    Biohydrogenation of 22:6n-3 by Butyrivibrio proteoclasticus P18

    Background: Rumen microbes metabolize 22:6n-3. However, pathways of 22:6n-3 biohydrogenation and ruminal microbes involved in this process are not known. In this study, we examine the ability of the well-known rumen biohydrogenating bacteria, Butyrivibrio fibrisolvens D1 and Butyrivibrio proteoclasticus P18, to hydrogenate 22:6n-3. Results: Butyrivibrio fibrisolvens D1 failed to hydrogenate 22:6n-3 (0.5 to 32 mu g/mL) in growth medium containing autoclaved ruminal fluid that either had or had not been centrifuged. Growth of B. fibrisolvens was delayed at the higher 22:6n-3 concentrations; however, total volatile fatty acid production was not affected. Butyrivibrio proteoclasticus P18 hydrogenated 22:6n-3 in growth medium containing autoclaved ruminal fluid that either had or had not been centrifuged. Biohydrogenation only started when volatile fatty acid production or growth of B. proteoclasticus P18 had been initiated, which might suggest that growth or metabolic activity is a prerequisite for the metabolism of 22:6n-3. The amount of 22:6n-3 hydrogenated was quantitatively recovered in several intermediate products eluting on the gas chromatogram between 22:6n-3 and 22:0. Formation of neither 22:0 nor 22:6 conjugated fatty acids was observed during 22:6n-3 metabolism. Extensive metabolism was observed at lower initial concentrations of 22:6n-3 (5, 10 and 20 mu g/mL) whereas increasing concentrations of 22:6n-3 (40 and 80 mu g/mL) inhibited its metabolism. Stearic acid formation (18:0) from 18:2n-6 by B. proteoclasticus P18 was retarded, but not completely inhibited, in the presence of 22:6n-3 and this effect was dependent on 22:6n-3 concentration. Conclusions: For the first time, our study identified ruminal bacteria with the ability to hydrogenate 22:6n-3. The gradual appearance of intermediates indicates that biohydrogenation of 22:6n-3 by B. proteoclasticus P18 occurs by pathways of isomerization and hydrogenation resulting in a variety of unsaturated 22 carbon fatty acids. During the simultaneous presence of 18:2n-6 and 22:6n-3, B. proteoclasticus P18 initiated 22:6n-3 metabolism before converting 18:1 isomers into 18:0

It shows how to identify nursing diagnoses according to NANDA Taxonomy II in Post Anesthesia Care Unit (PACU). The sample consisted of 30 adult patients undergoing elective surgery with general anesthesia, and evaluation of physical conditions according to American Society of Anesthesiologists (ASA 1). The field of study was the PACU, which has 8 beds in a mixed network hospital in the city of Santos. Data were collected through a structured method, with data on gender, age, medical diagnoses, intervention-surgical anesthetic, surgical time, anesthetic time, vital signs including blood pressure at admission, assessment of the index of Aldrete KrouliK; and complications such as nausea, vomiting and pain. After the analysis of the nursing problems, nursing diagnoses were identified, with the greatest frequency being hypothermia, nausea, and acute pain.  Este es un estudio descriptivo, exploratorio, con método cualitativo y rasgo no empírico, con análisis de frecuencia absoluta y relativa de los datos. Presenta como objetivo identificar los diagnósticos de enfermería según la Taxonomía II de la NANDA, en la Sala de Recuperación Post-Anestésica (SRPA). La muestra estaba constituida por 30 enfermos adultos, sometidos a cirugía selectiva, con anestesia general y evaluación de las condiciones físicas Americam Society of Anesthesiologists (ASA1). El campo de estudio fue la SPRA, la cual cuenta con 8 camas de un hospital de la red mixta de la ciudad de Santos. Los datos fueron colectados a través de un instrumento estructurado, con datos referentes al sexo, edad, diagnóstico médico, intervención anestésico quirúrgica, tiempo quirúrgico, tiempo anestésico, signos vitales, incluyendo verificación de la tensión arterial en la internación, índices de Aldrete Kroulik y complicaciones como náusea, vómito y dolor. Tras el análisis de los problemas de enfermería, fueron identificados los diagnósticos de enfermería, siendo los de mayor frecuencia la hipotermia, náusea y dolor agudo.Este é um estudo descritivo, exploratório, com método quantitativo e delineamento não experimental, com análise de freqüência absoluta e relativa dos dados. Apresenta como objetivo identificar os diagnósticos de enfermagem, segundo a Taxonomia II da NANDA, na Sala de Recuperação Pós-Anestésica (SRPA). A amostra foi constituída de 30 pacientes adultos, submetidos à cirurgia eletiva, com anestesia geral, e avaliação das condições físicas Americam Society of Anesthesiologists (ASA 1). O campo de estudo foi a SRPA, a qual conta com 8 leitos, de um Hospital da rede mista da cidade de Santos. Os dados foram coletados por meio de um instrumento estruturado, com dados referentes ao sexo, idade, diagnóstico médico, intervenção anestésico-cirúrgica, tempo cirúrgico, tempo anestésico, sinais vitais incluindo verificação de pressão arterial na internação, avaliação do índice de Aldrete KrouliK; e complicações como náusea, vômito e dor.     Health-related quality of life in patients with venous leg ulcer treated in primary care in Brazil and Portugal

