    Ceramide galactosyltransferase (UGT8) as a molecular marker of canine mammary tumor malignancy

     Thirty-two canine mammary tubulopapillary carcinomas and 14 simple adenomas were studied by immunohistochemistry for the expression of UDP-galactose:ceramide galactosyltransferase (UGT8). The majority of tissue specimens (57%) representing adenomas had no or weak reaction with anti-UGT8 antibodies (0-2 pts according to IRS scale) in comparison to the majority of carcinomas (90%) which stained with high intensities (3-9 pts according to IRS scale). When the average values of the reaction intensities (IRS) for malignant and benign tumors were compared, using the Mann-Whitney U-test, significant differences in UGT8 expression between them were found (P < 0.001). Mammary tubulopapillary carcinomas were further analyzed by IHC and the same rabbit polyclonal antibody directed against UGT8 according to their malignancy grade. It was found that the level of UGT8 increased in tumor specimens together with their grading. A comparison of the average values of the reaction intensity (IRS scale) revealed a significant difference (Mann-Whitney U-test, P < 0.05) in UGT8 expression between tumors representing malignancy grades G3 and G1. Based on the obtained results, it is proposed that UGT8 is associated with malignancy of canine mammary gland cells and may have a potential value as a diagnostic marker.

    THE LONG-TERM IMPACT OF AMELIORATING DOSES OF HARD COAL FLY ASH ON SHAPING THE CONTENT OF SELECTED MICROELEMENTS IN AGRICULTURAL SOIL

    The effect of ameliorating doses of hard coal fly ash (HCFA) on chemical properties of soil and on yields and chemical composition of crops was examined based on an experiment established in 1984. At first, typical agricultural crops were grown in the experiment, but in 1992 the field was turned into permanent grassland. The current study took place twenty-nine years after hard coal fly ash had been applied. Soil samples were collected from the 0–20 cm soil layer, corresponding to the compared treatments. The residual effect of HCFA doses increasing from 100 to 800 Mg·ha-1 caused a regular increase in the total content of Cd, Cr, Cu, Mn, Fe, Zn and Pb, as well as the forms of Zn, Cu, Mn, Fe and B soluble in 0.1 M HCl. The total content of the analyzed microelements increased by a maximum of 30% for cadmium up to 176% for zinc. The highest increase in the content of soluble forms ranged from 25% for Fe to 760% for boron. The share of Znbd, i.e. permanently bound to soil, to Zntot, i.e. the total zinc form, was positively correlated with pH in KCl, the C:N ratio and the content of Cu permanently bound with soil. Regarding the share of Febd in Fetot, correlations with pH in H2O and in KCl, content of organic matter (SOM) and the C:N ratio were demonstrated. No such correlations were proven with respect to the other elements. W doświadczeniu założonym w 1984 roku badano działanie melioracyjnych dawek popiołów ze spalania węgla kamiennego (hard coal fly ashes – HCFA) na właściwości fizykochemiczne gleby oraz na plonowanie i skład chemiczny roślin uprawnych. W pierwszych latach badań uprawiano typowe rośliny rolnicze, a w 1992 zmieniono charakter uprawy na trwały użytek zielony. Obecne badania wykonano po dwudziestu dziewięciu latach od zastosowania HCFA. Próbki gleby pobrano z poziomu 0–20 cm w nawiązaniu do porównywanych obiektów. Następcze działanie rosnących dawek HCFA od 100 do 800 Mg·ha-1 powodowało zarówno wyraźnie regularny wzrost zawartości całkowitej – Cd, Cr, Cu, Mn, Fe, Zn i Pb, jak i form rozpuszczalnych w 0.1 M HCl – Zn, Cu, Mn, Fe i B. Zawartość całkowita rozpatrywanych mikroskładników maksymalnie wzrosła o 30% w przypadku kadmu do 176 % w odniesieniu do cynku. Maksymalny wzrost zawartości form rozpuszczalnych kształtował się od 25 % w odniesieniu do żelaza do 760 % w przypadku boru. Udział Znbd tj. trwale związanego z glebą w stosunku do jego formy ogólnej Zntot, istotnie dodatnio korelował z pH w KCl i stosunek C:N oraz z Cu trwale związaną z glebą. W odniesieniu do udziału Febd w Fetot wykazano korelacje z pH w H2O, pH w KCl, SOM i C:N ratio. W odniesieniu do pozostałych badanych pierwiastków takich zależności nie wykazano

    Inhibin-α, E-cadherin, calretinin and Ki-67 antigen in the immunohistochemical evaluation of canine and human testicular neoplasms

