    Pacific barkcloth under the microscope - characterisation of condition, decoration and structure

    The creation of barkcloth begins with harvesting the inner bark of certain types of trees followed by soaking and beating with grooved beaters, a process which often leaves undulations on the surface of the bark, also known as the beater mark. The cloth can then be decorated using colorants, applied as particulate pigments, dyes or paints. The resulting material is highly ornate with varying surface textures and colours. The usefulness of stereo and standard light microscopy, ranging from around x10 for low and up to x200 for high-magnification microscopy, and macro photography to examine the subtleties of the surface of the cloth is highlighted in this research, with examples of barkcloth from the Hunterian, University of Glasgow, Glasgow and Royal Botanic Gardens, Kew, London collections. Examples of beater marks, particulate dye material, cracking painted surfaces, as well as the presence of two species used to create one cloth are shown, aspects that are not immediately obvious when examining the cloths with the naked eye. To further enhance the analysis from microscopy XRF and FTIR were used when appropriate. Observing the material in this way can increase appreciation for the aesthetic aspects of barkcloth and can further knowledge of the materials used in production. This can inform condition reports, storage requirements, and potential conservation treatments

    Solvent vapour use - the unintended consequences in textile conservation

    The impact of solvent vapour from two solvents, acetone and industrial denatured alcohol (IDA), commonly used in adhesive reactivation in textile conservation, when applied to artificially soiled silk fabric through either Gore-Tex® or Reemay® membranes for exposure times of either 1 or 3 min, was measured using microscopy and ImageJ to monitor the movement of the solid particulates of the soiling, and Fourier transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR) to monitor the movement of the oily components. Analysis using these techniques successfully showed the movement of large particulates through the textile and a decrease in the presence of oil. Analysis with FTIR-ATR showed that the application of solvent vapour by a poultice method produces more than just a surface change and the vapour can penetrate far enough into the fibres to cause a change in the level of oil through the entire textile. The results showed that the solvents and barrier membranes have characteristic differences which impact on the changes on the soiling. The application of acetone produced a greater change in the movement of oil on the front and back of the samples than the application of IDA, while a comparison between the barrier membranes showed a greater change occurring in the level of oil with the use of Gore-Tex® rather than Reemay®. The time of exposure to solvent vapour made little difference to the changes to the textile soiling. Quantitatively significant results were gathered from analysis of the changes in the oil measured with FTIR-ATR, and qualitative changes in the large particulate soiling, demonstrating the potential usefulness of ImageJ open access software in future historic textile soiling monitoring studies

    Applying genomic and transcriptomic advances to mitochondrial medicine

    Next-generation sequencing (NGS) has increased our understanding of the molecular basis of many primary mitochondrial diseases (PMDs). Despite this progress, many patients with suspected PMD remain without a genetic diagnosis, which restricts their access to in-depth genetic counselling, reproductive options and clinical trials, in addition to hampering efforts to understand the underlying disease mechanisms. Although they represent a considerable improvement over their predecessors, current methods for sequencing the mitochondrial and nuclear genomes have important limitations, and molecular diagnostic techniques are often manual and time consuming. However, recent advances in genomics and transcriptomics offer realistic solutions to these challenges. In this Review, we discuss the current genetic testing approach for PMDs and the opportunities that exist for increased use of whole-genome NGS of nuclear and mitochondrial DNA (mtDNA) in the clinical environment. We consider the possible role for long-read approaches in sequencing of mtDNA and in the identification of novel nuclear genomic causes of PMDs. We examine the expanding applications of RNA sequencing, including the detection of cryptic variants that affect splicing and gene expression and the interpretation of rare and novel mitochondrial transfer RNA variants

