    Biotin-avidin sandwich elisa with specific human isotypes IgG1 and IgG4 for Culicidae mosquito blood meal identification from an epizootic yellow fever area in Brazil

    With a view toward investigating the feeding behavior of Culicidae mosquitoes from an area of epizootic yellow fever transmission in the municipalities of Garruchos and Santo Antônio das Missões, Rio Grande do Sul State, Brazil, specimens were collected by aspiration from September 2005 to April 2007. The engorged females were submitted to blood meal identification by enzyme-linked immunosorbent assay (ELISA). A total of 142 blood-engorged samples were examined for human or monkey blood through species-specific IgG. Additional tests for specificity utilizing isotypes IgG1 and IgG4 of human monoclonal antibodies showed that only anti-human IgG1 was effective in recognizing blood meals of human origin. The results indicated a significant difference (p = 0.027) in detection patterns in samples of Haemagogus leucocelaenus recorded from human blood meals at Santo Antônio das Missões, which suggests some degree of exposure, since it was an area where epizootic outbreaks have been reported.PAHO Pan American Health Organization - Secretary of Health Surveillance, Brazilian Ministry of Healt

    Food Quality and Nutritional Profile of Students from a Public School in Brazil

    Feeding is directly related to nutritional status and alterations as obesity and malnutrition, affecting human body and increasing the risk of morbidity and mortality. This study aimed to evaluate the nutritional status of children and adolescents in a public school located in Uberaba MG Brazil by comparing macro and micronutrients on the school menu with PNAE (Brazilian government program) recommendations. A descriptive cross-sectional study including 547 individuals was carried out. Nutritional assessment was done based on anthropometric Z-score analysis. Only the food ingestion made at the school was considered, in order to verify if the meals offered by the school meet the percentage of nutrients recommended by PNAE. With regard to the nutritional status, there was a small proportion of students (0.18%) with low weight (-3 ? Z < -2). There was 15% prevalence of overweight (overweight, obesity, and severe obesity) among students. Menus composition revealed that macronutrients, fiber, and energy intake was lower than that recommended by PNAE for all age groups studied. The prevalence of a considerable number of overweight students is a warning for interventions to prevent obesity.     Seronegative patients vaccinated with cytomegalovirus gB-MF59 vaccine have evidence of neutralising antibody responses against gB early post-transplantation

    Background Human cytomegalovirus (HCMV) causes a ubiquitous infection which can pose a significant threat for immunocompromised individuals, such as those undergoing solid organ transplant (SOT). Arguably, the most successful vaccine studied to date is the recombinant glycoprotein-B (gB) with MF59 adjuvant which, in 3 Phase II trials, demonstrated 43–50% efficacy in preventing HCMV acquisition in seronegative healthy women or adolescents and reduction in virological parameters after SOT. However, the mechanism of vaccine protection in seronegative recipients remains undefined. Methods We evaluated samples from the cohort of seronegative SOT patients enroled in the Phase II glycoprotein-B/MF59 vaccine trial who received organs from seropositive donors. Samples after SOT (0–90 days) were tested by real-time quantitative PCR for HCMV DNA. Anti-gB antibody levels were measured by ELISA. Neutralization was measured as a decrease in infectivity for fibroblast cell cultures revealed by expression of immediate-early antigens. Findings Serological analyses revealed a more rapid increase in the humoral response against gB post transplant in vaccine recipients than in those randomised to receive placebo. Importantly, a number of patient sera displayed HCMV neutralising responses – neutralisation which was abrogated by pre-absorbing the sera with recombinant gB. Interpretation We hypothesise that the vaccine primed the immune system of seronegative recipients which, when further challenged with virus at time of transplant, allowed the host to mount rapid immunological humoral responses even under conditions of T cell immune suppression during transplantation

    Electron Spin Resonance and Thermoluminescence dating of shells and sediments from Sambaqui (shell mound) Santa Marta II, Brazil

    In Tupi, the word Sambaqui means “mound of shells”. These archaeological sites are cultural vestiges left by the prehistoric occupation of the Brazilian coast from five to six thousand years ago. Mollusks, fishes, and other marine edible foods were important for the survival of this population. The remains of foods, mainly shells, were heaped up, giving a mound of different proportions, which became part of the landscape of the Brazilian coastal plain. Due to the large number of Sambaquis in Brazil and considering that Sambaqui Santa Marta II, Laguna, SC, has not yet been dated, Electron Spin Resonance (ESR) measurements were performed in aragonite shells collected from different layers of Sambaqui Santa Marta II, starting from the base to the center of the Sambaqui. Radiocarbon analysis by Accelerator Mass Spectrometry (AMS) at Beta Analytic laboratory were also performed for comparison with ESR results. Before measurements, shells were chemically etched, after drying, were pulverized and sieved. The sediments were separated into small portions which were irradiated with gamma radiation from a 60Co source with doses from 5 up to 120 Gy. Ages around 2,000 to 4,000 years have been obtained. The results obtained are consistent with the dates of others Sambaquis of the region, possibly were built at the same time

    The formation of the coastal plain of the Brazilian sea is mainly due to the fluctuation of relative sea level in the past. Armação dos Buzios or simply Buzios is a municipality in the microregion of lakes, in the state of Rio de Janeiro. In this region there is a lowland area about two meters above current sea level. This lowland areais also known as coastal plain of Una River. It is expected that during the Holocene period the sea level reached a maximum about 2.5 meters above the current level. During that time billions of mollusks lived and proliferated in the shallow waters around the coastal plain of the Una River. As they died their shells formed a layer in the soil including Malhada Marsh that belongs to the Una River plain. In this study, shells were collected from this region and dated using the techniques of Electronic Paramagnetic Resonance (EPR). Ages from 2480±130 to 4490±270 years were obtained. Radiocarbon dating were performed at the Beta Analytic Lab, USA for comparison with the EPR results. Shells ages obtained are compatible with geological data from the Holocene period relative to past sea level fluctuations

    Obstacle Avoidance Using Echo Sounder Sonar

    Developing obstacle avoidance algorithms for low cost Autonomous Underwater Vehicles using single beam returned echo sounder sonar is a difficult task. In this paper, we propose an intelligent obstacle avoidance algorithm that maps the obstacle, avoids it with a guarantee that it will not get stuck and efficiently traverses a path towards the destination using navigation functions. We present a complete obstacle avoidance system with the help of hybrid automata, probabilistic mapping and navigation functions. Simulation results are presented showing the validity of our approach

    <p>Abstract</p> <p>Background</p> <p>Aspirin esterase (AE) activity can account for part of aspirin pharmacokinetics in the circulation, possibly being associated with the impairment of aspirin effectiveness as an inhibitor of platelet aggregation.</p> <p>Aims</p> <p>The study was aimed at investigating the correlations of serum AE activity with cholinesterase (ChE) and metabolic variables in healthy subjects in comparison to subjects with type 2 diabetes mellitus (T2DM).</p> <p>Methods</p> <p>In cardiovascular disease-free T2DM subjects and healthy controls, the AE activity levels and/or the correlation patterns between AE and the other variables were analyzed.</p> <p>Results</p> <p>Neither AE nor ChE activities were higher in the subjects with T2DM. Serum AE activity strongly correlated with ChE as well as glucose/lipids variables such as total cholesterol and triglyceride in healthy subjects, while the correlations between AE and glucose/lipids variables were not present in T2DM subjects.</p> <p>Conclusions</p> <p>These data may reflect the pathophysiological changes between healthy and T2DM subjects. Our data may thus provide the basis for future studies to unravel the mechanisms.</p