    Introduction. The steady increase of dogs with diagnosed testicular neoplasms observed in recent years prompted us to carry out immunohistochemical (IHC) studies for their better characterization. The aim of the study was to analyze most common canine testicular neoplasms (seminomas, Leydig cell and Sertoli cell tumors) with selected IHC markers and to compare the expressions of these proteins in corresponding canine and human testicular tumors. Material and methods. Studies were carried out on testicular canine tumors: 40 cases of seminoma, 40 cases of Leydig cell tumor and 40 cases of Sertoli cell tumor. Moreover, 15 cases of human seminomas and 5 cases of human Leydig cell tumors were also analyzed. Immunohistochemistry was performed on paraffin sections by standard technique using monoclonal anti-human antibodies against E-cadherin, inhibin-α, calretinin and Ki-67. The slides were subjected to computer-aided image analysis and the intensity of the immunoreactivity was assessed by a semi-quantitative scoring system. Results. Due to the very low prevalence of the Sertoli cell-derived tumors in the human population, we were able to examine the markers’ expression only in the canine gonadal tumors. We revealed that, apart from E-cadherin in Leydig cell tumors and calretinin in seminomas, the expression of all the analyzed markers in canine and human testicular tumors was similar. E.g. there was no immunoexpression of inhibin-α in 75% of canine and 100% of human cases of seminoma. The immunoreactivity of Ki-67 was intense in 40% of canine and 60% of human seminomas. Immunoexpression of inhibin-α in Leydig cell tumor was intense in 70% of canine and 100% of human cases, respectively. Also the immunoreactivity of calretinin was intense in 75% of cases of canine and 100% of human Leydig cell tumors. In 50% of canine and 40% of human Leydig cell tumors, the immunoexpression of Ki-67 was weak. Conclusions. The applied anti-human monoclonal antibodies against common antigens and markers of human testicular neoplasms could be routinely used for the immunohistochemical evaluation of canine testicular tumors.

    Vemurafenib and dabrafenib downregulates RIPK4 level

    Vemurafenib and dabrafenib are BRAF kinase inhibitors (BRAFi) used for the treatment of patients with melanoma carrying the V600E BRAF mutation. However, melanoma cells develop resistance to both drugs when used as monotherapy. Therefore, mechanisms of drug resistance are investigated, and new molecular targets are sought that could completely inhibit melanoma progression. Since receptor-interacting protein kinase (RIPK4) probably functions as an oncogene in melanoma and its structure is similar to the BRAF protein, we analyzed the impact of vemurafenib and dabrafenib on RIPK4 in melanomas. The in silico study confirmed the high similarity of BRAF kinase domains to the RIPK4 protein at both the sequence and structural levels and suggests that BRAFi could directly bind to RIPK4 even more strongly than to ATP. Furthermore, BRAFi inhibited ERK1/2 activity and lowered RIPK4 protein levels in BRAF-mutated melanoma cells (A375 and WM266.4), while in wild-type BRAF cells (BLM and LoVo), both inhibitors decreased the level of RIPK4 and enhanced ERK1/2 activity. The phosphorylation of phosphatidylethanolamine binding protein 1 (PEBP1) - a suppressor of the BRAF/MEK/ERK pathway - via RIPK4 observed in pancreatic cancer did not occur in melanoma. Neither downregulation nor upregulation of RIPK4 in BRAF- mutated cells affected PEBP1 levels or the BRAF/MEK/ERK pathway. The downregulation of RIPK4 inhibited cell proliferation and the FAK/AKT pathway, and increased BRAFi efficiency in WM266.4 cells. However, the silencing of RIPK4 did not induce apoptosis or necroptosis. Our study suggests that RIPK4 may be an off-target for BRAF inhibitors

    The correlation of mutations and expressions of genes within the PI3K/Akt/mTOR pathway in breast cancer : a preliminary study

    There is an urgent need to seek new molecular biomarkers helpful in diagnosing and treating breast cancer. In this elaboration, we performed a molecular analysis of mutations and expression of genes within the PI3K/Akt/mTOR pathway in patients with ductal breast cancer of various malignancy levels. We recognized significant correlations between the expression levels of the studied genes. We also performed a bioinformatics analysis of the data available on the international database TCGA and compared them with our own research. Studies on mutations and expression of genes were conducted using High-Resolution Melt PCR (HRM-PCR), Allele-Specific-quantitative PCR (ASP-qPCR), Real-Time PCR molecular methods in a group of women with ductal breast cancer. Bioinformatics analysis was carried out using web source Ualcan and bc-GenExMiner. In the studied group of women, it was observed that the prevalence of mutations in the studied PIK3CA and AKT1 genes was 29.63%. It was stated that the average expression level of the PIK3CA, PIK3R1, PTEN genes in the group of breast cancer patients is lower in comparison to the control group, while the average expression level of the AKT1 and mTOR genes in the studied group was higher in comparison to the control group. It was also indicated that in the group of patients with mutations in the area of the PIK3CA and AKT1 genes, the PIK3CA gene expression level is statistically significantly lower than in the group without mutations. According to our knowledge, we demonstrate, for the first time, that there is a very strong positive correlation between the levels of AKT1 and mTOR gene expression in the case of patients with mutations and without mutations

    Surgical treatment of rectal cancer in Poland — a report from a prospective, multi-centre observational study PSSO_01 conducted under the auspices of the Polish Society of Surgical Oncology

