    The higher civil service and bureaucracy: A comparative analysis of Great Britain and the' United States

    Magister Artium - MAThis thesis is a comparative study of the higher civil service and bureaucracy of Great Britain and the United States. The study analyzes the political framework of the British and the United States systems of governance, examples of administrative reforms in the two systems, and the impact of education, socialization, recruitment, and civil servants as policy-makers. The methodology used in this study involves longitudinal as well as cross-national comparison. In dealing with differences between Great Britain and the United States, the study concentrates on the antecedent variables (constitution, political framework, cultural and administrative reform), intermediate variables (education, socialization and recruitment procedures), and the dependant variables (status of senior civil servants as policy- makers)In the first part of the study, the constitutional allocations of political power, history and the political system in which the higher civil service and bureaucracy operate are analyzed. The purpose here is to show that the bureaucracy and the civil service do not exist in a vacuum, they are influenced by constitutional, political and cultural constraints. The second part of the thesis deals with the education, socialization and the recruitment of the higher civil servants of Great Britain and the United States. This section points to the disproportionate representation of educated, high-status officials at the top of the political and administrative hierarchy of both countries. In Britain, however, there are social traditions built into the education system. The education and recruitment process concentrates on a general approach. In the United States, on the other hand, the specialist tradition dominates the civil service. Thus, United States higher civil servants are essentially specialists. The third part of the study analyzes the impact of education, socialization and recruitment processes on the role and performance of senior civil servants as policy-makers, in both societies. It is evident that civil servants are involved in the process of policy-making and, therefore, have a political role. This is due to the intricacies of bureaucracy and the fact that civil servants relative permanency, experience and expertise gives them a vast amount of knowledge that is relevant to policy-making. The conclusions suggest that the generalist approach applied in Great Britain hampers the capability of senior civil servants when it comes to negotiating with interest groups involved in policymaking.     Characterization of South African waste foundry molding sand : metallic contaminants

    Waste foundry sands are invariably contaminated to some extent during the manufacturing of cast alloys. As such the sand is regarded as a hazardous material that requires exceptional precautions for its disposal. Therefore, the study is initiated to identify, quantify and to classify metallic contaminants present in these sands. To achieve these objectives in conjunction with the South African Waste Management Act which plays as the guideline for industrial waste disposal, samples were collected from various South African foundries. In the present study, ten waste sand samples were characterised using XRF, XRD, SEM-EDS and Sulphur analyser. It was found after comparison with a virgin sand used as control or reference sand, that the cast alloy and the moulding additives are the main pollutants present in the waste foundry sand. The additional sulphur and acid potential characterisation, showed that the waste foundry sand has a low potential for sulphuric acid and acid sulphate soil formation when submerged in aquatic medium. The leaching behaviour and the total metallic concentration of the waste was similar to the virgin soil thus their similar classification in the same waste class category

    Case report: mechanisms of HIV elite control in two African women

    Background The majority of people living with HIV require antiretroviral therapy (ART) for controlling viral replication, however there are rare HIV controllers who spontaneously and durably control HIV in the absence of treatment. Understanding what mediates viral control in these individuals has provided us with insights into the immune mechanisms that may be important to induce for a vaccine or functional cure for HIV. To date, few African elite controllers from high incidence settings have been described. We identified virological controllers from the CAPRISA 002 cohort of HIV-1 subtype C infected women in KwaZulu Natal, South Africa, two (1%) of whom were elite controllers. We examined the genetic, clinical, immunological and virological characteristics of these two elite HIV controllers in detail, to determine whether they exhibit features of putative viral control similar to those described for elite controllers reported in the literature. Case presentation In this case report, we present clinical features, CD4+ T cell and viral load trajectories for two African women over 7 years of HIV infection. Viral load became undetectable 10 months after HIV infection in Elite Controller 1 (EC1), and after 6 weeks in Elite Controller 2 (EC2), and remained undetectable for the duration of follow-up, in the absence of ART. Both elite controllers expressed multiple HLA Class I and II haplotypes previously associated with slower disease progression (HLA-A*74:01, HLA-B*44:03, HLA-B*81:01, HLA-B*57:03, HLA-DRB1*13). Fitness assays revealed that both women were infected with replication competent viruses, and both expressed higher mRNA levels of p21, a host restriction factor associated with viral control. HIV-specific T cell responses were examined using flow cytometry. EC1 mounted high frequency HIV-specific CD8+ T cell responses, including a B*81:01-restricted Gag TL9 response. Unusually, EC2 had evidence of pre-infection HIV-specific CD4+ T cell responses. Conclusion We identified some features typical of elite controllers, including high magnitude HIV-specific responses and beneficial HLA. In addition, we made the atypical finding of pre-infection HIV-specific immunity in one elite controller, that may have contributed to very early viral control. Agricultural areas must expand unless unprecedented rapid yield improvements are achieved. While these transformations are challenging to accomplish and often associated with increased greenhouse gas emissions, the alternative for a nutrition-secure future is to rely increasingly on imports, which would outsource emissions and be economically and politically challenging given the large import increases required

