    The Proteome of Biologically Active Membrane Vesicles from Piscirickettsia salmonis LF-89 Type Strain Identifies Plasmid-Encoded Putative Toxins

    Indexación: Scopus.Piscirickettsia salmonis is the predominant bacterial pathogen affecting the Chilean salmonid industry. This bacterium is the etiological agent of piscirickettsiosis, a significant fish disease. Membrane vesicles (MVs) released by P. salmonis deliver several virulence factors to host cells. To improve on existing knowledge for the pathogenicity-associated functions of P. salmonis MVs, we studied the proteome of purified MVs from the P. salmonis LF-89 type strain using multidimensional protein identification technology. Initially, the cytotoxicity of different MV concentration purified from P. salmonis LF-89 was confirmed in an in vivo adult zebrafish infection model. The cumulative mortality of zebrafish injected with MVs showed a dose-dependent pattern. Analyses identified 452 proteins of different subcellular origins; most of them were associated with the cytoplasmic compartment and were mainly related to key functions for pathogen survival. Interestingly, previously unidentified putative virulence-related proteins were identified in P. salmonis MVs, such as outer membrane porin F and hemolysin. Additionally, five amino acid sequences corresponding to the Bordetella pertussis toxin subunit 1 and two amino acid sequences corresponding to the heat-labile enterotoxin alpha chain of Escherichia coli were located in the P. salmonis MV proteome. Curiously, these putative toxins were located in a plasmid region of P. salmonis LF-89.     In-situ measurements of temperature distribution during transmission laser welding of poly(aryletherketone)

    In this work, the temperature distribution inside the specimens has been measured during transmission laser welding. The material studied is a high performance thermoplastic of the polyaryletherketone (PAEK) family. The assembly consists of a quasi-amorphous sample as the upper part and a semi-crystalline sample as the lower part. The temperature fields were measured by infrared thermography with the camera sensor perpendicular to the welded interface. With an energy beam of 28 J.mm-2 and irradiation time of 15s, we have noticed that the maximum temperature inside the sample is kept far from the PAEK degradation one. Moreover, the temperature at the interface reaches the melting temperature thus assuring enough mobility for polymeric chains to get adhesion at the interface. Finally, the location and size of the heat-affected zone (HAZ) has been determined

    Study of the cosmogenic activation in NaI(Tl) crystals within the ANAIS experiment

    The direct detection of galactic dark matter particles requires ultra-low background conditions. NaI(Tl) crystals are applied in the search for these dark matter particles through their interactions in the detector by measuring the scintillation signal produced. The production of long-lived isotopes in materials due to the exposure to cosmic rays on Earth''s surface can be an hazard for these ultra-low background demanding experiments, typically performed underground. Therefore, production rates of cosmogenic isotopes in all the materials present in the experimental set-up, as well as the corresponding cosmic rays exposure history, must be both well-known in order to assess the relevance of this effect in the achievable sensitivity of a given experiment. Here, analysis of the cosmogenic studies developed from the ANAIS experiment NaI(Tl) detectors are presented. Installed inside a convenient shielding at the Canfranc Underground Laboratory just after finishing surface exposure to cosmic rays and thanks to the prompt data taking developed, identification and quantification of isotopes with half-lives of the order of tens of days were allowed, and thanks to the long-term operation of the detectors long-lived isotopes have been also identified and quantified. Main results for the activation yields of iodine and tellurium isotopes, 22Na, 113Sn, 109Cd, and tritium are presented in this work, together with the estimate of the production rates for their activation by cosmic nucleons while on Earth''s surface based on a selection of excitation functions over the entire energy range of cosmic nucleons

    Cosmogenic and primordial radioisotopes in copper bricks shortly exposed to cosmic rays

