    Clark\u27s Nutcracker Seed Harvest Patterns in Glacier National Park and a Novel Method for Monitoring Whitebark Pine Cones

    Clark\u27s Nutcracker (Nucifraga columbiana) is the primary seed disperser of whitebark pine (Pinus albicaulis), which is in decline throughout its range. There is concern that a decline in whitebark pine will lead to a subsequent decline in local populations of Clark\u27s Nutcracker. Because natural regeneration depends on the presence of Clark\u27s Nutcracker, the process of harvesting whitebark pine seeds needs to be fully understood. In addition, resource managers need a cost-effective method for monitoring nutcracker occurrence in whitebark pine stands during the seed harvest season. I visited eleven study sites in Glacier National Park, Montana, where I searched for Clark\u27s Nutcracker and surveyed whitebark pine cones for seed harvesting scars, the presence of which indicated that nutcrackers harvested seeds. I documented cone use patterns of Clark\u27s Nutcracker and the major cone predator, red squirrel (Tamiasciurus hudsonicus), at five sites. To identify factors that influence cone use, I ran a correlation analysis with nutcracker and red squirrel seed harvesting variables with physical, compositional, and whitebark pine-related factors. I found that nutcrackers harvested seed at every site that had cones available. Nutcrackers harvested seed from a greater proportion of whitebark pine cones in stands where they started intensively harvesting seeds earlier. Nutcrackers began intensively harvesting seeds earlier in stands with higher relative dominance of whitebark pine. Red squirrels depleted the cone source more rapidly in stands with greater whitebark pine mortality, and at one site depleted the cone source completely before nutcrackers began intensively harvesting seeds from that site. The results of this study suggest that Clark\u27s Nutcracker will continue to harvest seeds even as whitebark pine declines, but the decline in whitebark pine may lead to decreased seed dispersal due to greater pre-dispersal cone predation by red squirrels. Finally, I evaluated direct and indirect monitoring methods to identify a cost-effective method to accurately monitor Clark\u27s Nutcracker occurrence in whitebark pine stands during the seed harvest season. I found that surveying scars made by seed-harvesting nutcrackers on whitebark pine cones was the most accurate and economical method of monitoring Clark\u27s Nutcracker occurrence in an area with a low population of Clark\u27s Nutcracker

    Efficient stray-light suppression for resonance fluorescence in quantum dot-micropillars using self-aligned metal apertures

    Within this work we propose and demonstrate a technological approach to efficiently suppress excitation laser stray-light in resonance fluorescence experiments on quantum dot-micropillars. To ensure efficient stray-light suppression, their fabrication process includes a planarization step and the subsequent covering with a titanium mask to fabricate self-aligned apertures at the micropillar positions. These apertures aim at limiting laser straylight in side-excitation vertical-detection configuration, while enabling detection of the optical signal through the top facet of the micropillars. Beneficial effects of these apertures are proven and quantitatively evaluated within a statistical study in which we determine and compare the stray-light suppression of 48 micropillars with and without metal apertures. Actual resonance fluorescence experiments on single quantum dots coupled to the cavity mode prove the relevance of the proposed approach and demonstrate that it will foster further studies on cavity quantum electrodynamics phenomena under coherent optical excitation.PostprintPeer reviewe

    Beyond germline genetic testing - heterozygous pathogenic variants in PMS2 in two children with osteosarcoma and ependymoma

    Background Lynch syndrome (LS) is not considered part of childhood cancer predisposition syndromes. Case presentation Analysis of a pediatric osteosarcoma (OS) displayed hypermutation (16.8), alternative lengthening of telomeres (ALT), loss of PMS2 expression in tumor tissue (retained in non-neoplastic cells), PMS2 loss of heterozygosity (LOH), and high-degree of microsatellite instability (MSI) tested by PCR. A heterozygous duplication c.1076dup p.(Leu359Phefs*6) in exon 10 of NM_000535.6:PMS2 was detected by SNV analysis in peripheral blood, confirming diagnosis of LS in the patient. The tumor molecular features suggest LS-associated development of OS. In a second case, whole-genome sequencing identified a heterozygous SNV c.1 A > T p.? in exon 1 of PMS2 in tumor and germline material of a girl with ependymoma. Tumor analysis displayed evidence for ALT and low mutational burden (0.6), PMS2 expression was retained, MSI was low. Multiplex ligation-dependent probe amplification identified no additional PMS2 variant and germline MSI testing did not reveal increased gMSI ratios in the patient´s lymphocytes. Thus, CMMRD was most closely excluded and our data do not suggest that ependymoma was related to LS in the child. Conclusions Our data suggest that the LS cancer spectrum may include childhood cancer. The importance of LS in pediatric cancers necessitates prospective data collection. Comprehensive molecular workup of tumor samples is necessary to explore the causal role of germline genetic variants

    High resolution MRI for quantitative assessment of inferior alveolar nerve impairment in course of mandible fractures: an imaging feasibility study

    The purpose of this study was to evaluate a magnetic resonance imaging (MRI) protocol for direct visualization of the inferior alveolar nerve in the setting of mandibular fractures. Fifteen patients suffering from unilateral mandible fractures involving the inferior alveolar nerve (15 affected IAN and 15 unaffected IAN from contralateral side) were examined on a 3 T scanner (Elition, Philips Healthcare, Best, the Netherlands) and compared with 15 healthy volunteers (30 IAN in total). The sequence protocol consisted of a 3D STIR, 3D DESS and 3D T1 FFE sequence. Apparent nerve-muscle contrast-to-noise ratio (aNMCNR), apparent signal-to-noise ratio (aSNR), nerve diameter and fracture dislocation were evaluated by two radiologists and correlated with nerve impairment. Furthermore, dislocation as depicted by MRI was compared to computed tomography (CT) images. Patients with clinically evident nerve impairment showed a significant increase of aNMCNR, aSNR and nerve diameter compared to healthy controls and to the contralateral side (p < 0.05). Furthermore, the T1 FFE sequence allowed dislocation depiction comparable to CT. This prospective study provides a rapid imaging protocol using the 3D STIR and 3D T1 FFE sequence that can directly assess both mandible fractures and IAN damage. In patients with hypoesthesia following mandibular fractures, increased aNMCN R, aSNR and nerve diameter on MRI imaging may help identify patients with a risk of prolonged or permanent hypoesthesia at an early time

    Introduction A growing body of evidence suggests that hearing loss is a significant and potentially modifiable risk factor for cognitive impairment. Although the mechanisms underlying the associations between cognitive decline and hearing loss are unclear, listening effort has been posited as one of the mechanisms involved with cognitive decline in older age. To date, there has been a lack of research investigating this association, particularly among adults with mild cognitive impairment (MCI).Methods and analysis 15–25 cognitively healthy participants and 15–25 patients with MCI (age 40–85 years) will be recruited to participate in an exploratory study investigating the association between cognitive functioning and listening effort. Both behavioural and objective measures of listening effort will be investigated. The sentence-final word identification and recall (SWIR) test will be administered with single talker non-intelligible speech background noise while monitoring pupil dilation. Evaluation of cognitive function will be carried out in a clinical setting using a battery of neuropsychological tests.
