    Objectifs : l'exploration subjective de l'odorat utilise des tests variables d'un pays Ă  l'autre en fonction de traits culturels. Notre objectif est de mettre au point des outils de reprĂ©sentation graphique et d'analyse d'un test semi-quantitatif rĂ©alisĂ© en France, le test Biolfa. MĂ©thode : Nous avons repris les 131 tests olfactomĂ©triques rĂ©alisĂ©s dans le service d'oto-rhino-laryngologie du CHU de Brest entre avril 2006 et mai 2008. Nous avons utilisĂ© une reprĂ©sentation en radar du test pour effectuer une analyse synthĂ©tique. La discordance entre les seuils de perception et de reconnaissance des odeurs a Ă©tĂ© appelĂ©e " Rinne olfactif ". RĂ©sultats : Il n'existe pas de profil olfactomĂ©trique type des principales Ă©tiologies responsables de trouble de l'odorat. Un outil de mesure quantitative de "Rinne olfactif " a Ă©tĂ© mis au point.     L'implant cochlĂ©aire a bouleversĂ© le pronostic fonctionnel des patients hypoacousiques sĂ©veres. Les vertiges font partie des plaintes post-opĂ©ratoires couramment exprimĂ©es. Le rĂŽle de l'implant cochlĂ©aire dans ces symptĂŽmes et dans la physiologie vestibulaire est mal connu. Notre travail, grace a une Ă©tude menĂ©e Ă  long terme aprĂ©s implantation, a permis de mesurer les plaintes des patients implantĂ©s cochlĂ©aires. Les plaintes spĂ©cifiquement exprimĂ©es concernant le fonctionement de l'implant, sont frĂ©quentes mais modĂ©rĂ©es. Nous avons pu, pour les patients les plus symptomatiques, corrĂ©ler ces plaintes avec une dĂ©viation de la verticale visuelle subjective lors de l extinction de l implant. Par ailleurs, nousdĂ©crivons l'existence de potentiels Ă©voquĂ©s myogĂ©niques obtenus par stimulation de l'implant. L existence de tels potentiels n est corrĂ©lĂ©e Ă  aucune plainte fonctionnelle. Au total, la fonction vestibulaire de base ou la rĂ©ponse Ă  une stimulation par l'implant n'a aucun caractĂ©re prĂ©dictif des plaintes fonctionnelles. En revanche, il semble que les patients les plus vertigineux et les moins bien compensĂ©s soient plus sensibles aux stimulations par l implant. Ces observations accrĂ©ditent l idĂ©e d une stimulation frĂ©quente, de faible intensitĂ©, du nerf vestibulaire par l implant. Cette stimulation semble soumise Ă  compensation et n ĂȘtre symptomatique qu'en cas de fluctuation de la fonction vestibulaire.BREST-BU MĂ©decine-Odontologie (290192102) / SudocSudocFranceF

    Perception of Odors and Tastes in Autism Spectrum Disorders: A Systematic Review of Assessments

    International audienceOlfaction and gustation are major sensory functions implied in processing environmental stimuli. Some evidences suggest that loss of olfactory function is an early biomarker for neurodegenerative disorders and atypical processing of odor and taste stimuli is present in several neurodevelopmental disorders, notably in Autism Spectrum Disorders (ASD). In this paper, we conducted a systematic review investigating the assessments of olfaction and gustation with psychophysics methods in individuals with ASD. Pubmed, PMC and Sciencedirect were scrutinized for relevant literature published from 1970 to 2015. In this review, fourteen papers met our inclusion criteria. They were analyzed critically in order to evaluate the occurrence of olfactory and gustatory dysfunction in ASD, as well as to report the methods used to assess olfaction and gustation in such conditions. Regarding to these two senses, the overall number of studies is low. Most of studies show significant difference regarding to odor or taste identification but not for detection threshold. Overall, odor rating through pleasantness, intensity and familiarity do not differ significantly between control and individuals with ASD. The current evidences can suggest the presence of olfactory and gustatory dysfunction in ASD. Therefore, our analysis show a heterogeneity of findings. This is due to several methodological limitations such as the tools used or population studied. Understanding these disorders could help to shed light on other atypical behavior in this population such as feeding or social behavior

    Development of a Murine model to dissect the CpG-oligonucleotide-enhancement of the killing of human B Cells by rituximab.

    International audienceAs a model to dissect the effects of CpG-oligonucleotides (CpG) on rituximab (RTX)-mediated therapeutic killing of autoimmune or malignant B lymphocytes, nude mice were grafted with Daudi human B cells. These mice were then injected with RTX alone or together with CpG. The human B cell aggregate was measured, and the reactive infiltrate analyzed after selective depletion of murine circulating cells. Macrophages (MØ) were identified in infiltrates, but not polymorphonuclear neutrophils (PMN), as confirmed by the failure of quantitative polymerase chain reaction to detect transcripts for PMN-specific myeloperoxidase in graft extracts. Evidence that MØ predominate over PMN in the anti-B cell RTX-induced immune mechanisms, include the presence of MØ-derived cytokines, and the lack of consequences of depletion of NK cells or B lymphocytes on the CpG-mediated effects on RTX. Interestingly however, removal of circulating PMN reduced the number of MØ attracted by the Daudi B cells. Our interpretation that CpG-induced complement activation is required for PMN to influence MØ was first based on overproduction of C5a in treated mice. This excess was due to the binding of the inhibitor of the alternative pathway of complement to CpG, as demonstrated by the elution of factor H from CpG-affinity-chromatography columns. Thus MØ are recruited to the tissue in the presence of C5a, and exploited locally by RTX
