    Biosolids, mycorrhizal fungi and Eucalypts for phytostabilization purposes of sulphidic mine tailings

    11 pages, 4 figures, 5 tables, 33 references.In mine tailings facilities, phytostabilisation is an attractive technology for long-term remediation. We tested the effect of the addition of biosolids combined with a native arbuscular mycorrhizal inoculum (AMF) on growth of a eucalypt (E. cladocalyx), and on trace element stabilization of sulphidic gold mine tailings. A glasshouse trial was established using four substrates: tailings (T); tailings with a layer of 5 cm topsoil (TS); tailings amended with 100 dry t ha-1 biosolids (LB), and tailings amended with 500 dry t ha-1 biosolids (HB). Half pots were inoculated with a mixture of Glomus sp. (WUM51 - 9227), Scutelaspora aurigloba (WUM51 - 53) and Acaulospora levis (WUM46) culture mix, and others were uninoculated controls. Two seeds per pot were sown in the pots and after 30 days all pots were uniformly thinned to 1 plant/pot. Plants were destructively harvested at 150 days after the sowing. Leaf and stem weights, leaf area and plant height were measured for each plant. Nutrients and trace element concentrations in leaves and stems were also determined. Addition of biosolids significantly increased mycorrhizal colonization (both ectomycorrhizal (ECM) and arbuscular mycorrhizal) in roots compared with the other treatments. Biosolids clearly improved the establishment and growth of the eucalypts. At low biosolid doses, mycorrhizal inoculum increased plant biomass production and the effectiveness of nutrient uptake. Trace element concentrations in trees of the biosolid-amended pots were in general high. It is therefore important to reduce the uptake of toxic elements by plants, which can be done successfully by adjusting amendment addition and the use of mycorrhizal inoculation, and then monitoring trace element contents of different taxa growing in the affected area.Dr. E.     The effect of contrasting biosolids application strategies on soil quality

    Purpose: Incorporating biosolids into the soil improves plant yield compared with surface application, but it can result in the increased uptake of trace elements. However, there is a lack of knowledge about how different types of biosolids applications affect soil quality. We aimed to determine the effect of the type and rate of biosolids application on soil quality and the mobility of contaminants. Methods: Soil quality was determined by soil fertility (inorganic N, exchangeable P, Mg, Ca, K), exchangeable trace and non-essential elements (Al, Mn, Zn, Cu and Cd) and biological activity (dehydrogenase activity). We measured the properties of soil pore water, bulk soil and rhizosphere in a pot and a rhizobox experiment, with increasing concentration of biosolids (equiv. 16 t ha¯¹, 48 t ha¯¹ and 145 t ha¯¹ dry weight), applied on the surface, incorporated to 25 cm, or incorporated into a patch. Results and discussion: The incorporation of biosolids into the soil increased the exchangeable Zn, Cu, Cr, Ni and Cd, compared with surface application. The surface application of biosolids increased the inorganic N in the soil compared with biosolids incorporation (680 mg kg¯¹ vs. 380 mg kg¯¹), and decreased soil pH by 1.1 units. This aligned with solubilisation of Al (43 mg kg¯¹ vs. 6 mg kg¯¹) and Mn (43 mg kg¯¹ vs. 33 mg kg¯¹) and explains the decreased microbial activity in the soil compared with the unamended soil. Incorporating biosolids in the soil increased the biological activity, likely due to biosolids-borne microbes. The root systems significantly increased microbial activity, pH, and the concentration of NH₄⁺, NO₃⁻, and exchangeable P, S, Mg, Na, Zn, Cu and Ni, and significantly decreased exchangeable concentration of Mn and Fe

    Cognitive Principles of Schematisation for Wayfinding Assistance

    People often need assistance to successfully perform wayfinding tasks in unfamiliar environments. Nowadays, a huge variety of wayfinding assistance systems exists. All these systems intend to present the needed information for a certain wayfinding situation in an adequate presentation. Some wayfinding assistance systems utilize findings for the field of cognitive sciences to develop and design cognitive ergonomic approaches. These approaches aim to be systems with which the users can effortless interact with and which present needed information in a way the user can acquire the information naturally. Therefore it is necessary to determinate the information needs of the user in a certain wayfinding task and to investigate how this information is processed and conceptualised by the wayfinder to be able to present it adequately. Cognitive motivated schematic maps are an example which employ this knowledge and emphasise relevant information and present it in an easily readable way. In my thesis I present a transfer approach to reuse the knowledge of well-grounded knowledge of schematisation techniques from one externalisation such as maps to another externalization such as virtual environments. A analysis of the informational need of the specific wayfinding task route following is done one the hand of a functional decomposition as well as a deep analysis of representation-theoretic consideration of the external representations maps and virtual environments. Concluding from these results, guidelines for transferring schematisation principles between different representation types are proposed. Specifically, this thesis chose the exemplary transfer of the schematisation technique wayfinding choremes from a map presentation into a virtual environment to present the theoretic requirements for a successful transfer. Wayfinding choremes are abstract mental concepts of turning action which are accessible as graphical externalisation integrated into route maps. These wayfinding choremes maps emphasis the turning action along the route by displaying the angular information as prototypes of 45° or 90°. This schematisation technique enhances wayfinding performance by supporting the matching processes between the map representation and the internal mental representation of the user. I embed the concept of wayfinding choremes into a virtual environment and present a study to test if the transferred schematisation technique also enhance the wayfinding performance. The empirical investigations present a successful transfer of the concept of the wayfinding choremes. Depending on the complexity of the route the embedded schematization enhance the wayfinding performance of participants who try to follow a route from memory. Participants who trained and recall the route in a schematised virtual environment make fewer errors than the participants of the unmodified virtual world. This thesis sets an example of the close research circle of cognitive behavioural studies to representation-theoretical considerations to applications of wayfinding assistance and their evaluations back to new conclusions in cognitive science. It contributes an interdisciplinary comprehensive inspection of the interplay of environmental factors and mental processes on the example of angular information and mental distortion of this information

