    Quantitative localized proton-promoted dissolution kinetics of calcite using scanning electrochemical microscopy (SECM)

    Scanning electrochemical microscopy (SECM) has been used to determine quantitatively the kinetics of proton-promoted dissolution of the calcite (101Ģ…4) cleavage surface (from natural ā€œIceland Sparā€) at the microscopic scale. By working under conditions where the probe size is much less than the characteristic dislocation spacing (as revealed from etching), it has been possible to measure kinetics mainly in regions of the surface which are free from dislocations, for the first time. To clearly reveal the locations of measurements, studies focused on cleaved ā€œmirrorā€ surfaces, where one of the two faces produced by cleavage was etched freely to reveal defects intersecting the surface, while the other (mirror) face was etched locally (and quantitatively) using SECM to generate high proton fluxes with a 25 Ī¼m diameter Pt disk ultramicroelectrode (UME) positioned at a defined (known) distance from a crystal surface. The etch pits formed at various etch times were measured using white light interferometry to ascertain pit dimensions. To determine quantitative dissolution kinetics, a moving boundary finite element model was formulated in which experimental time-dependent pit expansion data formed the input for simulations, from which solution and interfacial concentrations of key chemical species, and interfacial fluxes, could then be determined and visualized. This novel analysis allowed the rate constant for proton attack on calcite, and the order of the reaction with respect to the interfacial proton concentration, to be determined unambiguously. The process was found to be first order in terms of interfacial proton concentration with a rate constant k = 6.3 (Ā± 1.3) Ɨ 10ā€“4 m sā€“1. Significantly, this value is similar to previous macroscopic rate measurements of calcite dissolution which averaged over large areas and many dislocation sites, and where such sites provided a continuous source of steps for dissolution. Since the local measurements reported herein are mainly made in regions without dislocations, this study demonstrates that dislocations and steps that arise from such sites are not needed for fast proton-promoted calcite dissolution. Other sites, such as point defects, which are naturally abundant in calcite, are likely to be key reaction sites

    Asbestos modulates thioredoxin-thioredoxin interacting protein interaction to regulate inflammasome activation

    BACKGROUND: Asbestos exposure is related to various diseases including asbestosis and malignant mesothelioma (MM). Among the pathogenic mechanisms proposed by which asbestos can cause diseases involving epithelial and mesothelial cells, the most widely accepted one is the generation of reactive oxygen species and/or depletion of antioxidants like glutathione. It has also been demonstrated that asbestos can induce inflammation, perhaps due to activation of inflammasomes. METHODS: The oxidation state of thioredoxin was analyzed by redox Western blot analysis and ROS generation was assessed spectrophotometrically as a read-out of solubilized formazan produced by the reduction of nitrotetrazolium blue (NTB) by superoxide. Quantitative real time PCR was used to assess changes in gene transcription. RESULTS: Here we demonstrate that crocidolite asbestos fibers oxidize the pool of the antioxidant, Thioredoxin-1 (Trx1), which results in release of Thioredoxin Interacting Protein (TXNIP) and subsequent activation of inflammasomes in human mesothelial cells. Exposure to crocidolite asbestos resulted in the depletion of reduced Trx1 in human peritoneal mesothelial (LP9/hTERT) cells. Pretreatment with the antioxidant dehydroascorbic acid (a reactive oxygen species (ROS) scavenger) reduced the level of crocidolite asbestos-induced Trx1 oxidation as well as the depletion of reduced Trx1. Increasing Trx1 expression levels using a Trx1 over-expression vector, reduced the extent of Trx1 oxidation and generation of ROS by crocidolite asbestos, and increased cell survival. In addition, knockdown of TXNIP expression by siRNA attenuated crocidolite asbestos-induced activation of the inflammasome. CONCLUSION: Our novel findings suggest that extensive Trx1 oxidation and TXNIP dissociation may be one of the mechanisms by which crocidolite asbestos activates the inflammasome and helps in development of MM

    Size-selective concentration of chondrules and other small particles in protoplanetary nebula turbulence

    Size-selective concentration of particles in a weakly turbulent protoplanetary nebula may be responsible for the initial collection of chondrules and other constituents into primitive body precursors. This paper presents the main elements of this process of turbulent concentration. In the terrestrial planet region, both the characteristic size and size distribution of chondrules are explained. "Fluffier" particles would be concentrated in nebula regions which were at a lower gas density and/or more intensely turbulent. The spatial distribution of concentrated particle density obeys multifractal scaling}, suggesting a close tie to the turbulent cascade process. This scaling behavior allows predictions of the probability distributions for concentration in the protoplanetary nebula to be made. Large concentration factors (>10^5) are readily obtained, implying that numerous zones of particle density significantly exceeding the gas density could exist. If most of the available solids were actually in chondrule sized particles, the ensuing particle mass density would become so large that the feedback effects on gas turbulence due to mass loading could no longer be neglected.     Terahertz oscillations in an In<sub>0.53</sub>Ga<sub>0.47</sub>As submicron planar gunn diode

