    easySTORM: a robust, lower-cost approach to localisation and TIRF microscopy

    TIRF and STORM microscopy are super-resolving fluorescence imaging modalities for which current implementations on standard microscopes can present significant complexity and cost. We present a straightforward and low-cost approach to implement STORM and TIRF taking advantage of multimode optical fibres and multimode diode lasers to provide the required excitation light. Combined with open source software and relatively simple protocols to prepare samples for STORM, including the use of Vectashield for non-TIRF imaging, this approach enables TIRF and STORM imaging of cells labelled with appropriate dyes or expressing suitable fluorescent proteins to become widely accessible at low cost

    Musculoskeletal ultrasound involves the study of many superficial targets, especially in the hands, wrists, and feet. Many of these areas are within the first 3 cm of the skin surface and are ideal targets for ultra-high-frequency ultrasound. The high spatial resolution and the superb image quality achievable allow foreseeing a wider use of this novel technique, which has the potential to bring innovation to diagnostic imaging

    May Measurement Month (MMM) 2017: an analysis of blood pressure screening results in Bangladesh-South Asia

    Elevated blood pressure (BP) is a growing burden worldwide, leading to over 10 million deaths each year. Based on the findings of the non-communicable disease Risk Factors Survey Bangladesh 2010, the prevalence of hypertension in adults 25 years or older in Bangladesh is 20.1%. The Bangladesh Demographic Health Survey, 2011 showed that approximately 50% of those affected are unaware of their hypertensive condition. The May Measurement Month 2017 (MMM17) is a global initiative of the International Society of Hypertension (ISH) aimed at raising awareness of high BP. We participated in MMM17 to raise awareness of hypertension screening and identify those with elevated BP who were unaware, and those on treatment with still uncontrolled hypertension. Following the standard protocol designed by the ISH, we participated in MMM17, an opportunistic cross-sectional survey of volunteers aged ≥18. It was carried out in May 2017. BP measurement, the definition of hypertension and statistical analysis followed the standard MMM protocol. Data were collected from 35 screening sites in 33 districts in Bangladesh. Personnel from several government and non-government organizations volunteered in this huge event. A total of 11 418 individuals were screened during MMM17, of which 5401 (47.3%) were found to have hypertension. Of 8365 individuals not receiving anti-hypertensive medication, 2348 (28.1%) were hypertensive. Of 3053 individuals receiving anti-hypertensive medication, 1594 (52.2%) had uncontrolled BP. MMM17 was the largest BP screening campaign undertaken in Bangladesh. This study suggests that opportunistic screening can identify significant numbers of people with raised BP. A periodic public health programme at a national level needs to be initiated to increase hypertension detection and control rate and thus for the prevention of cardiovascular diseases

    Reconstructing historic Glacial Lake Outburst Floods through numerical modelling and geomorphological assessment:Extreme events in the Himalaya

    Recession of high‐mountain glaciers in response to climatic change frequently results in the development of moraine‐dammed glacial lakes. Moraine dam failure is often accompanied by the release of large volumes of water and sediment, termed a Glacial Lake Outburst Flood (GLOF). Chukhung Glacier is a small (~3 km2) receding valley glacier in Mt. Everest (Sagarmatha) National Park, Nepal. Unlike many Himalayan glaciers, which possess a thick mantle of supraglacial debris, its surface is relatively clean. The glacier terminus has receded 1.3 km from its maximum Holocene position, and in doing so provided the space for an ice‐contact moraine‐dammed lake to develop. The lake had a maximum volume of 5.5 × 105 m3 and drained as a result of breaching of the terminal moraine. An estimated 1.3 × 105 m3 of material was removed from the terminal moraine during breach development. Numerical dam‐breach modelling, implemented within a Generalised Likelihood Uncertainty Estimation (GLUE) framework, was used to investigate a range of moraine‐dam failure scenarios. Reconstructed outflow peak discharges, including failure via overtopping and piping mechanisms, are in the range 146–2200 m3 s‐1. Results from two‐dimensional hydrodynamic GLOF modelling indicate that maximum local flow depths may have exceeded 9 m, with maximum flow velocities exceeding 20 m s‐1 within 700 m of the breach. The floodwaters mobilised a significant amount of material, sourced mostly from the expanding breach, forming a 300 m long and 100 m wide debris fan originating at the breach exit. moraine‐dam. These results also suggest that inundation of the entire floodplain may have been achieved within ten minutes of initial breach development, suggesting that debris fan development was rapid. We discuss the key glaciological and geomorphological factors that have determined the evolution of a hazardous moraine‐dammed lake complex and the subsequent generation of a GLOF and its geomorphological impact

    Superconformal Flavor Simplified

    A simple explanation of the flavor hierarchies can arise if matter fields interact with a conformal sector and different generations have different anomalous dimensions under the CFT. However, in the original study by Nelson and Strassler many supersymmetric models of this type were considered to be 'incalculable' because the R-charges were not sufficiently constrained by the superpotential. We point out that nearly all such models are calculable with the use of a-maximization. Utilizing this, we construct the simplest vector-like flavor models and discuss their viability. A significant constraint on these models comes from requiring that the visible gauge couplings remain perturbative throughout the conformal window needed to generate the hierarchies.
