Portrait de l'utilisation des TIC par les collégiens ayant des troubles d'apprentissage, les bons lecteurs et les très faibles lecteurs
Les auteurs présentent les points de vue qu’adoptent des spécialistes du milieu collégial et des étudiants ayant des troubles d’apprentissage sur l’efficacité des technologies de l’information et de la communication. Cet article met en lumière les résultats de la seconde phase d’une étude menée par le Réseau de Recherche Adaptech, avec la collaboration du Collège Dawson, du Cégep André-Laurendeau et du Collège Montmorency qui porte sur cette question. Les étudiants ayant des TA semblent moins connaitre les TIC que les bons lecteurs, alors que les très faibles lecteurs se situent entre ces deux groupes. Le travail de sensibilisation à l’utilisation des TIC doit être entrepris ou poursuivi

Phase Ib study of the combination of pexidartinib (PLX3397), a CSF-1R inhibitor, and paclitaxel in patients with advanced solid tumors.
Purpose:To evaluate the safety, recommended phase II dose (RP2D) and efficacy of pexidartinib, a colony stimulating factor receptor 1 (CSF-1R) inhibitor, in combination with weekly paclitaxel in patients with advanced solid tumors. Patients and Methods:In part 1 of this phase Ib study, 24 patients with advanced solid tumors received escalating doses of pexidartinib with weekly paclitaxel (80 mg/m2). Pexidartinib was administered at 600 mg/day in cohort 1. For subsequent cohorts, the dose was increased by ⩽50% using a standard 3+3 design. In part 2, 30 patients with metastatic solid tumors were enrolled to examine safety, tolerability and efficacy of the RP2D. Pharmacokinetics and biomarkers were also assessed. Results:A total of 51 patients reported ≥1 adverse event(s) (AEs) that were at least possibly related to either study drug. Grade 3-4 AEs, including anemia (26%), neutropenia (22%), lymphopenia (19%), fatigue (15%), and hypertension (11%), were recorded in 38 patients (70%). In part 1, no maximum tolerated dose was achieved and 1600 mg/day was determined to be the RP2D. Of 38 patients evaluable for efficacy, 1 (3%) had complete response, 5 (13%) partial response, 13 (34%) stable disease, and 17 (45%) progressive disease. No drug-drug interactions were found. Plasma CSF-1 levels increased 1.6- to 53-fold, and CD14dim/CD16+ monocyte levels decreased by 57-100%. Conclusions:The combination of pexidartinib and paclitaxel was generally well tolerated. RP2D for pexidartinib was 1600 mg/day. Pexidartinib blocked CSF-1R signaling, indicating potential for mitigating macrophage tumor infiltration

Les auteurs communiquent enfin leurs recommandations pour que l’utilisation que l’on fait des TIC dans les collèges favorise la réussite scolaire
AXL modulates extracellular matrix protein expression and is essential for invasion and metastasis in endometrial cancer
The receptor tyrosine kinase AXL promotes migration, invasion, and metastasis. Here, we evaluated the role of AXL in endometrial cancer. High immunohistochemical expression of AXL was found in 76% (63/83) of advanced-stage, and 77% (82/107) of high-grade specimens and correlated with worse survival in uterine serous cancer patients. In vitro, genetic silencing of AXL inhibited migration and invasion but had no effect on proliferation of ARK1 endometrial cancer cells. AXL-deficient cells showed significantly decreased expression of phospho-AKT as well as uPA, MMP-1, MMP-2, MMP-3, and MMP-9. In a xenograft model of human uterine serous carcinoma with AXL-deficient ARK1 cells, there was significantly less tumor burden than xenografts with control ARK1 cells. Together, these findings underscore the therapeutic potentials of AXL as a candidate target for treatment of metastatic endometrial cancer
Comment bien utiliser les TIC au collégial : le point de vue des étudiants
Sachant que la motivation est au cœur de l’apprentissage, les auteures de cet article se sont intéressées aux technologies de l’information et de la communication (TIC) qui pouvaient augmenter la motivation des étudiants à s’impliquer dans leurs apprentissages et dans les activités proposées en classe. Les professeurs du collégial participent de plus en plus au dynamisme du courant TIC en enseignement supérieur, privilégiant l’utilisation des technologies en classe. D’ailleurs, plusieurs recherches se sont déjà penchées sur l’intégration des TIC à l’enseignement collégial, mais peu se sont attardées en profondeur à toute la complexité pédagogique qu’elle implique. Cette complexité découle du fait que la technologie n’est pas utilisée dans le vide : une intégration pédagogique réussie des TIC doit s’insérer dans un cadre respectant certains principes et où les activités pédagogiques impliquant les TIC s’articulent finement à des méthodes servant des objectifs explicites. Dans une étude, les auteures ont tenté de mieux comprendre comment favoriser les apprentissages ainsi que la participation des étudiants par l’utilisation des TIC en classe en s’attardant à la perception qu’ont ceux-ci des différentes technologies et de la manière dont elles sont mises à profit par leurs professeurs durant la session. L’article expose les grandes lignes de cette recherche en pistant les lecteurs sur les TIC que préfèrent les étudiants ou sur celles qui semblent avoir un effet positif sur leurs apprentissages, de leur point de vue. Le texte présente, par ailleurs, des problèmes fréquents associés aux technologies que les étudiants ont relevés et des stratégies qu’il est possible de mettre en place pour les contrer. Les chercheuses espèrent fournir aux professeurs quelques réflexions quant aux meilleures façons d’intégrer les TIC en classe, en fonction de la perspective étudiante

