection of caspase activity, JNK and PARP activation, and TUNEL and DNA ladder methods. In the final part of the work, we discussed the results and compared them with the literature. Newly developed method using the Hoechst 33258 probe is fully comparable with TUNEL method. Moreover, our method has a number of advantages (e.g. speed, quantification, low cost) over the TUNEL assay.     This thesis deals with performance evaluation of a selected company using the START model and financial analysis ratios. The content of the thesis is conceived in three main parts – theoretical background, analysis of the problem and the current situation, own proposals for solutions and the contribution of the proposed solutions. The aim of this thesis is to find strengths and opportunities for improvement by applying the START model and analysing the financial situation of the selected company. The result of this thesis is development of suggestions for the company's problematic areas that emerged from the methods and analyses used to help improve its performance
