    This essay details a 3D simulation of a number of control methods used for maneuvering of teleoperated USAR robots. The implementation was produced in the Unity3D engine. The simulation implemented different variations on field-ofview angle, turning algorithms, and camera view perspectives. An evaluation using volunteer test operators was conducted and discussed. The sample size was too small to draw any definitive conclusions. Further testing is advised.Denna uppsats behandlar en 3D-simulering samt användartester av flera olika kontrollmetoder som används vid fjärrstyrning av obemannade räddningsrobotar. Implementationen skapades med Unity3D-plattformen. De styrmetoder som testades var olika stora synfältsvinklar på kameran, olika algoritmer för att styra robotens svängning, samt olika kameraperspektiv.     Study of the cytological features of bone marrow mesenchymal stem cells from patients with neuromyelitis optica.

    Neuromyelitis optica (NMO) is a refractory autoimmune inflammatory disease of the central nervous system without an effective cure. Autologous bone marrow‑derived mesenchymal stem cells (BM‑MSCs) are considered to be promising therapeutic agents for this disease due to their potential regenerative, immune regulatory and neurotrophic effects. However, little is known about the cytological features of BM‑MSCs from patients with NMO, which may influence any therapeutic effects. The present study aimed to compare the proliferation, differentiation and senescence of BM‑MSCs from patients with NMO with that of age‑ and sex‑matched healthy subjects. It was revealed that there were no significant differences in terms of cell morphology or differentiation capacities in the BM‑MSCs from the patients with NMO. However, in comparison with healthy controls, BM‑MSCs derived from the Patients with NMO exhibited a decreased proliferation rate, in addition to a decreased expression of several cell cycle‑promoting and proliferation‑associated genes. Furthermore, the cell death rate increased in BM‑MSCs from patients under normal culture conditions and an assessment of the gene expression profile further confirmed that the BM‑MSCs from patients with NMO were more vulnerable to senescence. Platelet‑derived growth factor (PDGF), as a major mitotic stimulatory factor for MSCs and a potent therapeutic cytokine in demyelinating disease, was able to overcome the decreased proliferation rate and increased senescence defects in BM‑MSCs from the patients with NMO. Taken together, the results from the present study have enabled the proposition of the possibility of combining the application of autologous BM‑MSCs and PDGF for refractory and severe patients with NMO in order to elicit improved therapeutic effects, or, at the least, to include PDGF as a necessary and standard growth factor in the current in vitro formula for the culture of NMO patient‑derived BM‑MSCs

    Root tuber extract of Polygooum cillinerve (Nakai) Ohwi exerts antimicrobial and immunomodulatory activities

    Purpose: To investigate the antibacterial and immunomodulatory activities of the root tuber extract of Polygonum ciliinerve (Nakai) ohwi, a traditional Chinese medicine.Methods: Minimum inhibitory concentrations (MICs) of root tuber extract of Polygonum ciliinerve (Nakai) ohwi (rPC) and the synergistic effects between rPC and antibiotics were evaluated by broth dilution. In vitro adhesion and invasion assays were used to determine the effect of rPC on bacterial adhesion and invasion. Furthermore, the immunomodulatory effects of rPC were assessed by western blot.Results: rPC treatment inhibited the growth of Staphylococcus aureus (MIC = 12 mg/ml) and Escherichia coli (MIC = 64 mg/mL). rPC also showed synergistic effects with penicillin (fractional inhibitory concentration, FIC = 0.45), vancomycin (FIC = 0.333), moxifloxacin (FIC = 0.25), and levofloxacin (FIC = 0.356). The adhesion and invasion of bacteria were also suppressed by rPC treatment. Moreover, rPC exhibited an immunomodulatory effect during bacterial infection.Conclusion: rPC shows antibacterial and immunomodulatory activites when assessed by multiple methods, and therefore is a potential therapeutic alternative agent for the treatment of bacterial infections.Keywords: Polygonum ciliinerve root tuber, Synergistic effect, Immunomodulatory, Antibacteria

    Hepatitis B virus infection and replication in a new cell culture system established by fusing HepG2 cells with primary human hepatocytes

    BackgroundHepatitis B virus (HBV) infection is strictly species and tissue specific, therefore none of the cell models established previously can reproduce the natural infection process of HBV in vitro. The aim of this study was to establish a new cell line that is susceptible to HBV and can support the replication of HBV.MethodsA hybrid cell line was established by fusing primary human hepatocytes with HepG2 cells. The hybrid cells were incubated with HBV-positive serum for 12 hours. HBV DNA was detected by quantitative fluorescence polymerase chain reaction (QF-PCR). HBsAg (surface antigen) and HBeAg (extracellular form of core antigen) were observed by electrochemiluminescence (ECL). HBcAg (core antigen) was detected by the indirect immunofluorescence technique. HBV covalently closed circular DNA (cccDNA) was analyzed by Southern blot hybridization and quantified using real-time PCR.ResultsA new cell line was established and named HepCHLine-7. The extracellular HBV DNA was observed from Day 2 and the levels ranged from 9.80 (± 0.32) × 102 copies/mL to 3.12 (± 0.03) × 104 copies/mL. Intracellular HBV DNA was detected at Day 2 after infection and the levels ranged from 7.92 (± 1.08) × 103 copies/mL to 5.63 (± 0.11) × 105 copies/mL. HBsAg in the culture medium was detected from Day 4 to Day 20. HBeAg secretion was positive from Day 5 to Day 20. HBcAg constantly showed positive signals in approximately 20% (± 0.82%) of hybrid cells. Intracellular HBV cccDNA could be detected as early as 2 days postinfection and the highest level was 15.76 (± 0.26) copies/cell.ConclusionHepCHLine-7 cells were susceptible to HBV and supported the replication of HBV. They are therefore suitable for studying the complete life cycle of HBV
