    Stability Testing of Beclomethasone Dipropionate Nanoemulsion

    Purpose: To perform stability studies on a nanoemulsion formulation containing beclomethasone dipropionate (BD) and prepared by spontaneous emulsification method.Method: A nanoemulsion (o/w) containing BD was prepared using eucalyptus oil, Tween-40, ethanol and distilled water. The nanoemulsions were characterized by droplet size, pH, viscosity, conductivity and refractive index. Stability studies were performed according to International Council on Harmonization (ICH) guidelines over a period of 3 months. Droplet size, pH, viscosity, conductivity and refractive index were determined monthly for 3 months. The shelf-life of the nanoemulsion formulation was determined by accelerated stability testing.Results: The droplet size, conductivity, viscosity, pH and refractive index of the optimized formulations did not change significantly (p ≥ 0.05) after 3 months of storage at room temperature (25 ºC).     Suitability of using palm oil mill effluent as a medium for lipase production

    Lipases are enzymes that can be secreted by several microorganisms using agro-industrial residues as potential substrates. This work screened ten microorganisms for their potential to produce lipase in palm oil mill effluent (POME)-based medium. Among the 10 organisms, the most promising strain was Candida cylindracea (ATCC 14830) which showed appreciable activity both on agar plates and liquid cultures. Medium supplementation by NH4Cl and olive oil led to an enzyme activity of 2.07 U/ml. However, supplementation with organic nitrogen sources resulted in better enzyme activity. Addition of malt extract, peptone and olive oil into the medium greatly influenced the lipase production. Among the oils that were tested, olive oil was found to be the best for the expression of extracellular lipase at 0.5% (v/v) with an activity of 4.02 U/ml in an optimized POME supplemented medium.Key words: Lipase, agro-industrial residue, palm oil mill effluent, Candida cylindrace

    FedRolex: Model-Heterogeneous Federated Learning with Rolling Sub-Model Extraction

    Most cross-device federated learning (FL) studies focus on the model-homogeneous setting where the global server model and local client models are identical. However, such constraint not only excludes low-end clients who would otherwise make unique contributions to model training but also restrains clients from training large models due to on-device resource bottlenecks. In this work, we propose FedRolex, a partial training (PT)-based approach that enables model-heterogeneous FL and can train a global server model larger than the largest client model. At its core, FedRolex employs a rolling sub-model extraction scheme that allows different parts of the global server model to be evenly trained, which mitigates the client drift induced by the inconsistency between individual client models and server model architectures. We show that FedRolex outperforms state-of-the-art PT-based model-heterogeneous FL methods (e.g. Federated Dropout) and reduces the gap between model-heterogeneous and model-homogeneous FL, especially under the large-model large-dataset regime. In addition, we provide theoretical statistical analysis on its advantage over Federated Dropout and evaluate FedRolex on an emulated real-world device distribution to show that FedRolex can enhance the inclusiveness of FL and boost the performance of low-end devices that would otherwise not benefit from FL.     Prolonged heating of honey increases its antioxidant potential but decreases its antimicrobial activity

    Background: Honey is renowned for its antioxidant, antimicrobial and medicinal properties. It is typically heated and filtered during processing to avoid crystallization. However, few studies have reported its heating effect and the antioxidant and antimicrobial activities of honey.Materials and Methods: In this study, honey samples were collected from eight different honey-producing sites in Bangladesh and heated to 50ºC, 70ºC and 90ºC for 12 hours, while the antioxidant potentials and antimicrobial properties of the heated samples were compared with those of the unheated samples. Antimicrobial properties were investigated against one Gram-positive and eight Gram-negative bacteria. Other physicochemical and biochemical properties were also determined.Results: Interestingly, there was a proportionate increase in the amount of antioxidant compounds (phenolics and flavonoids) as well as antioxidant potentials proportional with the amount of heat introduced, whereas the antimicrobial properties of the honey samples were reduced with increasing heat.Conclusion: In conclusion, though prolonged heat treatment of honey can increase its antioxidant potential, antimicrobial activities are compromised.Keywords: Honey, heat, antioxidant, antimicrobial

    GPT-FL: Generative Pre-trained Model-Assisted Federated Learning

    In this work, we propose GPT-FL, a generative pre-trained model-assisted federated learning (FL) framework. At its core, GPT-FL leverages generative pre-trained models to generate diversified synthetic data. These generated data are used to train a downstream model on the server, which is then fine-tuned with private client data under the standard FL framework. We show that GPT-FL consistently outperforms state-of-the-art FL methods in terms of model test accuracy, communication efficiency, and client sampling efficiency. Through comprehensive ablation analysis, we discover that the downstream model generated by synthetic data plays a crucial role in controlling the direction of gradient diversity during FL training, which enhances convergence speed and contributes to the notable accuracy boost observed with GPT-FL. Also, regardless of whether the target data falls within or outside the domain of the pre-trained generative model, GPT-FL consistently achieves significant performance gains, surpassing the results obtained by models trained solely with FL or synthetic data

