    Pulmonary hypertension is called PH for short. It is caused by the pulmonary artery vascular disease leading to pulmonary vascular resistance, and the increase right lung compartment load, which resulting in weakening or even collapse of the right ventricular function. The establishment of rat PH model under the action of monocrotaline is a repeatable, simple and accessible operation technique, which has been widely used in the treatment of pulmonary hypertension.     Combining the Tyrosine Kinase Inhibitor Cabozantinib and the mTORC1/2 Inhibitor Sapanisertib Blocks ERK Pathway Activity and Suppresses Tumor Growth in Renal Cell Carcinoma.

    UNLABELLED: Current treatment approaches for renal cell carcinoma (RCC) face challenges in achieving durable tumor responses due to tumor heterogeneity and drug resistance. Combination therapies that leverage tumor molecular profiles could offer an avenue for enhancing treatment efficacy and addressing the limitations of current therapies. To identify effective strategies for treating RCC, we selected ten drugs guided by tumor biology to test in six RCC patient-derived xenograft (PDX) models. The multitargeted tyrosine kinase inhibitor (TKI) cabozantinib and mTORC1/2 inhibitor sapanisertib emerged as the most effective drugs, particularly when combined. The combination demonstrated favorable tolerability and inhibited tumor growth or induced tumor regression in all models, including two from patients who experienced treatment failure with FDA-approved TKI and immunotherapy combinations. In cabozantinib-treated samples, imaging analysis revealed a significant reduction in vascular density, and single-nucleus RNA sequencing (snRNA-seq) analysis indicated a decreased proportion of endothelial cells in the tumors. SnRNA-seq data further identified a tumor subpopulation enriched with cell-cycle activity that exhibited heightened sensitivity to the cabozantinib and sapanisertib combination. Conversely, activation of the epithelial-mesenchymal transition pathway, detected at the protein level, was associated with drug resistance in residual tumors following combination treatment. The combination effectively restrained ERK phosphorylation and reduced expression of ERK downstream transcription factors and their target genes implicated in cell-cycle control and apoptosis. This study highlights the potential of the cabozantinib plus sapanisertib combination as a promising treatment approach for patients with RCC, particularly those whose tumors progressed on immune checkpoint inhibitors and other TKIs. SIGNIFICANCE: The molecular-guided therapeutic strategy of combining cabozantinib and sapanisertib restrains ERK activity to effectively suppress growth of renal cell carcinomas, including those unresponsive to immune checkpoint inhibitors

    Baichuan 2: Open Large-scale Language Models

    Large language models (LLMs) have demonstrated remarkable performance on a variety of natural language tasks based on just a few examples of natural language instructions, reducing the need for extensive feature engineering. However, most powerful LLMs are closed-source or limited in their capability for languages other than English. In this technical report, we present Baichuan 2, a series of large-scale multilingual language models containing 7 billion and 13 billion parameters, trained from scratch, on 2.6 trillion tokens. Baichuan 2 matches or outperforms other open-source models of similar size on public benchmarks like MMLU, CMMLU, GSM8K, and HumanEval. Furthermore, Baichuan 2 excels in vertical domains such as medicine and law. We will release all pre-training model checkpoints to benefit the research community in better understanding the training dynamics of Baichuan 2.Comment: Baichuan 2 technical report.     Normative modeling of brain morphometry in clinical high risk for psychosis