Standard Colonic Lavage Alters the Natural State of Mucosal-Associated Microbiota in the Human Colon
Past studies of the human intestinal microbiota are potentially confounded by the common practice of using bowel-cleansing preparations. We examined if colonic lavage changes the natural state of enteric mucosal-adherent microbes in healthy human subjects.Twelve healthy individuals were divided into three groups; experimental group, control group one, and control group two. Subjects in the experimental group underwent an un-prepped flexible sigmoidoscopy with biopsies. Within two weeks, subjects were given a standard polyethylene glycol-based bowel cleansing preparation followed by a second flexible sigmoidoscopy. Subjects in control group one underwent two un-prepped flexible sigmoidoscopies within one week. Subjects in the second control group underwent an un-prepped flexible sigmoidoscopy followed by a second flexible sigmoidoscopy after a 24-hour clear liquid diet within one week. The mucosa-associated microbial communities from the two procedures in each subject were compared using 16S rRNA gene based terminal restriction fragment length polymorphism (T-RFLP), and library cloning and sequencing.Clone library sequencing analysis showed that there were changes in the composition of the mucosa-associated microbiota in subjects after colonic lavage. These changes were not observed in our control groups. Standard bowel preparation altered the diversity of mucosa-associated microbiota. Taxonomic classification did not reveal significant changes at the phylum level, but there were differences observed at the genus level.Standard bowel cleansing preparation altered the mucosal-adherent microbiota in all of our subjects, although the degree of change was variable. These findings underscore the importance of considering the confounding effects of bowel preparation when designing experiments exploring the gut microbiota
Cytoplasmic p53 couples oncogene-driven glucose metabolism to apoptosis and is a therapeutic target in glioblastoma.
Cross-talk among oncogenic signaling and metabolic pathways may create opportunities for new therapeutic strategies in cancer. Here we show that although acute inhibition of EGFR-driven glucose metabolism induces only minimal cell death, it lowers the apoptotic threshold in a subset of patient-derived glioblastoma (GBM) cells. Mechanistic studies revealed that after attenuated glucose consumption, Bcl-xL blocks cytoplasmic p53 from triggering intrinsic apoptosis. Consequently, targeting of EGFR-driven glucose metabolism in combination with pharmacological stabilization of p53 with the brain-penetrant small molecule idasanutlin resulted in synthetic lethality in orthotopic glioblastoma xenograft models. Notably, neither the degree of EGFR-signaling inhibition nor genetic analysis of EGFR was sufficient to predict sensitivity to this therapeutic combination. However, detection of rapid inhibitory effects on [18F]fluorodeoxyglucose uptake, assessed through noninvasive positron emission tomography, was an effective predictive biomarker of response in vivo. Together, these studies identify a crucial link among oncogene signaling, glucose metabolism, and cytoplasmic p53, which may potentially be exploited for combination therapy in GBM and possibly other malignancies