    Cosmogenic activation is the most common source of radioactivity in copper, being 60 Co the most significant because of its long half-life (5.27 y) and saturation activity at sea level of 1 mBq/kg. Copper bricks, which had been exposed to cosmic rays for 41 days after their casting, were used to replace the internal 10 cm of the lead shielding of a HPGe detector placed at the Canfranc Underground Laboratory.     The total phenols in virgin olive oil are highly dependent on cultivar, but also on ripening stage and other agronomic factors. The focus of most studies on agronomic factors has been irrigation, while fertilization has received less attention. Most of the fertilization works find that nitrogen over-fertilization leads to a decrease in phenol contents in virgin olive oil (VOO) and extra virgin olive oil (EVOO), under rain-fed or irrigation management. Ortho-diphenols also decrease with high doses of nitrogen, with no effect on secoiridoids. Phosphorous has a minor effect on irrigated trees; while the role of potassium is controversial, with a lack of trials with calcium and micro-nutrients. Due to the great impact of the fertilization on the phenol content and quality of VOO, new research is necessary with focus aimed at different cultivars and agronomic factors.Los fenoles del aceite de oliva tienen un fuerte componente varietal, estando influenciados por el estado de maduración y otros factores agronómicos. Entre estos últimos, el efecto del riego es el más conocido, mientras que la fertilización está menos estudiada. La mayoría de los trabajos existentes sobre fertilización, muestran que el incremento en el abonado nitrogenado disminuye los niveles de fenoles totales del aceite de oliva virgen o virgen extra, tanto en secano como en regadío. También se han observado niveles inferiores de ortodifenoles en tratamientos con aplicación de nitrógeno, sin detectar diferencias en el contenido de secoiridoides. El fósforo tiene una influencia menor y la del potasio es algo controvertida, quedando por explorar la influencia del calcio y de los micronutrientes. Debido al considerable impacto que tiene la fertilización en los fenoles así como en la calidad del aceite, sería necesario incrementar los estudios de fertilización con diferentes variedades y sistemas productivos

    Influence of irrigation and fertilization on the sterol and triterpene dialcohol compositions of virgin olive oil

    The objective of this work was to evaluate the influence of irrigation and fertilization with nitrogen and potassium on the sterol and triterpene dialcohol contents in two trials of cv. Arbequina in super-intensive orchards in Madrid and Lleida (Spain), using a completely randomized block design. No significant differences in total sterols between deficit and full irrigation treatments were observed. Under very dry conditions, the sterol levels from fully irrigated trees were higher than from rain-fed treatments and the triterpene dialcohol erythrodiol+ uvaol content was lower in the irrigated treatments in Lleida. In the fertilizer trial with full irrigation, total sterols were higher in the two N treatments compared to the unfertilized one; while erythrodiol + uvaol decreased. The application of K fertilizer had no effect on total sterol or triterpene dialcohol contents. A proper fertilization and irrigation are vital to obtain high quality EVOOs that meet the regulatory range in sterol and erythrodiol + uvaol contents.Influencia del riego y la fertilización en los esteroles y dialcoholes triterpénicos del aceite de oliva. El objetivo de este trabajo fue evaluar la influencia del riego y del abonado nitrogenado y potásico en los esteroles y los dialcoholes tritepénicos, en sendos ensayos de arbequina en superintensivo, con un diseño en bloques al azar, en Madrid y Lleida. No se observaron diferencias significativas de esteroles totales entre el riego deficitario y el riego completo. En condiciones de sequía extrema, los esteroles del riego completo fueron superiores al secano y los dialcoholes tritepénicos eritrodiol y uvaol fueron inferiores en los tratamientos de riego en Lleida. En el ensayo de abonado con riego completo, los esteroles totales fueron superiores en las dos aplicaciones de nitrógeno respecto al testigo no tratado y también disminuyeron el eritrodiol + uvaol. El abonado potásico no afectó a los esteroles totales ni a los dihalcoholes triterpénicos. Una fertilización y riego adecuados son de vital importancia para obtener AOVEs de alta calidad que cumplan con los límites legales del contenido en esteroles y eritrodiol + uvaol.info:eu-repo/semantics/publishedVersio