    Background Venous ulcers constitute an important public health problem as they can cause disability with consequences for multiple dimensions of quality of life. Objective To describe the quality of life in patients with venous leg ulcer treated in primary care in two cities from Brazil and Portugal. Methods This was a cross-sectional comparative study with a non-probabilistic sample of 171 patients with venous leg ulcers who were treated in primary care in two cities from Brazil and Portugal, namely, Natal and Évora. A form covering sociodemographic and health data and the Medical Outcomes Study 36-Item Short-Form Health Survey were used, and descriptive and inferential analyses were performed. Results Significant differences in age and income were observed between the two samples. Patients with venous leg ulcer from Brazil had lower income and were younger than those from Portugal. Quality of life scores were significantly higher in Portugal for the physical aspects, pain, and social functioning, among domains, and for the physical health dimension and total score of QOL. Conclusion The quality of life was better in Portugal than in Brazil and the differences between the countries need further investigation

    A Technique for Measuring Petal Gloss, with Examples from the Namaqualand Flora

    The degree of floral gloss varies between species. However, little is known about this distinctive floral trait, even though it could be a key feature of floral biotic and abiotic interactions. One reason for the absence of knowledge is the lack of a simple, repeatable method of gloss measurement that can be used in the field to study floral gloss. A protocol is described for measuring gloss in petal samples collected in the field, using a glossmeter. Repeatability of the technique is assessed. We demonstrate a simple yet highly accurate and repeatable method that can easily be implemented in the field. We also highlight the huge variety of glossiness found within flowers and between species in a sample of spring-blooming flowers collected in Namaqualand, South Africa. We discuss the potential uses of this method and its applications for furthering studies in plant-pollinator interactions. We also discuss the potential functions of gloss in flowers

    The Enzymatic Activity of Type 1 Iodothyronine Deiodinase (D1) is Low in Liver Hemangioma: A Preliminary Study

    Type 1 iodothyronine deiodinase (D1) is a crucial enzyme which converts the prohormone thyroxine (T4) into active tri-iodothyronine (T3). There has been strong evidence that the metabolism of thyroid hormones is disturbed in some neoplastic tissues such as thyroid, renal, and breast cancer. However, there are few available data about D1 enzyme activity in benign tumors such as hemangioma, which is the most common primary liver tumor. Hence this study aimed to determine the enzymatic activity of D1 in hemangiomas in relation to healthy liver tissue. Seven tumors and healthy control tissues were obtained from patients who had liver resection due to hemangioma. The activity was assessed by measurement of radioactive iodine released by deiodination catalyzed by D1. It was found that D1 activity was significantly lower in the hemagiomas than in the healthy surrounding tissue (p = 0.0017). The results indicated that thyroid hormones play important roles not only in the regulation of cell metabolism, but also in cell growth, division, and apoptosis. The active form T3 acts through its nuclear receptors and influences the up- and down-regulation of target genes. Healthy liver tissue expresses a high level of D1, but disturbed D1 activity may result in changes in the local concentration of T3 which may impair gene transcription. These finding demonstrate a low enzymatic activity of D1 in liver hemangioma and suggest an as yet unknown role of thyroid hormones in this type of benign liver tumor

