    Loading of Cells with Fluorescent Probe to Study Intracellular Acid-base Homeostasis in Lactic Acid Bacteria

    Here we describe a protocol which we have used to study the homeostasis intracellular in vivo in lactic acid bacteria (LAB) using a fluorescent probe. This type of probes can be used for determining changes in the pH of cytoplasm with high sensitivity, temporal resolution and technical simplicity as well as accessing the rate of change of intracellular pH in response to a stimulus from kinetic measurements on short time scales (Breeuwer et al., 1996; Molenaar et al., 1991).     Scalable model for industrial coffee roasting chamber

    Abstract The temperature profile of the coffee beans during the roasting phase determines the colour, aroma and flavour of the coffee. In order to reproduce these desired characteristics, the control of the coffee beans temperature has a key role in the roasting process. A proper model of the plant is required to design an intelligent control. Recently, several physical models that share the main physical equations have been proposed, but they have physical parameters specific of each process. In such scenario, each plant requires an ad hoc identification of the model parameters. This work proposes a model of the roasting chamber that can be used on plants of different sizes by scaling only geometrical parameters directly measurable on the roasting plant. The proposed model was identified on a 120 kg plant and then applied to a 360 kg one. The obtained results show in both cases similar accuracy (FIT = 75.49%, MPE=4.66%)

    Multivariate analysis of organic acids in fermented food from reversed-phase high-performance liquid chromatography data

    Multivariate calibration coupled to RP-HPLC with diode array detection (HPLC-DAD) was applied to the identification and the quantitative evaluation of the short chain organic acids (malic, oxalic, formic, lactic, acetic, citric, pyruvic, succinic, tartaric, propionic and α-cetoglutaric) in fermented food. The goal of the present study was to get the successful resolution of a system in the combined occurrence of strongly coeluting peaks, of distortions in the time sensors among chromatograms, and of the presence of unexpected compounds not included in the calibration step. Second-order HPLC-DAD data matrices were obtained in a short time (10 min) on a C18 column with a chromatographic system operating in isocratic mode (mobile phase was 20 mmol L−1 phosphate buffer at pH 2.20) and a flow-rate of 1.0 mL min−1 at room temperature. Parallel factor analysis (PARAFAC) and unfolded partial least-squares combined with residual bilinearization (U-PLS/RBL) were the second-order calibration algorithms select for data processing. The performance of the analytical parameters was good with an outstanding limit of detection (LODs) for acids ranging from 0.15 to 10.0 mmol L−1 in the validation samples. The improved method was applied to the analysis of many dairy products (yoghurt, cultured milk and cheese) and wine. The method was shown as an effective means for determining and following acid contents in fermented food and was characterized by reducibility with simple, high resolution and rapid procedure without derivatization of analytes.Fil: Mortera, Pablo.     Verso un modello di governance resiliente del patrimonio pubblico : azioni di rigenerazione urbana sull’ex complesso ospedaliero G. B. Giustinian

    Il contributo si pone l’obiettivo di presentare un progetto di rigenerazione urbana del centro storico di Venezia partendo da una proposta di governance alternativa e resiliente degli asset pubblici. La metodologia con cui ù stato elaborato sviluppa delle linee guida per un approccio metodologico progettuale che rende questo processo esportabile nel contesto generale dei centri storici italiani. Il progetto si avvale del contributo del dipartimento di Amministrazione Finanza e Controllo dell’università di Ca’ Foscari. Esso si occupa di un intervento urbanistico, architettonico ed economico. In particolare, vengono esaminati i principali mercati economici del contesto urbano - analisi PEST - per prevederne i futuri trend e di conseguenza adattare le azioni di progetto alla previsione. In seguito, viene delineato il quadro strategico dei soggetti promotori e degli stake holder al fine di coordinare l’analisi multi-obiettivi con il business model. Lo sviluppo del business plan viene infine utilizzato per proporre un sistema di intervento coordinato sulle principali situazioni urbane in cui il processo di rigenerazione ù attuabile in funzione della disponibilità del patrimonio e degli stakeholders

    Thermal stability of water ice in Ceres' crater Oxo

    Dwarf planet Ceres, target of the NASA Dawn mission, exhibits evidences of ammoniated phyllosilicates on its surface [1], compatible with a likely outer Solar System origin. Considerable amounts of water ice have recently been detected in some craters by the Visible InfraRed mapping spectrometer (VIR) onboard Dawn in some small fresh crater, such as Oxo, located at about 40° N. The exposure mechanism of water ice is unknown: cryovolcanism, cometary type sublimation/recondensation [2]or impacts with other bodies are likely mechanisms. The evaluation of the time stability of the water ice is crucial to understand the plausible mechanism for its existence. For this purpose, we developed a 3D finite-elements model (FEM) by using the topography given by the shape model of Ceres derived on the basis of images acquired by the Framing Camera in the Survey mission phase. The illumination conditions are provided by the SPICE toolkit. We performed several simulations by analyzing the effect of thermal inertia and albedo on the temperature and rate of ice sublimation.     Exploring potential risk factors of antimicrobial use in beef cattle.

    Livestock species are major contributors to the increase of antimicrobial (AM) resistance which is a worldwide concern for both human and animal health. The over-use of AM is widely acknowledged, however, unlike pigs, poultry and dairy cattle, knowledge on potential risk factors affecting AM usage (AMU) in beef industry is limited. Hence, this study aimed to investigate the impact of farm, breed, sex and season of arrival of purchased beef cattle on AMU in Italian beef cattle. Data on 1063 batches were collected from January 2016 to April 2019 from specialised beef fattening farms located in the north of Italy. Information on breed, sex, date of arrival, performance traits and AM agents used on farm was collected, and the treatment incidence 100 (TI100) indexes per batch were calculated using the defined daily dose animal estimated according to Italian summaries of product characteristics. Factors affecting TI100 indexes were investigated using a cross-classified multilevel model. Farms largely differed in terms of AMU. Males had greater AMU than females (P < 0.001), likely due to their higher susceptibility to disease. Statistically significant differences were observed between seasons of arrival with summer and spring having lower TI100 indexes than winter and autumn (P < 0.001). Indeed, winter is commonly linked to an increase in respiratory diseases in beef cattle. Finally, the TI100it indexes tended to be different among breeds with Blonde d'Aquitaine and Limousine having greater AMU compared to the other breeds. Results of this study provided valuable information on potential risk factors of AMU in beef production which may be useful to address its reduction. For instance, the development of tailored management strategies for specific breeds, targeted approaches to improve the health of males as well as greater care towards batches purchased in winter are possible advice to implement on-farm for a more responsible AM stewardship