    Introduction. Since 2016, as part of the PSSO_01 multi-centre research project conducted under the auspices of the Polish Society of Surgical Oncology, clinical data on rectal cancer treatment have been collected. The objective of the study was to illustrate the state of early results of surgical treatment. Material and methods. The research project is multi-centre in nature. Data shall be collected electronically. The study protocol does not impose or suggest any course of procedure. It only systematizes the way data are collected for scientific purposes. The analysis of early results of surgical treatment was compared with the results of population studies from other European countries (Netherlands, Belgium). Results. By the end of June 2018, 736 patients were registered in the study. In 399 (54.2%) an anterior resection was performed. More than half of patients undergoing subsequent surgical treatment (54.2%) receive neoadjuvant treatment, with the percentage of patients undergoing radiotherapy or radiochemical treatment for lower rectal cancer being about 70%. Most patients (96%) are operated in elective procedure. The percentage of laparoscopic surgeries is low (8.6%). Postoperative complications are observed in 21.1% of patients. Severe complications (grades III–V according to Clavien-Dindo classification) occur in 7.6% of patients undergoing surgery. Postoperative mortality is 1.1%. Discussion. Although the project does not have the character of a registry and does not allow for drawing wider conclusions concerning the compliance with the standards of qualification for neoadjuvant treatment, the important information is that more than half of rectal cancer patients receive preoperative treatment, and the percentage of severe postoperative complications does not exceed 10%. Conclusions. The results of the PSSO_01 project are representative and reflect the actual situation concerning surgical treatment of rectal cancer patients in Poland

    Useful immunohistochemical indicators in canine mast cell tumours

    Morphological and immunohistochemical analysis of 45 canine mast cell tumours was performed to determine whether the proteins examined are useful for a more precise description of tumour morphology and a more reliable determination of the prognosis in patients. Tissue sections were stained according to the standard haematoxylin and eosin (HE) technique and with toluidine blue to demonstrate cytoplasmic granules. Immunohistochemical studies were performed, using the cell markers CD117 (c-kit), p16 and von Willebrand factor (FVIII). In CD117 three different staining patterns were observed: (1) membranous reaction, (2) intense staining of cytoplasm, and (3) a diffuse, delicate cytoplasmic reaction. Von Willebrand antibody was evaluated on the basis of the number of blood vessels stained. p16 expression was evaluated by scoring positive nuclear reaction. Positive expression was demonstrated for all examined antigens, but their level of expression differed depending on the grades of tumour malignancy. Statistical analysis of the results documented a pronounced positive correlation between the markers studied and the grade of tumour malignancy (P < 0.001). It was shown that each of the cell markers examined represents a useful prognostic indicator for patients with mast cell tumours.     Leczenie chirurgiczne raka odbytnicy w Polsce — raport z prospektywnego, wieloośrodkowego badania obserwacyjnego PSSO_01 prowadzonego pod auspicjami Polskiego Towarzystwa Chirurgii Onkologicznej

    Wstęp. Od 2016 roku w ramach wieloośrodkowego projektu badawczego PSSO_01 prowadzonego pod auspicjami Polskiego Towarzystwa Chirurgii Onkologicznej, gromadzone są dane kliniczne dotyczące leczenia raka odbytnicy. Celem pracy było zobrazowanie stanu dotyczącego wczesnych wyników leczenia chirurgicznego. Materiał i metody. Projekt badawczy ma charakter wieloośrodkowy. Dane gromadzone są elektronicznie. Protokół badania nie narzuca ani nie sugeruje żadnego sposobu postępowania, systematyzuje jedynie sposób zbierania danych w celach naukowych. Analiza dotycząca wczesnych wyników leczenia chirurgicznego została porównana z rezultatami populacyjnych badań z innych krajów europejskich (Holandia, Belgia). Wyniki. Do końca czerwca 2018 roku w badaniu zarejestrowano 736 chorych. U 399 (54,2%) wykonano resekcję przednią. Leczenie neoadiuwantowe otrzymała ponad połowa chorych, poddanych następnie leczeniu operacyjne­mu (54,2%), przy czym odsetek pacjentów poddanych radio- bądź radiochemioterapii z powodu raka dolnej części odbytnicy wyniósł około 70%. Większość chorych (96%) operowana była w trybie planowym. Odsetek operacji wykonanych techniką laparoskopową jest niski (8,6%). Powikłania pooperacyjne zaobserwowano u 21,1% chorych. Ciężkie powikłania (III–V st. wg klasyfikacji Claviena-Dindo) sięgały 7,6% operowanych chorych. Śmiertelność po­operacyjna wyniosła 1,1%. Dyskusja. Chociaż projekt nie ma charakteru rejestru i nie pozwala na wyciągniecie szerszych wniosków dotyczących przestrzegania standardów kwalifikacji do leczenia neoadiuwantowego, istotną informacją jest to, że ponad połowa chorych na raka odbytnicy otrzymuje leczenie przedoperacyjne, a odsetek ciężkich powikłań pooperacyjnych nie przekracza 10%. Wnioski. Wyniki projektu PSSO_01 są reprezentatywne i odzwierciedlają faktyczną sytuację dotyczącą leczenia chirurgicznego chorych na raka odbytnicy w Polsce