    Shared N417-dependent epitope on the SARS-CoV-2 Omicron, Beta, and Delta Plus variants

    As severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to evolve, several variants of concern (VOCs) have arisen which are defined by multiple mutations in their spike proteins. These VOCs have shown variable escape from antibody responses and have been shown to trigger qualitatively different antibody responses during infection. By studying plasma from individuals infected with either the original D614G, Beta, or Delta variants, we showed that the Beta and Delta variants elicit antibody responses that are overall more cross-reactive than those triggered by D614G. Patterns of cross-reactivity varied, and the Beta and Delta variants did not elicit cross-reactive responses to each other. However, Beta-elicited plasma was highly cross-reactive against Delta Plus (Delta+), which differs from Delta by a single K417N mutation in the receptor binding domain, suggesting that the plasma response targets the N417 residue. To probe this further, we isolated monoclonal antibodies from a Beta-infected individual with plasma responses against Beta, Delta+, and Omicron, which all possess the N417 residue. We isolated an N417-dependent antibody, 084-7D, which showed similar neutralization breadth to the plasma. The 084-7D MAb utilized the IGHV3-23*01 germ line gene and had somatic hypermutations similar to those of previously described public antibodies which target the 417 residue. Thus, we have identified a novel antibody which targets a shared epitope found on three distinct VOCs, enabling their cross-neutralization. Understanding antibodies targeting escape mutations, such as K417N, which repeatedly emerge through convergent evolution in SARS-CoV-2 variants, may aid in the development of next-generation antibody therapeutics and vaccines. IMPORTANCE : The evolution of SARS-CoV-2 has resulted in variants of concern (VOCs) with distinct spike mutations conferring various immune escape profiles. These variable mutations also influence the cross-reactivity of the antibody response mounted by individuals infected with each of these variants. This study sought to understand the antibody responses elicited by different SARS-CoV-2 variants and to define shared epitopes. We show that Beta and Delta infections resulted in antibody responses that were more cross-reactive than the original D614G variant, but they had differing patterns of cross-reactivity. We further isolated an antibody from Beta infection which targeted the N417 site, enabling cross-neutralization of Beta, Delta+, and Omicron, all of which possess this residue.     Group B Streptococcus vaccine development: present status and future considerations, with emphasis on perspectives for low and middle income countries.

    Globally, group B Streptococcus (GBS) remains the leading cause of sepsis and meningitis in young infants, with its greatest burden in the first 90 days of life. Intrapartum antibiotic prophylaxis (IAP) for women at risk of transmitting GBS to their newborns has been effective in reducing, but not eliminating, the young infant GBS disease burden in many high income countries. However, identification of women at risk and administration of IAP is very difficult in many low and middle income country (LMIC) settings, and is not possible for home deliveries. Immunization of pregnant women with a GBS vaccine represents an alternate pathway to protecting newborns from GBS disease, through the transplacental antibody transfer to the fetus in utero. This approach to prevent GBS disease in young infants is currently under development, and is approaching late stage clinical evaluation. This manuscript includes a review of the natural history of the disease, global disease burden estimates, diagnosis and existing control options in different settings, the biological rationale for a vaccine including previous supportive studies, analysis of current candidates in development, possible correlates of protection and current status of immunogenicity assays. Future potential vaccine development pathways to licensure and use in LMICs, trial design and implementation options are discussed, with the objective to provide a basis for reflection, rather than recommendations

    Meta-evaluations in government and government institutions: A case study example from the Australian Centre for International Agricultural Research

    In this paper we draw on impact assessment work of the Australian Centre for International Agricultural Research (ACIAR) to present an example of meta-evaluation - an evaluation of evaluations - in an agricultural research, development and extension setting. We explore quality issues relating to evaluation studies in the context of government institutions. Program evaluation standards (PES) are divided into categories of utility, feasibility, propriety and accuracy to provide a framework for the meta-evaluation. The PES are presented as a universal measure of evaluation study quality. The intent of using them here is to judge the adequacy of PES as a universal quality measure or meta-evaluation base and to extract useful insights from ACIAR program evaluation activities when developing a meta-evaluation model for the Limpopo Department of Agriculture (LDA). Our meta-evaluation is undertaken of 63 impact assessment reports. First, the literature guiding the conduct of a meta-evaluation is reviewed. Second, an assessment (the meta-evaluation) of the evaluation studies is carried out for 19 sampled reports from a population of relevant reports fitting the dimension of the analysis, and results are presented and discussed. Also, lessons learned are presented, using the framework provided by the meta-evaluation criteria. Third, taking into account the lessons learned, implications are drawn for a proposed systematic meta-evaluation of the LDA. Finally, we conclude that all the PES cannot be equally emphasized in a meta-evaluation model. At ACIAR, 70% of the standards were at least partially addressed. Therefore, we succeeded in using the PES in judging the ACIAR evaluation quality. As such, they can be an important base when developing an evaluation model but should be applied in a contextualized manner