    Importance The lack of robust neuroanatomical markers of psychosis risk has been traditionally attributed to heterogeneity. A complementary hypothesis is that variation in neuroanatomical measures in individuals at psychosis risk may be nested within the range observed in healthy individuals. Objective To quantify deviations from the normative range of neuroanatomical variation in individuals at clinical high risk for psychosis (CHR-P) and evaluate their overlap with healthy variation and their association with positive symptoms, cognition, and conversion to a psychotic disorder. Design, Setting, and Participants This case-control study used clinical-, IQ-, and neuroimaging software (FreeSurfer)–derived regional measures of cortical thickness (CT), cortical surface area (SA), and subcortical volume (SV) from 1340 individuals with CHR-P and 1237 healthy individuals pooled from 29 international sites participating in the Enhancing Neuroimaging Genetics Through Meta-analysis (ENIGMA) Clinical High Risk for Psychosis Working Group. Healthy individuals and individuals with CHR-P were matched on age and sex within each recruitment site. Data were analyzed between September 1, 2021, and November 30, 2022. Main Outcomes and Measures For each regional morphometric measure, deviation scores were computed as z scores indexing the degree of deviation from their normative means from a healthy reference population. Average deviation scores (ADS) were also calculated for regional CT, SA, and SV measures and globally across all measures. Regression analyses quantified the association of deviation scores with clinical severity and cognition, and 2-proportion z tests identified case-control differences in the proportion of individuals with infranormal (z < −1.96) or supranormal (z > 1.96) scores. Results Among 1340 individuals with CHR-P, 709 (52.91%) were male, and the mean (SD) age was 20.75 (4.74) years. Among 1237 healthy individuals, 684 (55.30%) were male, and the mean (SD) age was 22.32 (4.95) years. Individuals with CHR-P and healthy individuals overlapped in the distributions of the observed values, regional z scores, and all ADS values. For any given region, the proportion of individuals with CHR-P who had infranormal or supranormal values was low (up to 153 individuals [<11.42%]) and similar to that of healthy individuals (<115 individuals [<9.30%]). Individuals with CHR-P who converted to a psychotic disorder had a higher percentage of infranormal values in temporal regions compared with those who did not convert (7.01% vs 1.38%) and healthy individuals (5.10% vs 0.89%). In the CHR-P group, only the ADS SA was associated with positive symptoms (β = −0.08; 95% CI, −0.13 to −0.02; P = .02 for false discovery rate) and IQ (β = 0.09; 95% CI, 0.02-0.15; P = .02 for false discovery rate). Conclusions and Relevance In this case-control study, findings suggest that macroscale neuromorphometric measures may not provide an adequate explanation of psychosis risk

    Scientific Computing with Diffractive Optical Neural Networks

    Diffractive optical neural networks (DONNs) are emerging as high‐throughput and energy‐efficient hardware platforms to perform all‐optical machine learning (ML) in machine vision systems. However, the current demonstrated applications of DONNs are largely image classification tasks, which undermine the prospect of developing and utilizing such hardware for other ML applications. Herein, the deployment of an all‐optical reconfigurable DONNs system for scientific computing is demonstrated numerically and experimentally, including guiding two‐dimensional quantum material synthesis, predicting the properties of two‐dimensional quantum materials and small molecular cancer drugs, predicting the device response of nanopatterned integrated photonic power splitters, and the dynamic stabilization of an inverted pendulum with reinforcement learning. Despite a large variety of input data structures, a universal feature engineering approach is developed to convert categorical input features to images that can be processed in the DONNs system. The results open up new opportunities for employing DONNs systems for a broad range of ML applications

    The modulation of selectivity of highly reactive carbon radical cross-coupling for the construction of C-C bonds represents a challenging task in organic chemistry. N-Heterocyclic carbenes (NHCs) catalyzed radical transformations opened a new avenue for acyl radical cross-coupling chemistry. With this method, highly selective cross-coupling of acyl radical with alkyl radical for efficient construction of C-C bonds were succussfully realized. However, the cross-coupling reaction of acyl radical with vinyl radicals represents an uncharted domain. We herein describe NHCs and photocatalysis co-catalyzed radical 1,4-sulfonylacylation of 1,3-enynes, providing structurally diversified valuable tetrasubstituted allenyl ketones. Mechanistic studies indicated that ketyl radicals are formed from aroyl fluorides via oxidative quenching process of excited photocatalysis, allenyl radicals are generated from chemo specific sulfonyl radical addition to the 1,3-enynes, finally, unprecedented key allenyl and ketyl radical cross-coupling provides tetrasubstituted allenyl ketones

    A Novel Multiport Hybrid Wave Energy System for Grid-Connected and Off-Grid Applications

    Direct drive wave energy converters (DDWECs) have gradually become the mainstream of wave energy converters (WECs). In order to make better use of wave energy, energy storage devices and other renewable energy sources are often used to suppress power fluctuation in DDWECs. However, the addition of other energy sources will increase the complexity of the converter system and the number of power switches. Considering the flexibility of nine-switch converters (NSCs), this paper proposes a novel nine-switch grid-connected/off-grid multiport hybrid wave energy system (HWES). First, the system structure and modulation principle are described. Then, a model for a generator, a grid and energy storage are built, including a control strategy of each part. Finally, a simulation for the grid-connected/off-grid application and an experiment on the off-grid HWES are carried out. The results show that the multiport wave energy system can achieve the objective of stable and reliable power transmission by reducing power devices