    Chemodynamics of the Milky Way. I. The first year of APOGEE data

    We investigate the chemo-kinematic properties of the Milky Way disc by exploring the first year of data from the Apache Point Observatory Galactic Evolution Experiment (APOGEE), and compare our results to smaller optical high-resolution samples in the literature, as well as results from lower resolution surveys such as GCS, SEGUE and RAVE. We start by selecting a high-quality sample in terms of chemistry (____sim 20.000 stars) and, after computing distances and orbital parameters for this sample, we employ a number of useful subsets to formulate constraints on Galactic chemical and chemodynamical evolution processes in the Solar neighbourhood and beyond (e.g., metallicity distributions -- MDFs, [____alpha/Fe] vs. [Fe/H] diagrams, and abundance gradients). Our red giant sample spans distances as large as 10 kpc from the Sun. We find remarkable agreement between the recently published local (d << 100 pc) high-resolution high-S/N HARPS sample and our local HQ sample (d << 1 kpc). The local MDF peaks slightly below solar metallicity, and exhibits an extended tail towards [Fe/H] == -1, whereas a sharper cut-off is seen at larger metallicities. The APOGEE data also confirm the existence of a gap in the [____alpha/Fe] vs. [Fe/H] abundance diagram. When expanding our sample to cover three different Galactocentric distance bins, we find the high-[____alpha/Fe] stars to be rare towards the outer zones, as previously suggested in the literature. For the gradients in [Fe/H] and [____alpha/Fe], measured over a range of 6 < < R < < 11 kpc in Galactocentric distance, we find a good agreement with the gradients traced by the GCS and RAVE dwarf samples. For stars with 1.5 << z << 3 kpc, we find a positive metallicity gradient and a negative gradient in [____alpha/Fe]

    High-Throughput Analysis of Synthetic Peptides for the Immunodiagnosis of Canine Visceral Leishmaniasis

    Globally, the number of new human cases of visceral leishmaniasis (VL) is estimated to be approximately 500,000 per year. This is the most severe of all forms of leishmaniasis, and the zoonotic form of VL, caused by Leishmania infantum (also known as Leishmania chagasi), represents 20% of human visceral leishmaniasis worldwide; additionally, its prevalence is increasing in urban and peri-urban areas of the tropics. In Brazil, the identification and elimination of infected dogs, which act as a reservoir for Leishmania parasites, is a control measure employed in addition to the use of insecticides against the vectors and the identification and treatment of infected humans. Currently, the diagnostic methods employed to identify infected animals are not able to detect all of these dogs, which compromises the effectiveness of control measures. Moreover, one of the most important issues in controlling VL is the difficulty of diagnosing asymptomatic dogs, which act as parasite reservoirs. Therefore, to contribute to the improvement of the diagnostic methods for CVL, we aimed to identify and characterize new antigens that were more sensitive and specific and could be applied in epidemiologic surveys

    New Insight into the Antifibrotic Effects of Praziquantel on Mice in Infection with Schistosoma japonicum

    Schistosomiasis is a parasitic disease infecting more than 200 million people in the world. Although chemotherapy targeting on killing schistosomes is one of the main strategies in the disease control, there are few effective ways of dealing with liver fibrosis caused by the parasite infection in the chronic and advanced stages of schistosomiasis. For this reason, new strategies and prospective drugs, which exert antifibrotic effects, are urgently required.-induced liver fibrosis was inhibited by PZQ treatment for 30 days. Furthermore, we analyzed the effects of praziquantel on mouse primary hepatic stellate cells (HSCs). It is indicated that mRNA expressions of Col1α1, Col3α1, α-SMA, TGF-β, MMP9 and TIMP1 of HSCs were all inhibited after praziquantel anti-parasite treatments.The significant amelioration of hepatic fibrosis by praziquantel treatment validates it as a promising drug of anti-fibrosis and offers potential of a new chemotherapy for hepatic fibrosis resulting from schistosomiasis