    The Significance of Short Food Supply Chains: Trends and Bottlenecks from the SKIN Thematic Network

    peer-reviewedShort Food Supply Chains (SFSCs) are central to the alternative food movement discourse. SFSCs are based upon the interrelations among actors who are directly involved in the production, processing, distribution, and consumption of food products. They depend upon actors mobilising resources of various kinds: skills; knowledge; labour; capital; buildings etc. External factors such as policies and regulations can also encourage the creation of these shorter chains. The development of SFSCs can still be hindered by a range of other factors. Nevertheless, bottlenecks can be overcome via the sharing of information on successful SFSCs through the dissemination of Good Practices between various actors and territories. The Short Supply Chain Knowledge and Innovation (SKIN) project uses the term ‘good’ rather than ‘best’ practice to draw attention to the subjective lens through which a practice is ultimately evaluated by an end-user. This paper first outlines the many issues that confront SFSC actors which represent bottlenecks to the adoption of ‘Good Practices’. It then documents the Good Practices collected as part of the SKIN project as tangible examples of how SFSCs overcome such challenges. Lessons learnt from project highlights are subsequently assessed in an effort to mitigate and offer solutions to the challenges associated with SFSCs. The paper demonstrates the considerable latent potential inherent to SFSCs. However, in order for the agricultural sector to realise the full promise of short supply chains it must first be conscious of the issues pertinent to their prosperity

    Effect of Wet and Dry Distillers Grains Plus Solubles and Supplemental Fat Level on Performance of Yearling Finishing Cattle

    Two finishing trials were conducted to compare the addition of fat from either wet or dry distillers byproducts (WDGS; DDGS) to that provided from one of two commercially available fat sources (corn oil, Trial 1; tallow, Trial 2). In Trial 1, feed conversion and ADG decreased linearly as level of corn oil increased, while feed conversion and ADG were improved numerically as the level of WDGS increased in the diet. In Trial 2, no differences in performance were observed comparing cattle fed dry distillers grains plus solubles to cattle fed diets containing tallow. WDGS provided 12 and 17% more net energy for gain when fed at 20 and 40% of the diet compared to a dry rolled/high moisture corn mix

    Phosphorus Requirement for Finishing Heifers

    Sixty head of crossbred heifers (614 lb initial BW) were individually fed one of five levels of phosphorus of 0.10, 0.17, 0.24, 0.31, 0.38 % of diet DM with supplemental P from monosodium phosphate. Heifers were fed an energy-dense diet composed primarily of corn starch and corn fiber to minimize P from corn. When compared to the other four levels, heifers fed at the 0.10 % P level had lower DMI, ADG and final BW. Heifers fed 0.10%P had lower plasma P concentration. Breakpoint analysis of ADG suggests that the P requirement for finishing heifers is 0.115% P of diet DM within the range of 8.2 to 10.3g P/day (0.104 and 0.127% P on a DM basis)

    Impact of Cleaning Frequency on Nitrogen Balance in Open Feedlot Pens

    Pen cleaning frequency of feedlot pens was evaluated during the summer of 2001 and 2002. Dry matter, organic matter and nitrogen recoveries were evaluated on a per head basis. Cleaning pens monthly compared to cleaning at the end of the feeding period resulted in significantly more DM, OM and N recovered. Cleaning pens every month increased N removal by 7.0 lb per steer (49.9% increase) above manure N removed at the end of the feeding period

    Effects of Corn Processing Method and Crude Protein Level with the Inclusion of Wet Corn Gluten Feed on Finishing Steer Performance

    Three hundred twenty crossbred steer calves were used to evaluate corn processing method and crude protein level in finishing diets that included wet corn gluten feed. There was no response due to crude protein level (14 vs 15%) observed in this trial. As corn processing method became more intensive (fine-grinding, high-moisture ensiling, and steam-flaking corn) compared to dry-rolling, daily intake was reduced. Daily gain was similar across corn processing methods. Feed efficiency and cost of gain improved as corn processing method intensity increased

    A finishing trial was conducted to evaluate level of wet corn gluten feed in steam-flaked corn based finishing diets. Feed efficiency and daily gain were similar among all levels (10, 20, 25, 30, and 35%, DM basis) of inclusion of Sweet Bran® wet corn gluten feed evaluated in this trial. These data indicate wet corn gluten feed is similar in energy content to steam-flaked corn based on animal performance

    One hundred sixty crossbred steer calves were stratified by weight and allotted into four groups to test three sorting strategies against an unsorted control to compare methods of sorting long yearling steers to decrease variation in carcass weight and fat thickness, increase pounds of carcass weight sold, and increase profitability. Sorting by weight upon entry into the feedlot or by weight and fat thickness at the end of the feeding period increased average carcass weight. No statistical differences in variation or profitability were found, although numerical differences were present
