    Ferritin nanovehicle for targeted delivery of cytochrome C to cancer cells

    In this work, we have exploited the unique properties of a chimeric archaeal-human ferritin to encapsulate, deliver and release cytochrome c and induce apoptosis in a myeloid leukemia cell line. The chimeric protein combines the versatility in 24-meric assembly and cargo incorporation capability of Archaeglobus fulgidus ferritin with specific binding of human H ferritin to CD71, the “heavy duty” carrier responsible for transferrin-iron uptake. Delivery of ferritin-encapsulated cytochrome C to the Acute Promyelocytic Leukemia (APL) NB4 cell line, highly resistant to transfection by conventional methods, was successfully achieved in vitro. The effective liberation of cytochrome C within the cytosolic environment, demonstrated by double fluorescent labelling, induced apoptosis in the cancer cells

Disciplinary referrals are on the rise in American schools and are a cause of concern for teachers, administrators, parents, and community members. School personnel currently implement programs designed to curb discipline problems in the classroom and foster productive behavior among adolescents. There is some debate and conflicting literature on whether sports participation has a beneficial influence on students. The writings of Virgina Chomitz, who hypothesized a positive relationship between academic achievement and physical fitness, guided this research. This study analyzed grade point average (GPA) and discipline referral data for 4,433 students in a suburban American high school over a 3-year time period. The GPA and referral data for athletes and nonathletes were compared using multiple t tests, and it was found that athletes had significantly higher GPAs and lower discipline referrals when compared to nonathletes. These results, which match the original hypotheses, provide greater insight into how participating in athletics may improve a student\u27s behavior and academic achievement. With this knowledge, educators may place greater emphasis on athletics as a method to promote achievement and positive experiences among high school students. These positive effects will contribute to social change for students at an individual level and for entire school environments

    β-hexachlorocyclohexane: a small molecule with a big impact on human cellular biochemistry

    Organochlorine pesticides (OCPs) belong to a heterogeneous class of organic compounds blacklisted by the Stockholm Convention in 2009 due to their harmful impact on human health. Among OCPs, β-hexachlorocyclohexane (β-HCH) is one of the most widespread and, at the same time, poorly studied environmental contaminant. Due to its physicochemical properties, β-HCH is the most hazardous of all HCH isomers; therefore, clarifying the mechanisms underlying its molecular action could provide further elements to draw the biochemical profile of this OCP. For this purpose, LNCaP and HepG2 cell lines were used as models and were subjected to immunoblot, immunofluorescence, and RT-qPCR analysis to follow the expression and mRNA levels, together with the distribution, of key biomolecules involved in the intracellular responses to β-HCH. In parallel, variations in redox homeostasis and cellular bioenergetic profile were monitored to have a complete overview of β-HCH effects. Obtained results strongly support the hypothesis that β-HCH could be an endocrine disrupting chemical as well as an activator of AhR signaling, promoting the establishment of an oxidative stress condition and a cellular metabolic shift toward aerobic glycolysis. In this altered context, β-HCH can also induce DNA damage through H2AX phosphorylation, demonstrating its multifaceted mechanisms of action

    A dynamic load balancing algorithm for Quality of Service and mobility management in next generation home networks

    Heterogeneity of connection technologies and nodes mobility open new challenges in home networks control strategies. Moreover, user's needs are changing towards applications requiring high transmission speeds such as 3D gaming, enhanced interactivity and high definition video. Each of those applications puts several constraints on the network capabilities to guarantee requirements on the Quality of Service. In this paper we introduce an innovative concept based on fast load balancing algorithm operating on top of a convergence layer, in order to rapidly react to network changes and contemporaneously to satisfy strict application demands. We formulated the load balancing problem as a Multi-Commodity Flow and resolved it with a column generation approach using Lagrangian Relaxation and Dijkstra algorithm. The load balancing problem computational complexity is decreased with respect to state of the art load balancing solutions based on linear programming techniques. Proof of concept simulation results are reported

    Antioxidant properties of the decarboxylated dimer of aminoethylcysteine ketimine: assessment of its ability to scavenge peroxynitrite.

    The natural sulfur compound aminoethylcysteine ketimine decarboxylated dimer (AECK dimer) has been investigated for its ability to act as peroxynitrite scavenger. It has been found that the product efficiently protects against the nitration of tyrosine and the inactivation of oz-antiproteinase by peroxynitrite. The tyrosine nitration can be completely prevented by 100 mu M AECK dimer which appears as effective as the antioxidants glutathione and N-acetylcysteine. The AECK dimer was also found to limit surface charge alteration of low density lipoprotein induced by peroxynitrite. These findings indicate that the AECK dimer is a strong protective agent against peroxynitrite damage and that it could play an important role in the defence against oxidative stress in human diseases

    Music and physical activity in psychological well-being

    The present study was designed to examine the effects of listening to music during exercise of moderate intensity on mood, state anxiety, and time to exhaustion as well as to evaluate sex differences in 27 physically active (14 men, 13 women) subjects between the ages of 20 and 30 years. Participants completed the Profile of Mood States and the State Anxiety Inventory before and after treadmill running in Music and No music conditions. Music and No Music conditions were randomly assigned, and participants exercised at 75% of their Heart Rate Reserve until voluntary exhaustion. Analysis indicated participants reported statistically significant mean changes on Tension, Depression, Fatigue, Confusion, and State Anxiety. However, the findings for emotions yielded no significant effect of music, except findings suggested that women, but not men, reported greater mean Fatigue after exercising in the presence of music than in its absence. Also, there was a statistically significant finding suggesting that women exercised longer with music than without

    Aminoethylcysteine ketimine is a sulfur-containing cyclic compound produced by the enzymatic α-deamination of the parent aminoethylcysteine that has been detected in bovine brain and cerebellum. Aminoethylcysteine ketimine is known to dimerize spontaneously and easily lose one carboxyl group. This decarboxylated compound, simply named the dimer, has been recently detected in normal human urine. In this article we provide evidence on the occurrence of the dimer in the bovine cerebellum

    "The critical infrastructure protection is a key point from a social and economic point of view. The FP7 MICIE project has achieved promising results in evaluating impact of failures and faults in interdependent physical systems, as critical infrastructures. In order to achieve this goal, the consortium developed an online risk prediction tool, able to acquire information by local physical systems through their control centres and obtain accurate and synchronised predictions using shared interdependency models. However, results of MICIE project are not enough in order to quickly and effectively react to all adverse events that may occur over the system of systems and, in particular, to face cyber threats and attacks. The EC FP7 CockpitCI project aims to improve resilience and dependability of CIs through the design and the implementation of an alerting system that provides to CI operators an efficient tool to support them in the prevention of cyber attacks impact on real systems and in the implementation of possible consequence containment strategies in case of attack..