    Sub-nanosecond signal propagation in anisotropy engineered nanomagnetic logic chains

    Energy efficient nanomagnetic logic (NML) computing architectures propagate and process binary information by relying on dipolar field coupling to reorient closely-spaced nanoscale magnets. Signal propagation in nanomagnet chains of various sizes, shapes, and magnetic orientations has been previously characterized by static magnetic imaging experiments with low-speed adiabatic operation; however the mechanisms which determine the final state and their reproducibility over millions of cycles in high-speed operation (sub-ns time scale) have yet to be experimentally investigated. Monitoring NML operation at its ultimate intrinsic speed reveals features undetectable by conventional static imaging including individual nanomagnetic switching events and systematic error nucleation during signal propagation. Here, we present a new study of NML operation in a high speed regime at fast repetition rates. We perform direct imaging of digital signal propagation in permalloy nanomagnet chains with varying degrees of shape-engineered biaxial anisotropy using full-field magnetic soft x-ray transmission microscopy after applying single nanosecond magnetic field pulses. Further, we use time-resolved magnetic photo-emission electron microscopy to evaluate the sub-nanosecond dipolar coupling signal propagation dynamics in optimized chains with 100 ps time resolution as they are cycled with nanosecond field pulses at a rate of 3 MHz. An intrinsic switching time of 100 ps per magnet is observed. These experiments, and accompanying macro-spin and micromagnetic simulations, reveal the underlying physics of NML architectures repetitively operated on nanosecond timescales and identify relevant engineering parameters to optimize performance and reliability.Comment: Main article (22 pages, 4 figures), Supplementary info (11 pages, 5 sections

    Obesity, Disability and Self-Perceived Health Outcomes in Australian Adults: A Longitudinal Analysis Using 14 Annual Waves of the HILDA Cohort.

    Background: Both obesity and disability have been widely recognised as major public health challenges because they play significant roles in determining self-perceived general and mental health. Longitudinal studies of the relationship between obesity and disability with self-reported health outcomes are scarce. Therefore, the objective of the present study is to examine the relationship between obesity and disability with self-perceived general and mental health among Australian adults aged 15 years and above. Methods: Data were extracted from the most recent 14 waves (waves 6 through 19) of the annual individual person dataset of the Household, Income and Labour Dynamics in Australia (HILDA) survey. The longitudinal random-effects logistic regression model was adopted to investigate the relationships between obesity and disability with self-reported health outcomes. Results: The results revealed that obese individuals and adults with some form of disability are more likely to report poor or fair general and mental health. The odds of self-reporting poor or fair general health were 2.40 and 6.07 times higher among obese (aOR: 2.40, 95% CI: 2.22-2.58) and adults with some form of disability (aOR: 6.07, 95% CI: 5.77-6.39), respectively, relative to adults with healthy weight and those without disability . The results also showed that self-rated poor or fair mental health were 1.22 and 2.40 times higher among obese adults (aOR: 1.22, 95% CI: 1.15-1.30) and adults with disability (aOR: 2.40, 95% CI: 2.30-2.51), respectively, compared to their healthy weight peers and peers without disability. Conclusion: As governmental and non-governmental organisations seek to improve the community's physical and mental well-being, these organisations need to pay particular attention to routine health care prevention, specific interventions, and treatment practices, especially for obese and/or people with disabilities

    Crystal structures of murine norovirus-1 RNA-dependent RNA polymerase in complex with 2-thiouridine or ribavirin

    AbstractMurine norovirus-1 (MNV-1) shares many features with human norovirus (HuNoV) and both are classified within the norovirus genus of Caliciviridae family. MNV-1 is used as the surrogate for HuNoV research since it is the only form that can be grown in cell culture. HuNoV and MNV-1 RNA dependent RNA polymerase (RdRp) proteins with the sequence identity of 59% show essentially identical conformations. Here we report the first structural evidence of 2-thiouridine (2TU) or ribavirin binding to MNV-1 RdRp, based on the crystal structures determined at 2.2Å and 2.5Å resolutions, respectively. Cellular and biochemical studies revealed stronger inhibitory effect of 2TU on the replication of MNV-1 in RAW 264.7 cells, compared to that of ribavirin. Our complex structures highlight the key interactions involved in recognition of the nucleoside analogs which block the active site of the viral RNA polymerase
