    Validating design principles for creative collaboration

    Design Science Research (DSR) lacks accepted validation approaches to validate its outcomes (design principles and design solutions). The quality of such a validation has to be ensured so that the outcomes of DSR are as methodologically thorough and as practically relevant as possible. This study introduces a pragmatic validation approach to ensure the quality of design principles. It uses a three-step approach, combining (1) evaluating the design solution as a case in practice, (2) qualifying the implementation of the set of design principles, and (3) assessing the validity of evaluation and qualification results. The validation approach is illustrated by assessing the evaluation results of a creativity facilitation course (design solution) developed according to a set of design principles

    Orally administered 5-aminolevulinic acid for isolation and characterization of circulating tumor-derived extracellular vesicles in glioblastoma patients

    Background: In glioblastoma (GB), tissue is required for accurate diagnosis and subtyping. Tissue can be obtained through resection or (stereotactic) biopsy, but these invasive procedures provide risks for patients. Extracellular vesicles (EVs) are small, cell-derived vesicles that contain miRNAs, proteins, and lipids, and possible candidates for liquid biopsies.     Small molecules to regulate the GH/IGF1 axis by inhibiting the growth hormone receptor synthesis

    Growth hormone (GH) and insulin-like growth factor-1 (IGF1) play an important role in mammalian development, cell proliferation and lifespan. Especially in cases of tumor growth there is an urgent need to control the GH/IGF1 axis. In this study we screened a 38,480-compound library, and in two consecutive rounds of analogues selection, we identified active lead compounds based on the following criteria: inhibition the GH receptor (GHR) activity and its downstream effectors Jak2 and STAT5, and inhibition of growth of breast and colon cancer cells. The most active small molecule (BM001) inhibited both the GH/IGF1 axis and cell proliferation with an IC50 of 10-30 nM of human cancer cells. BM001 depleted GHR in human lymphoblasts. In preclinical xenografted experiments, BM001 showed a strong decrease in tumor volume in mice transplanted with MDA-MB-231 breast cancer cells. Mechanistically, the drug acts on the synthesis of the GHR. Our findings open the possibility to inhibit the GH/IGF1 axis with a small molecule

    A novel antifolate suppresses growth of FPGS-deficient cells and overcomes methotrexate resistance

    Cancer cells make extensive use of the folate cycle to sustain increased anabolic metabolism. Multiple chemotherapeutic drugs interfere with the folate cycle, including methotrexate and 5-fluorouracil that are commonly applied for the treatment of leukemia and colorectal cancer (CRC), respectively. Despite high success rates, therapy-induced resistance causes relapse at later disease stages. Depletion of folylpolyglutamate synthetase (FPGS), which normally promotes intracellular accumulation and activity of natural folates and methotrexate, is linked to methotrexate and 5-fluorouracil resistance and its association with relapse illustrates the need for improved intervention strategies. Here, we describe a novel antifolate (C1) that, like methotrexate, potently inhibits dihydrofolate reductase and downstream one-carbon metabolism. Contrary to methotrexate, C1 displays optimal efficacy in FPGS-deficient contexts, due to decreased competition with intracellular folates for interaction with dihydrofolate reductase. We show that FPGS-deficient patient-derived CRC organoids display enhanced sensitivity to C1, whereas FPGS-high CRC organoids are more sensitive to methotrexate. Our results argue that polyglutamylation-independent antifolates can be applied to exert selective pressure on FPGS-deficient cells during chemotherapy, using a vulnerability created by polyglutamylation deficiency

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Background: Information Technology (IT) has reached a level of maturity, where IT can really meet the complexity of the construction industry. IT has reached the ability to support construction not just any more in the facilitating processes as financial services and human resources management (HR), but also to play a key role in the primary process due to the development of the building information modeling (BIM). However, implementation of BIM in a company is still an challenging exercise. Purpose of this paper: The development of an implementation strategy based on the needs of project support. Method: Desk research gives insights in the roles and phases in a project, levels of hierarchy in a company and two case studies in two large European construction companies deliver insight in the actual situation. Results &amp; Discussion: The complexity of BIM implementation will be presented in a clear analysis of BIM for the different roles, activities, phases of an building project. Moreover a BIM implementation approach will be presented useful for small and large construction companies.</p