    Status of the ANAIS Dark Matter Project at the Canfranc Underground Laboratory

    The ANAIS (Annual modulation with Nal(Tl) Scintillators) experiment aims at the confirmation of the DAMA/LIBRA signal using the same target and technique at the Canfranc Underground Laboratory (LSC). Along 2016, 112.5 kg of ultra pure Nal(Tl) crystals will be installed at LSC in a 3×3 modules matrix configuration. The ANAIS-25 and ANAIS-37 set-ups have been taking data at the LSC testing the detector performance, the DAQ and analysis systems, and assessing the background. Main results coming from both set-ups will be summarized in this paper, focusing on the excellent detector performance and background understanding. Prospects for the experiment will be also briefly revised

    First Results on Dark Matter Annual Modulation from the ANAIS-112 Experiment

    ANAIS is a direct detection dark matter experiment aiming at the testing of the DAMA/LIBRA annual modulation result, which, for about two decades, has neither been confirmed nor ruled out by any other experiment in a model independent way. ANAIS - 112, consisting of 112.5 kg of sodium iodide crystals, has been taking data at the Canfranc Underground Laboratory, Spain, since August 2017. This Letter presents the annual modulation analysis of 1.5 years of data, amounting to 157.55 kg yr. We focus on the model independent analysis searching for modulation and the validation of our sensitivity prospects. ANAIS - 112 data are consistent with the null hypothesis (p values of 0.67 and 0.18 for [2-6] and [1-6] keV energy regions, respectively). The best fits for the modulation hypothesis are consistent with the absence of modulation (S-m = -0.0044 +/- 0.0058 cpd/kg/keV and -0.0015 +/- 0.0063 cpd/kg/keV, respectively). They are in agreement with our estimated sensitivity for the accumulated exposure, which supports our projected goal of reaching a 3 sigma sensitivity to the DAMA/LIBRA result in five years of data taking

    The ANAIS-112 experiment at the Canfranc Underground Laboratory

    The ANAIS experiment aims at the confirmation of the DAMA/LIBRA signal at the Canfranc Underground Laboratory (LSC). Several 12.5 kg NaI(Tl) modules produced by Alpha Spectra Inc. have been operated there during the last years in various set-ups; an outstanding light collection at the level of 15 photoelectrons per keV, which allows triggering at 1 keV of visible energy, has been measured for all of them and a complete characterization of their background has been achieved. In the first months of 2017, the full ANAIS-112 set-up consisting of nine Alpha Spectra detectors with a total mass of 112.5 kg was commissioned at LSC and the first dark matter run started in August, 2017. Here, the latest results on the detectors performance and measured background from the commissioning run will be presented and the sensitivity prospects of the ANAIS-112 experiment will be discussed

    Preliminary results of ANAIS-25

    The ANAIS (Annual Modulation with NaI(Tl) Scintillators) experiment aims at the confirmation of the DAMA/LIBRA signal using the same target and technique at the Canfranc Underground Laboratory. 250 kg of ultrapure NaI(Tl) crystals will be used as a target, divided into 20 modules, each coupled to two photomultipliers. Two NaI(Tl) crystals of 12.5 kg each, grown by Alpha Spectra from a powder having a potassium level under the limit of our analytical techniques, form the ANAIS-25 set-up. The background contributions are being carefully studied and preliminary results are presented: their natural potassium content in the bulk has been quantified, as well as the uranium and thorium radioactive chains presence in the bulk through the discrimination of the corresponding alpha events by PSA, and due to the fast commissioning, the contribution from cosmogenic activated isotopes is clearly identified and their decay observed along the first months of data taking. Following the procedures established with ANAIS-0 and previous prototypes, bulk NaI(Tl) scintillation events selection and light collection efficiency have been also studied in ANAIS-25