    The length of the transit region of a Gunn diode determines the natural frequency at which it operates in fundamental mode ā€“ the shorter the device, the higher the frequency of operation. The long-held view on Gunn diode design is that for a functioning device the minimum length of the transit region is about 1.5Ī¼m, limiting the devices to fundamental mode operation at frequencies of roughly 60 GHz. Study of these devices by more advanced Monte Carlo techniques that simulate the ballistic transport and electron-phonon interactions that govern device behaviour, offers a new lower bound of 0.5Ī¼m, which is already being approached by the experimental evidence that has shown planar and vertical devices exhibiting Gunn operation at 600nm and 700nm, respectively. The paper presents results of the first ever THz submicron planar Gunn diode fabricated in In&lt;sub&gt;0.53&lt;/sub&gt;Ga&lt;sub&gt;0.47&lt;/sub&gt;A on an InP substrate, operating at a fundamental frequency above 300 GHz. Experimentally measured rf power of 28 ĀµW was obtained from a 600 nm long Ɨ120 Āµm wide device. At this new length, operation in fundamental mode at much higher frequencies becomes possible ā€“ the Monte Carlo model used predicts power output at frequencies over 300 GHz

    A perspective from extinct radionuclides on a Young Stellar Object: The Sun and its accretion disk

    Meteorites, which are remnants of solar system formation, provide a direct glimpse into the dynamics and evolution of a young stellar object (YSO), namely our Sun. Much of our knowledge about the astrophysical context of the birth of the Sun, the chronology of planetary growth from micrometer-sized dust to terrestrial planets, and the activity of the young Sun comes from the study of extinct radionuclides such as 26Al (t1/2 = 0.717 Myr). Here we review how the signatures of extinct radionuclides (short-lived isotopes that were present when the solar system formed and that have now decayed below detection level) in planetary materials influence the current paradigm of solar system formation. Particular attention is given to tying meteorite measurements to remote astronomical observations of YSOs and modeling efforts. Some extinct radionuclides were inherited from the long-term chemical evolution of the Galaxy, others were injected into the solar system by a nearby supernova, and some were produced by particle irradiation from the T-Tauri Sun. The chronology inferred from extinct radionuclides reveals that dust agglomeration to form centimeter-sized particles in the inner part of the disk was very rapid (<50 kyr), planetesimal formation started early and spanned several million years, planetary embryos (possibly like Mars) were formed in a few million years, and terrestrial planets (like Earth) completed their growths several tens of million years after the birth of the Sun.Comment: 49 pages, 9 figures, 1 table.     Global Defects in Field Theory with Applications to Condensed Matter

    We review investigations on defects in systems described by real scalar fields in (D,1) space-time dimensions. We first work in one spatial dimension, with models described by one and two real scalar fields, and in higher dimensions. We show that when the potential assumes specific form, there are models which support stable global defects for D arbitrary. We also show how to find first-order differential equations that solve the equations of motion, and how to solve models in D dimensions via soluble problems in D=1. We illustrate the procedure examining specific models and showing how they may be used in applications in different contexts in condensed matter physics, and in other areas.Comment: 15 pages, 9 figure

    Demystifying Governance and its Role for Transitions in Urban Socialā€“Ecological Systems

    Governance is key to sustainable urban transitions. Governance is a system of social, power, and decisionā€making processes that acts as a key driver of resource allocation and use, yet ecologistsā€”even urban ecologistsā€“seldom consider governance concepts in their work. Transitions to more sustainable futures are becoming increasingly important to the management of many ecosystems and landscapes, and particularly so for urban systems. We briefly identify and synthesize important governance dimensions of urban sustainability transitions, using illustrations from cities in which longā€term socialā€“ecological governance research is underway. This article concludes with a call to ecologists who are interested in environmental stewardship, and to urban ecologists in particular, to consider the role of governance as a driver in the dynamics of the systems they study

    Acupuncture, Counseling, and Usual care for Depression (ACUDep): study protocol for a randomized controlled trial

    Background: The evidence on the effect of acupuncture or counseling for depression is not conclusive yet is sufficient to warrant further research. Our aim is to conduct a full-scale RCT to determine the clinical and cost effectiveness of acupuncture and counseling compared to usual care alone. We will explore the experiences and perspectives of patients and practitioners. Methods/Design: Randomized controlled trial with three parallel arms: acupuncture plus usual care, counseling plus usual care, and usual care alone, in conjunction with a nested qualitative study using in-depth interviews with purposive samples of trial participants. Participants: Patients aged over 18 years diagnosed with depression or mood disorder by their GP and with a score of 20 or above on the Beck Depression Inventory (BDI-II). Randomization: Computer randomization by York Trials Unit to acupuncture, counseling, and usual care alone in proportions of 2:2:1, respectively, with secure allocation concealment. Interventions: Patients allocated to acupuncture and counseling groups receive the offer of up to 12 weekly sessions. Both interventions allow flexibility to address patient variation, yet are constrained within defined protocols. Acupuncture is based on traditional Chinese medicine and counseling is non-directive within the humanistic tradition. Outcome: The PHQ-9 is the primary outcome measure, collected at baseline, 3, 6, 9, and 12 months. Also measured is BDI-II, SF-36 Bodily pain subscale, and EQ-5D. Texted mood scores are collected weekly over the first 15 weeks. Health-related resource use is collected over 12 months. Analysis: The sample size target was for 640 participants, calculated for an effect size of 0.32 on the PHQ-9 when comparing acupuncture with counseling given 90% power, 5% significance, and 20% loss to follow-up. Analysis of covariance will be used on an intention-to-treat basis. Thematic analysis will be used for qualitative data. We will compare incremental cost-effectiveness of the three treatment options at 12 months. Discussion: Ethical approval was obtained in October 2009. There were six subsequent protocol amendments, the last of which was approved in January 2012. Recruitment of 755 participants took place over 18 months. Data collection will be completed by June 2012. No interim analyses have been conducted