    Agricultural use of compost under different irrigation strategies in a hedgerow olive grove under Mediterranean conditions – a comparison with traditional systems

    Soil and water-efficient management are key factors in ensuring the olive sector's sustainable production practices. The use of compost based on olive waste (alperujo) as fertilizer could enhance ecosystem services while the need to transition to a zero-waste approach based on a circular economy is achieved. The present work includes a comparative study of the effect of alperujo compost (AC) vs. inorganic fertilization under different management systems: a traditional adult olive grove under rainfed conditions and a young hedgerow olive system, in which a factorial test of tree irrigation regimes (full, deficit and no irrigation) is implemented as well. At the hedgerow plots, the addition of AC and soil sampling time greatly impacted soil chemical parameters and, to a lesser extent, enzymatic activities, whereas irrigation regimes did not exert a marked influence. In the traditional rainfed system, the addition of AC proved to be an efficient tool for carbon sequestration. The first soil sampling revealed a clear stoichiometric relationship between soil organic matter (SOM) and the nitrogen, phosphorus and potassium (NPK) contents in both systems, whereas the correlations were weak and scarce in the second sampling at the hedgerow plots. This fact was related to the decay of the compost effect. Compost in combination with irrigation tended to trigger a certain priming effect on the native SOM with time since the carbon stocks were reduced between 6 % and 38 % from one sampling to the other in the hedgerow system, depending on the irrigation intensity. However, the deficit irrigation caused a less intense reduction of the SOM and essential nutrients representing the best alternative to maximizing the agronomic effects of the compost under a water-saving strategy. Recurrent application of compost would be necessary to maintain soil quality, especially with high tree densities. The combined management of AC and the deficit irrigation proved to be an efficient tool toward a zero-waste circular economy and a water conservation strategy.</p

    An Assessment of Contaminants in UK Road-Verge Biomass and the Implications for Use as Anaerobic Digestion Feedstock

    Biomass from harvested road-verge herbage has potential value as a feedstock for anaerobic digestion (AD) energy plants. However, the proximity to road traffic related pollution sources introduces the possibility of contamination by potentially toxic elements and polycyclic aromatic hydrocarbons. Potential sources of pollution from road traffic emissions are identified and the consequent likelihood of certain contaminants being present at elevated levels is assessed. Samples of road verge biomass harvested from selected locations in Lincolnshire UK for use in AD plants were analysed to produce a set of measurements for the presence of the contaminants of interest. The measured levels of these contaminants are compared to reported background levels in UK herbage and soils to assess if there is significant increased concentration in road-verge biomass. Samples of digestate from an AD plant using the road-verge biomass as feedstock were also analysed to determine if there is notable risk of transfer and concentration of contaminants into agricultural land where the digestate may be used for fertilisation. While elevated levels of contaminants were detected, they were not found in concentrations on road verge biomass at high enough levels to cause adverse effects or concerns for its safe use as an AD feedstock

    Deep-sequencing reveals broad subtype-specific HCV resistance mutations associated with treatment failure

    A percentage of hepatitis C virus (HCV)-infected patients fail direct acting antiviral (DAA)-based treatment regimens, often because of drug resistance-associated substitutions (RAS). The aim of this study was to characterize the resistance profile of a large cohort of patients failing DAA-based treatments, and investigate the relationship between HCV subtype and failure, as an aid to optimizing management of these patients. A new, standardized HCV-RAS testing protocol based on deep sequencing was designed and applied to 220 previously subtyped samples from patients failing DAA treatment, collected in 39 Spanish hospitals. The majority had received DAA-based interferon (IFN) a-free regimens; 79% had failed sofosbuvir-containing therapy. Genomic regions encoding the nonstructural protein (NS) 3, NS5A, and NS5B (DAA target regions) were analyzed using subtype-specific primers. Viral subtype distribution was as follows: genotype (G) 1, 62.7%; G3a, 21.4%; G4d, 12.3%; G2, 1.8%; and mixed infections 1.8%. Overall, 88.6% of patients carried at least 1 RAS, and 19% carried RAS at frequencies below 20% in the mutant spectrum. There were no differences in RAS selection between treatments with and without ribavirin. Regardless of the treatment received, each HCV subtype showed specific types of RAS. Of note, no RAS were detected in the target proteins of 18.6% of patients failing treatment, and 30.4% of patients had RAS in proteins that were not targets of the inhibitors they received. HCV patients failing DAA therapy showed a high diversity of RAS. Ribavirin use did not influence the type or number of RAS at failure. The subtype-specific pattern of RAS emergence underscores the importance of accurate HCV subtyping. The frequency of “extra-target” RAS suggests the need for RAS screening in all three DAA target regions
